Claims (14)
- 서열번호 1로 기재되는 담배 (Nicotiana tabacum)의 고온내성 유전자NtLHS1.
- 제 1항의 유전자NtLHS1를 포함하고도 1에 기재된 제한지도를 갖는 벡터 pGEM-T-Easy/NtLHS1.
- 제 2항의 벡터 pBADNH/NtLHS1를 포함하는 대장균 균주 DH5α/NtLHS1 (수탁번호: KCTC 0650BP).
- 제 1항의 유전자NtLHS1를 포함하고도 2에 기재된 제한지도를 갖는 발현벡터 pBADNH/NtLHS1.
- 제 1항의 유전자NtLHS1를 포함한 발현벡터를 포함하는 대장균 균주.
- 제 5항에 있어서, 발현벡터가 pBADNH/NtLHS1인 것을 특징으로 하는 대장균 균주 MC1061/NtLHS1.
- 제 1항의 유전자NtLHS1에 의해 코딩되고서열번호 2에 의해 기재되는 아미노산 서열을 갖는 고온내성 단백질 NtLHS1 또는 95% 이상의 아미노산 서열 상동성을 보이고 상기 NtLHS1과 동일한 활성을 갖는 변이형 단백질.
- 1) 제 5항의 대장균 균주를 배양한 후 발현유도물질을 첨가하여NtLHS1유전자의 발현을 유도하는 단계;2) 상기 배양액에서 세포를 수확하여 세포 용해액 (lysate)을 얻는 단계; 및3) 크로마토그래피를 이용하여 상기 세포 용해액을 정제하는 단계를 포함하는 제 7항의 단백질 NtLHS1 또는 그 변이형 단백질의 제조 방법.
- 제 8항에 있어서, 상기 대장균 균주는 제 4항의 균주 MC1061/NtLHS1이고, 상기 발현유도물질은 0.05 내지 0.3% (w/v)의 최종농도로 배양액에 첨가되는 아라비노즈이고, 상기 수확은 아라비노즈를 배양액에 첨가한 지 4시간 후에 수행하고, 상기 크로마토그래피는 Ni2+친화 크로마토그래피 및 젤 여과 크로마토그래피인 것을 특징으로 하는 제 5항의 단백질 NtLHS1 또는 그 변이형 단백질의 제조 방법.
- 제 1항의 유전자NtLHS1또는 제 7항의 단백질 NtLHS1의 변이형 단백질을 코딩하는 유전자를 진핵세포 또는 원핵세포에 도입하는 것으로 구성되는 고온내성을 가진 세포의 제조 방법.
- 제 10항에 있어서, 상기 진핵세포는 효모인 것을 특징으로 하는 고온내성을 가진 세포의 제조 방법.
- 제 10항에 있어서, 상기 원핵세포는 대장균인 것을 특징으로 하는 고온내성을 가진 세포의 제조 방법.
- 변성을 방지하고자 하는 단백질을 포함하는 계에 제 7항의 단백질 NtLHS1 또는 그 변이형 단백질을 첨가하는 것으로 구성되는 단백질의 고온변성 방지 방법.
- 제 1항의 유전자NtLHS1에 대한 안티센스 RNA 또는 제 7항의 단백질 NtLHS1의 변이형 단백질을 코딩하는 유전자에 대한 안티센스 RNA를 발현하는 유전자를 진핵세포 또는 원핵세포에 도입하는 것으로 구성되는 세포의 고온내성 억제 방법.
