Claims (7)
- (1) GST 융합 단백질 유전자를 함유하는 균주를 배양하는 단계와,(2) 상기 (1) 단계에서 배양된 배양액을 원심분리하여 세포가 용리된 용리액을 얻는 단계와,(3) 상기 (2) 단계의 용리액을 GSH 결합 수지에 적용하여 GSH 친화성 컬럼에 GST 융합 단백질을 결합시키는 단계와,(4) 2% 포름산을 함유하는 용리 완충액으로 GST 융합 단백질을 용리하는 단계, 및(5) GST 융합 단백질을 회수하는 단계를 포함하는, 용리 완충액으로 2% 포름산을 사용하는 것에 특징이 있는 재조합형 GST 융합 단백질의 정제방법.
- 제1항에 있어서, 상기 단계 (1) 은 i) 목적단백질을 코딩하는 DNA 서열을 제조하는 단계, ii) 상기 DNA 서열을 GST 코딩 DNA 서열을 포함하는 벡터에 작동가능하게 연결하여 재조합 발현 벡터를 제조하는 단계, iii) 상기 재조합 발현 벡터로 숙주를 형질전환시키는 단계, 및 iv) 생성된 형질전환체를 상기 DNA 서열이 발현되도록 하기에 적절한 배지 및 조건하에서 배양하는 단계를 포함하는 것인, GST 융합 단백질의 정제방법.
- 삭제
- 제1항에 있어서, 상기 GST에 융합되는 단백질은 사이클로필린, 호르몬, 효소, 사이토카인, 과립구 콜로니 자극인자, 적혈구 생성인자, 베타셀룰린(betacellulin, BTC) 또는 항체인 것인 GST 융합 단백질의 정제방법.
- 제1항에 있어서, 상기 GST에 융합되는 단백질은 사이클로필린 A인 것인 GST 융합 단백질의 정제방법.
- 제1항에 있어서, 상기 단계 (4)에 앞서 상기 단계 (3)의 GST 융합 단백질이 결합된 GSH 친화성 컬럼을 세척 완충액 으로 세척하는 단계를 더 포함하는 것인 재조합형 GST 융합 단백질의 정제방법.
- 제1항에 있어서, 상기 단계 (4)에서 용리된 분획을 1M Tris-HCl를 함유하는 완충액으로 중화하는 단계를 더 포함하는 것인 재조합형 GST 융합 단백질의 정제방법.
