Results of the PBMC-tumor cell co-culture assay are shown in FIG. Mouse 4T1 tumor cells that do not or stably express mouse Nectin-4 were co-cultured with or without BCY12141 with human PBMC and anti-CD3 and anti-CD28 stimulation for 48 hours. HTRF was then used to assay the concentration of the cytokine IL-2. The OX40-Nectin-4 bispecific heterotandem BCY12141 induced production of the cytokine IL-2 by human PBMCs in the presence of 4T1 cells expressing Nectin-4. In the absence of Nectin-4 expression in 4T1 cells, IL-2 levels were comparable to control baseline CD3+CD28 stimulation. This indicates that the OX40-nectin-4 bispecific molecule BCY12141 can induce an immune response in human PBMC only in the presence of cells expressing nectin-4.
