Abstract
본 발명은 다우리놀(daurinol)을 유효성분으로 하는 암 예방 및 치료용 약학적 조성물에 관한 것으로, 구체적으로 몽골 약용 식물인 하플로필럼 다우리쿰(Haplophyllum dauricum) 또는 하플로필럼 카파도시쿰(Haplophyllum cappadocicum)으로부터 분리한 다우리놀을 유효성분으로 하는 암 예방 및 치료용 약학적 조성물에 관한 것이다. 하플로필럼 다우리쿰(Haplophyllum dauricum)으로부터 분리한 다우리놀은 암세포의 세포주기 중 S 기를 선택적으로 정지시켜 세포주기를 교란하며, 순차적으로, 세포자살 반응을 일으킴으로써 항암 활성을 나타낸다. 따라서 본 발명의 다우리놀은 항암제 개발연구에 있어서 양성 대조군(positive control) 약물로서 신규 세포주기 교란제 및 신규 세포자살반응 유도제로서 사용가능하며, 궁극적으로는 암 예방 및 치료용 약학적 조성물로서 유용하게 이용될 수 있다.
Description
본 발명은 다우리놀(daurinol)을 유효성분으로 하는 암 예방 및 치료용 약학적 조성물에 관한 것으로, 상세하게는 몽골 약용 식물인 하플로필럼 다우리쿰(Haplophyllum dauricum) 또는 하플로필럼 카파도시쿰(Haplophyllum cappadocicum)으로부터 분리한 다우리놀을 유효성분으로 하는 암 예방 및 치료용 약학적 조성물에 관한 것이다.
항암제란 암세포에서 DNA와 직접 작용하여 암세포의 복제, 전사, 번역 과정을 차단하거나, 대사 경로에 개입하여 핵산 전구체의 합성을 방해하고, 암세포 분열을 저해함으로써 항암활성, 즉 암세포에 대한 독성을 나타내는 약제를 총칭한다. 항암제는 증식 의존성 약물로 활발히 분열 증식하는 세포에만 작용하는 특징을 가지며, 따라서 항암제는 일종의 항증식성 약제(antiproliferative drug) 라고 할 수 있다(서울대학교 의과대학, 종약학, pp 153-164, 2003, 서울대학교 출판부). 특히 세포주기 교란(cell cycle arrest)과 세포자살반응(apoptosis)은 암세포의 세포독성 또는 항증식 활성을 나타나게 하는 가장 중요한 메커니즘 중의 하나이다. 그러므로 세포주기 교란과 세포자살 반응 유도는 항암제 개발에서 가장 중요한 표적이 된다.
세포주기는 G1 기, S 기, G2 기 및 M 기의 4 단계로 구성된다. G1 기는 세포생장에 필요한 효소와 구조단백질 등 세포분열에 필요한 다양한 단백질들이 합성되는 기간이며, S 기는 세포내 DNA가 합성되는 기간으로 합성기이며, G2 기는 튜불린 단백질 등 유사분열을 준비하는 기간이며, M 기는 핵분열과 세포질 분열 등 세포가 분열되는 세포분열기이다. G1 기가 과도하게 길어 세포분열이 휴지 상태에 있는 경우를 G0 기라 한다. 일반적으로 세포분열에 소요되는 시간은 G1 기의 길이에 의하여 좌우된다. 암세포는 무한히 분열하는 특성을 가지며, 정상세포에 비하여 G0/G1 기에 머무는 시간이 짧은 특징을 가지며, 그 결과로 세포분열이 매우 빠르게 진행된다. 그러므로, 빠르게 세포분열되는 즉, 세포주기가 빠르게 반복되는 암세포에서 특정 세포주기를 정지 시킬 수 있는 약물을 처리하여 세포주기를 교란할 경우, 정상세포는 구별하면서 암세포의 증식만을 선택적으로 억제 할 수 있게 된다(서울대학교 의과대학, 종약학, pp 153-164, 2003, 서울대학교 출판부; Alberts B, et al ., Molecular Biology of the Cell (4th ed), pp 983-1026, 2002).
한편, 다우리놀은 아릴나프탈렌 리그난(arylnaphthalene lignan) 계열의 단일화학 성분으로, 1980 년도 초반에 본 발명자들이 몽골 약용 식물인 하플로필럼 다우리쿰(Haplophyllum dauricum)으로부터 처음 분리하였으며, 같은 속인 하플로필럼 카파도시쿰(Haplophyllum cappadocicum)등에서도 보고되었다(Batsuren D, et al., Chemistry of Natural Compounds, 17, 223-225, 1982; Gozler B, et al., Phytochemistry, 31, 2473-2475, 1992). 그러나 현재까지 다우리놀에 대한 약리학적 활성은 전혀 보고되지 않았다. 하플로필럼(Haplophyllum) 속 식물은 중앙아시아 지방에 광범위하게 분포 하고 있는 다년생 풀이며, 수백 년 간 다양한 질병을 대상으로 전통적인 민간요법에 이용되었다.
이에, 본 발명자들은 몽골 약용식물인 하플로필럼 다우리쿰(Haplophyllum dauricum)으로부터 분리된 다우리놀이 암세포주의 세포주기 중 S 기를 선택적으로 정지하고, 이어서 세포자살 반응을 일으킴으로써 세포독성을 나타내므로, 다우리놀이 신규 세포주기 교란제, 신규 세포자살반응 유도제, 신규 세포사멸 유도제, 암 예방 및 치료용 약학적 조성물에 이용될 수 있음을 밝힘으로써 본 발명을 완성하였다.
본 발명은 몽골 약용 식물인 하플로필럼 다우리쿰(Haplophyllum dauricum) 또는 하플로필럼 카파도시쿰(Haplophyllum cappadocicum)으로부터 분리한 다우리놀을 유효성분으로 하는 암 예방 및 치료용 약학적 조성물에 관한 것이다.
상기 목적을 달성하기 위하여, 본 발명은 다우리놀(daurinol)을 유효성분으로 함유하는 암 예방 및 치료용 약학적 조성물을 제공한다.
또한, 본 발명은 다우리놀(daurinol)을 유효성분으로 함유하는 세포주기 교란제를 제공한다.
또한, 본 발명은 다우리놀(daurinol)을 유효성분으로 함유하는 세포사멸 유도제를 제공한다.
아울러, 본 발명은 다우리놀(daurinol)을 유효성분으로 함유하는 세포자살반응 유도제를 제공한다.
이하, 본 발명을 상세히 설명한다.
본 발명은 다우리놀(daurinol)을 유효성분으로 함유하는 암 예방 및 치료용 약학적 조성물을 제공한다.
본 발명의 다우리놀(daurinol)은 몽골 약용 식물인 하플로필럼 다우리쿰(Haplophyllum dauricum) 또는 하플로필럼 카파도시쿰(Haplophyllum cappadocicum)으로부터 유래한 아릴나프탈렌 리그난 계열의 단일 화학성분으로서, 하기 화학식 1 로 표시된다.
상기 하플로필럼 다우리쿰[Haplophyllum dauricum (L.) G. Don.]은 몽골의 우불크한가이스키 아이막(Uvurkhangaiskii aimak) 지방에서 채집되었으며, 에탄올을 용매로 하여 추출되었다. 다우리놀은 상기 식물의 지상부에 많이 존재하지만, 이 식물의 뿌리에서도 분리할 수 있다. 추출물은 석유 에테르(petroleum ether), 클로로포름, 에틸아세테이트, 부탄올을 이용하여 분획되었으며, 이 중에서 클로로포름 분획물을 실라카겔 칼럼(1:10)을 이용하여 분리하였으며, 다양한 비율의 벤젠-클로로포름 용매를 이용하여 용리하였다. 메탄올을 용매로 하여, 분별결정(fractional recrystallization)함으로써 다우리놀은 분리하였으며, 다양한 분광학적 방법으로, UV-스펙트럼, IR-스펙트럼, 질량스펙트럼, 1H-NMR 스펙트럼 등을 분석하여 다우리놀의 구조를 확인하였다.
본 발명의 다우리놀은 상기와 같은 방법으로 하플로필럼 다우리쿰(Haplophyllum dauricum), 하플로필럼 카파도시쿰(Haplophyllum cappadocicum) 등의 특정 하플로필럼(Haplophyllum) 속 식물로부터 추출 분리하여 사용하거나, 당업계에서 널리 알려진 화학 합성 방법을 이용하여 제조하여 사용할 수도 있다.
본 발명자들은 다우리놀의 암세포 독성을 측정하기 위하여, 대장암 세포주인 HCT116과 DLD-1을 대상으로 MTT(methylthiazol-2-yl-2.5 diphenyl tetrazolium bromide) 방법을 수행하였다. 그 결과, 두 가지 세포주 모두에서 세포 독성이 매우 탁월하게 나타났으며, 암세포 증식 억제 효과는 농도 의존적이었다. 또한 세포 증식억제 효과는 24 시간 처리구에 비하여 48 시간 처리 시에 보다 탁월하게 나타남을 확인하였다(표 1 및 도 1 참조).
또한, 다우리놀을 처리한 대장암 세포주를 수득하여 고정하고, DNA를 선택적으로 염색하는 요오드화 프로피디엄(propidium iodide, PI)을 처리한 다음, 유세포 분석기를 이용하여 세포주기를 분석하였다. 그 결과, 다우리놀은 암세포에서 세포주기 중 S 기에 멈추게 함을 확인하였다(도 2, 도 3 및 도 4 참조). 상기 결과는 다우리놀이 세포 교란제로서 이용될 수 있음을 의미한다.
또한, 본 발명자들은 세포주기 분석을 통해 세포자살반응의 척도가 되는 sub G0/G1 비율이 대조군에 비하여, 다우리놀 처리군에서 현저히 증가됨을 확인하였다(표 2 및 도 5 참조). sub G0/G1은 G0/G1기보다 DNA 양이 더 적은 부분(예를 들면, 세포주기 그래프에서 G0/G1 피크의 앞쪽 또는 왼쪽 부분)을 의미한다. 도 2 및 도 3에서 "세포자살"이라고 표시된 부분이 sub G0/G1에 해당되며, 첫 번째 피크가 G0/G1기, 가운데 부분이 S기, 두 번째 피크가 G2기 및 M기에 해당된다.
이와 함께 다우리놀을 처리한 암세포를 수득하고, 아넥신 V(Annnexin V)-FITC(Fluorescein isothiocyanate)과 요오드화 프로피디엄을 동시 염색법을 통해 세포자살반응이 유도된 초기의 세포 비율이 대조군과 비교하여 현저히 증가됨을 확인하였다(도 6 및 도 7 참조). 이는 다우리놀이 세포자살반응 유도제로서 이용될 수 있음을 의미한다.
다우리놀을 처리한 후, 24 시간이 경과된 암세포주를 DIC(differential interference contrast) 광학 현미경으로 관찰해 보면, 세포 간 간격이 멀어져 세포 사이의 경계가 명확해지는 형태학적 특징을 보인다. 다우리놀 처리 후 48 시간이 경과한 시점에서 암세포주를 DIC(differential interference contrast) 광학 현미경으로 관찰하면, 암세포는 바닥으로부터 떨어져 나오기 시작한다. 이후 72 시간 및 96 시간이 경과한 시점에서 이러한 현상이 보다 두드러지게 나타나며(도 8 참조), 이는 다우리놀이 세포사멸반응을 유도한 결과이다.
상기에서 살펴보았듯이 본 발명의 다우리놀은 세포주기 중 S 기를 선택적으로 정지시키며, 세포자살 반응을 유도함으로서 항암효과를 가지므로 암 예방 및 치료용 약학적 조성물에 유용하게 이용될 수 있다.
상기 암의 예를 들자면, 후두암, 폐암, 식도암, 췌장암, 대장암, 간암, 위암, 설암, 피부암, 뇌종양, 자궁암, 유방암, 자궁경부암, 난소암, 신장암, 담낭암, 구강암, 결장암, 간암 또는 방광암 등이 있으며, 대장암인 것이 가장 바람직하나 이에 제한되는 것은 아니다.
상기 다우리놀은 임상 투여 시에 경구로 투여가 가능하며, 일반적인 의약품 제제의 형태로 사용될 수 있으며 다우리놀에 추가로 동일 또는 유사한 기능을 나타내는 유효성분을 1종 이상 함유할 수 있다. 투여를 위해서는 추가로 약제학적으로 허용 가능한 담체를 1종 이상 포함하여 제조할 수 있다. 약제학적으로 허용 가능한 담체는 식염수, 멸균수, 완충 식염수, 덱스트로스 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 성분 이상을 혼합하여 이용할 수 있으며, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수 있다. 또한, 희석제, 분산제, 계면활성제, 결합제 및 윤활제를 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 산제, 정제, 캡슐제, 환 또는 과립 등으로 제제화 할 수 있으며, 각 질환에 따라 또는 성분에 따라 바람직하게 제제화할 수 있다.
또한, 본 발명은 다우리놀(daurinol)을 유효성분으로 함유하는 세포주기 교란제, 세포사멸 유도제 및 세포자살반응 유도제를 제공한다.
상기에서 살펴보았듯이 본 발명의 다우리놀은 세포주기 중 S 기를 선택적으로 정지시키므로 세포주기 교란제에 유용하게 이용될 수 있으며, 또한 세포자살 반응을 유도함으로 세포사멸 유도제로 유용하게 이용될 수 있다.
이하 본 발명을 실시예, 실험예 및 제제예에 의해 상세히 설명한다.
단, 하기 실시예, 실험예 및 제제예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예, 실험예 및 제제예에 한정되는 것은 아니다.
<실시예 1> 다우리놀(daurinol)의 제조
다우리놀 추출에 사용된 식물인 하플로필럼 다우리쿰[Haplophyllum dauricum (L.) G. Don.]은 몽골의 우불크한가이스키 아이막(Uvurkhangaiskii aimak) 지방에서 채집되었다. 이 식물을 곱게 빻은 재료 6.5 ㎏을 에탄올을 용매로 하여 10회에 걸쳐 추출하였으며, 감압증류하여 농축하였다. 농축된 에탄올 추출물에 농축된 에탄올의 부피만큼 증류수를 첨가하여 희석(1:1)하고 이어서 여러 가지 용매를 이용하여 분획한 결과, 석유 에테르(petroleum ether) 층에서 261 g, 클로로포름 층에서 126 g, 에틸아세테이트 층에서 20 g, 부탄올 층에서 258 g의 분획물이 수득되었다. 이 중 클로로포름 분획물을 실라카겔 칼럼(1:10)을 이용하여 분리하였으며, 다양한 비율의 벤젠-클로로포름 용매를 이용하여 용리하였다. 메탄올을 이용한 분별결정(fractional recrystallization) 방법을 이용하여 다우리놀 0.82 g을 수득하였다.
재결정된 다우리놀의 녹는점은 256 ~ 257℃ 이었으며, 클로로포름-에틸아세테이트(19:1)를 이동상으로 Silufol UV-254 플레이트를 이용한 박층 크로마토그래피 (thin layer chromatography)를 수행한 결과, Rf 값은 0.54로 나타났으며, 이하 다양한 분광학적 방법으로 다우리놀의 구조를 확인하였으며, 하기 화학식 1과 같은 구조를 갖는다.

EPS-3T 분광계를 이용한 다우리놀의 UV-스펙트럼 조사 결과, λethanol max 227, 262, 296, 324, 352 ㎚ (logε 4.55, 4.68, 4.07, 4.09, 3.41)로 나타났다. UR-20 분광광도계를 이용한 IR-스펙트럼 조사 결과, 흡수 밴드(cm-1)가 3390(수산화기; hydroxy group), 1762 (C=O), 623, 1600(방향환; aromatic nucleus), 936(-OCH2O) 및 777(치환된 방향환; 1,2,4-trisubstituted benzene ring)에서 나타났다. Mkh-1303 분광계를 이용하여 질량스펙트럼(mass spectrum)을 분석한 결과, m/e(상대존재비; relative abundance %) : M+ 350 (100), M-CHO 321 (10, m* 350 → 321, 294, 5), M-CO2H 305 (10), M-C2H2O2 292 (4), M-CH3OCO 291 (15), 278 (4), 277 (11), 264 (5), 263 (13), 251 (6), 249 (5), 248 (6), 235 (5), 233 (7), 221 (6), 220 (8), 205 (5), 192 (6), 176 (7), 175 (4), 165 (7), 164 (6), 163 (13), 160 (11), 146 (6)으로 나타났다.
0.5 ㎖의 피리딘(pyridine) 존재하에서 2 ㎖의 무수아세트산(acetic anhydride)과 50 ㎎의 다우리놀을 상온에서 24 시간 동안 반응시켜, 다우리놀을 아세틸화(acetylation) 한 다음, 1H-NMR 스펙트럼을 조사한 결과(JNM-4H-100 분광계), 2.30 (3H, s, Ar-OCOCH3), 2.67 (3H, s, Ar-OCH3), 5.24 (2 H, s, Ar-CH2O-), 5.92 and 5.98 (1 H each, dd, Jgem= 1.2 Hz, Ar-OCH2O), 6.74 (1 H, br. s, H -2/), 6.70 (1 H, q, 8.4 및 1.5 Hz, H-6), 6.86 (1H, d, 8.5 Hz, H-5), 7.08 and 7.46 (1 H, s, each, H-5 및 H-8), 및 7.62 (1 H, br, s H-4)으로 나타났다. 1H-NMR 스펙트럼 분석시 중수소로 치환된 클로로포름 (CDCl3)을 용매로 사용하였다.

실험예
1>
다우리놀의
암세포 증식 억제 효능 검증
상기 실시예에서 분리한 다우리놀(daurinol)을 인간 대장암 세포주인 HCT116 및 DLD-1(American type of cell culture, ATCC)에 처리하여, 세포 증식 억제효과를 측정하였다. 상기 두개의 암 세포주는 10% 우태혈청(fetal bovine serum)과 항생제(페니실린 및 스트렙토마이신)가 포함된 MEM 배지를 이용하여 37℃, 5% CO2 에서 배양하였으며, 2 ~ 3 일마다 계대하여 세포주를 배양하였다.
HCT116 및 DLD-1 세포주를 5×103 세포/웰의 밀도로 96-웰 플레이트(96-well plate)에 접종하고, 24 시간 동안 배양한 후, DMSO(Dimethyl Sulphoxide)에 녹인 다우리놀을 0.1 ~ 200 ㎍/㎖의 다양한 농도로 24 시간 및 48 시간 동안 처리하였다. 다음으로, MTT(methylthiazol-2-yl-2.5 diphenyl tetrazolium bromide) 시약을 처리하고, 1 ~ 3 시간 경과 후, 450 nm에서 흡광도의 측정하여 세포 증식 정도를 확인하였다.
그 결과, 두 가지 암세포주 모두에서 세포 증식 억제 효과는 다우리놀 농도에 의존적이었으며, 매우 탁월하게 나타났다. 다우리놀에 대한 감수성은 DLD-1에서보다 HCT116에서 보다 높게 나타났다. 또한, 다우리놀을 24 시간 동안 처리한 경우보다 48 시간 동안 처리한 경우에 세포 증식억제 효과가 보다 탁월하게 나타났다(도 1). 이러한 현상이 나타나는 이유는 다우리놀을 처리한 후 24 시간이 지나면, 암세포가 세포주기 중 S 기에 정지를 일으키고, 48 시간이 경과한 후에는 세포자살반응이 뚜렷이 진행되기 때문이다. 또한 50%의 세포가 생존하는 농도인 IC50 값을 구하여 하기 표 1에 나타내었다. IC50 값이 낮을수록 세포 독성이 강함을 의미하는데, 다우리놀은 DLD-1에서보다 HCT116에서 강한 세포독성을 나타냄을 알 수 있었다.

<실험예 2> 세포주기 교란제로서의 다우리놀의 효능 검증
다음으로 본 발명자들은 상기 실시예 1에서 제조한 다우리놀(daurinol)의 세포주기 교란 효과를 하기와 같이 하여 검증하였다.
인간 대장암 세포인 HCT116 및 DLD-1을 60×15 cm 크기의 배양 접시 당 5×105 개를 접종하고, 24 시간 동안 배양한 후, DMSO(Dimethyl Sulphoxide)에 녹인 다우리놀을 5 ~ 20 uM 농도로, 24 시간, 48 시간 및 72 시간 동안 처리하였다. 다음으로, 세포를 수득하여 DPBS(Dulbecco's Phosphate Buffered Saline)로 세척한 다음, 70% 에탄올에 넣어 -20℃에서 하루 이상 고정하였다. 이 후, 세포를 다시 DPBS로 세척한 다음, 요오드화 프로피디엄(propidium iodide)을 이용하여 20 분간 염색하고, 유세포 분석기를 이용하여 분석하였다. 이때 사용된 요오드화 프로피디엄은 세포내 DNA만을 선택적으로 염색시키기 때문에, 요오드화 프로피디엄에 의한 형광량과 세포수를 변수로 하여 그래프를 도시하면, 세포주기의 모양이 형성된다. 이 그래프를 분석하면 다우리놀에 의한 세포주기 교란 효과와 세포자살 반응 여부를 확인할 수 있다.
HCT116 및 DLD-1 암세포주에 다양한 농도(5, 10 및 20 uM)의 다우리놀을 48 시간 동안 처리한 결과, 다우리놀을 처리하지 않은 대조군과 비교하여 다우리놀 처리군에서 S 기의 비율이 크게 증가하는 특성을 확인하였다(도 2 및 도 3). 또한 세포자살반응 유도 정도의 척도가 되는 sub G0/G1 비율이 다우리놀을 처리하지 않은 대조군에 비하여 다우리놀을 처리한 군에서 유의하게 증가함을 확인하였으며, 특히 HCT116 세포주에서 크게 증가함을 확인하였다(표 2).
또한, HCT116 암세포주에 5 uM 농도의 다우리놀을 다양한 시간(24, 48 및 72시간) 동안 처리 할 경우에서도, 다우리놀을 처리하지 않은 대조군과 비교하여 다우리놀 처리군에서 S 기의 비율이 크게 증가하는 특성이 확인되었다(도 4).
아울러, HCT116 암세포주에 5 uM 다우리놀을 처리한 실험군에서 세포자살반응 유도 정도의 척도가 되는 sub G0/G1 비율도 크게 증가함을 확인하였다(도 5).

<실험예 3> 다우리놀의 세포자살반응 유도 효능 검증
상기 실험예 2에서 살펴보았듯이, 하플로필럼 다우리쿰(Haplophyllum dauricum)으로부터 분리한 다우리놀(daurinol)은 세포자살반응의 척도인 sub G0/G1 비율은 대조군과 비교하여 크게 증가하였다. 본 발명자들은 이와 더불어 다우리놀의 세포자살 반응 유도를 보다 정확하게 확인하기 위하여, 아넥신(annexin) V-FITC(Fluorescein isothiocyanate)와 요오드화 프로피디엄(propidium iodide) 동시 염색법을 이용하였다.
정상적인 세포에서 세포막 구성 성분인 인지질 중 포스파티딜 세린은 항상 세포 안쪽으로 향하는 특징이 있는데, 초기 세포자살반응 유도 시 이 포스파티딜 세린은 세포 바깥쪽으로 노출되게 된다. 이때 포스파티딜 세린과 선택적으로 결합하는 아넥신(annexin) V를 녹색 형광 물질로 표시함으로서 세포자살 반응이 유도된 초기 세포의 비율을 확인할 수 있다.
HCT116 세포를 60×15 cm 크기의 배양 접시 당 5×105 개를 접종하고, 24 시간 동안 배양한 후, DMSO(Dimethyl Sulphoxide)에 녹인 다우리놀을 5 uM 농도로, 24 시간 및 48 시간 동안 처리하였다. 다음으로, 세포를 수득하여 DPBS(Dulbecco's Phosphate Buffered Saline)로 세척한 다음, 1×106 개 세포만을 옮겨서 아넥신(annexin) V-FITC(Fluorescein isothiocyanate) 및 요오드화 프로피디엄(propidium iodide)으로 15분간 염색한 뒤, 유세포 분석기를 이용하여 분석하였다.
그 결과, 5 uM 다우리놀을 처리한 HCT116 세포에서 대조군과 비교하여 세포자살반응이 현저히 유도됨을 확인하였다(도 6 및 도 7). 세포자살 반응의 유도는 48 시간이 경과 된 후에야 명백히 유도되었다. 이는 다우리놀이 세포주기 중 S 기를 멈추게 한 뒤, 이어서 세포자살 반응을 유도시킴을 의미한다.
<실험예 4> 다우리놀 처리에 의한 암세포주의 형태학적 변화 검증
본 발명자들은 최종적으로, 몽골 약용 식물인 하플로필럼 다우리쿰(Haplophyllum dauricum)으로부터 분리한 다우리놀(daurinol)을 처리한 암세포에서 나타나는 형태학적 특징을 DIC(differential interference contrast) 광학 현미경을 이용하여 확인하였다.
HCT116 암세포를 60×15 cm 크기의 배양접시 당 5×105 개를 접종하고, 24 시간 동안 배양하였다. 이후 DMSO(Dimethyl Sulphoxide)에 녹여 5 uM 농도의 다우리놀을 처리하고, 각각 12, 24, 48, 72 및 96 시간 경과 후에 DIC 광학 현미경을 이용하여 세포의 형태를 촬영하였다.
그 결과, 다우리놀 처리 후 24 시간이 경과한 시점에서 HCT116 암세포는 세포 간 간격이 멀어져 세포사이의 경계가 명확히 늘어남을 확인하였으며, 48 시간이 경과한 시점에서 HCT116 암세포는 바닥으로부터 떨어져 나오기 시작함을 알 수 있으며, 이후 72 및 96 시간이 경과한 시점에서 이러한 현상이 보다 두드러지게 나타났다(도 8).
하기에 본 발명의 조성물을 위한 제제예를 예시하나, 이에 제한되는 것은 아니다.
<제제예 1> 약학적 제제의 제조
<1-1> 산제의 제조
다우리놀(daurinol) 2 g
유당 1 g
상기의 성분을 혼합한 후, 기밀포에 충진하여 산제를 제조하였다.
<1-2> 정제의 제조
다우리놀(daurinol) 100 ㎎
옥수수전분 100 ㎎
유 당 100 ㎎
스테아린산 마그네슘 2 ㎎
상기의 성분을 혼합한 후, 통상의 정제의 제조방법에 따라서 타정하여 정제를 제조하였다.
<1-3> 캡슐제의 제조
다우리놀(daurinol) 100 ㎎
옥수수전분 100 ㎎
유 당 100 ㎎
스테아린산 마그네슘 2 ㎎
상기의 성분을 혼합한 후, 통상의 캡슐제의 제조방법에 따라서 젤라틴 캡슐에 충전하여 캡슐제를 제조하였다.
<1-4> 환의 제조
다우리놀(daurinol) 1 g
유당 1.5 g
글리세린 1 g
자일리톨 0.5 g
상기의 성분을 혼합한 후, 통상의 방법에 따라 1환 당 4 g이 되도록 제조하였다.
<1-5> 과립의 제조
다우리놀(daurinol) 150 ㎎
대두추출물 50 ㎎
포도당 200 ㎎
전분 600 ㎎
상기의 성분을 혼합한 후, 30% 에탄올 100 ㎎을 첨가하여 섭씨 60℃에서 건조하여 과립을 형성한 후 포에 충진하였다.
하플로필럼 다우리쿰(Haplophyllum dauricum) 또는 하플로필럼 카파도시쿰(Haplophyllum cappadocicum)으로부터 분리한 다우리놀(daurinol)은 암세포의 세포주기 중 S 기를 선택적으로 정지시켜 세포주기를 교란하며, 순차적으로, 세포자살 반응을 일으킴으로써 항암 활성을 나타내므로 본 발명의 다우리놀은 세포주기 교란제 및 세포자살반응 유도제로서 사용가능하며, 암 예방 및 치료용 약학적 조성물로서 유용하게 이용될 수 있다.
도 1은 다우리놀(daurinol)에 의한 암세포 증식 억제 효과를 나타낸 그래프이다.
도 2는 다우리놀 처리 농도에 따른 인간 대장암 세포(HCT116)의 세포주기 변화를 나타낸 그래프이다:
A: 대조군;
B: 5 uM 다우리놀 처리군;
C: 10 uM 다우리놀 처리군; 및
D: 20 uM 다우리놀 처리군.

도 7은 상기 도 5의 실험을 통해 얻어진 오른쪽 하단 사분면(아넥신 V-FITC으로 염색되었지만, 요오드화 프로피디엄으로 염색되지 않은 세포를 의미함)에 해당하는 세포의 비율을 분석 후 막대그래프로 도시한, 다우리놀 처리에 의한 인간 대장암 세포(HCT116)에서 초기 세포자살반응 유도 세포의 비율을 나타낸 그래프이다.

Claims (13)
- 다우리놀(daurinol)을 유효성분으로 함유하는 암 예방 및 치료용 약학적 조성물.
- 제 1항에 있어서, 상기 다우리놀은 하플로필럼 다우리쿰(Haplophyllum dauricum) 또는 하플로필럼 카파도시쿰(Haplophyllum cappadocicum)으로부터 분리되는 것을 특징으로 하는 암 예방 및 치료용 약학적 조성물.
- 제 1항에 있어서, 상기 다우리놀은 세포주기 중 S 기를 선택적으로 정지시켜 세포주기를 교란하며, 순차적으로, 세포자살 반응을 일으킴으로써 항암 활성을 갖는 것을 특징으로 하는 암 예방 및 치료용 약학적 조성물.
- 제 1항에 있어서, 암은 후두암, 폐암, 식도암, 췌장암, 대장암, 간암, 위암, 설암, 피부암, 뇌종양, 자궁암, 유방암, 자궁경부암, 난소암, 신장암, 담낭암, 구강암, 결장암, 간암, 방광암 및 대장암으로 이루어진 군으로부터 선택되어지는 것을 특징으로 하는 암 예방 및 치료용 약학적 조성물.
- 제 1항에 있어서, 상기 약학적 조성물은 다우리놀이 세포주기를 교란함으로써 암 예방 및 치료를 하는 것을 특징으로 하는 암 예방 및 치료용 약학적 조성물.
- 제 5항에 있어서, 상기 다우리놀은 하플로필럼 다우리쿰(Haplophyllum dauricum) 또는 하플로필럼 카파도시쿰(Haplophyllum cappadocicum)으로부터 분리되는 것을 특징으로 하는 암 예방 및 치료용 약학적 조성물.
