(免疫細胞に標的分子の局在を生成する工程)
本実施形態の免疫細胞の分析方法(以下、「方法」という)では、以下の工程を経て得られた免疫細胞を分析する:免疫細胞を含む全血と、免疫刺激因子とをインビトロで混合し、該免疫細胞の標的分子を標識する工程。この標的分子の標識工程について、以下に説明する。
本実施形態の分析方法では、上記の標的分子を標識する工程により得られた免疫細胞を分析する。具体的には、標識を検出することにより、免疫細胞における標的分子の局在に関する情報を取得する。
本発明の範囲には、細胞分析装置も含まれる。本発明のこの態様に係る実施形態について、図面を参照しつつ説明する。なお、以下の説明はすべて例示であって、本発明をこの説明に限定されない。
図2を参照して、細胞分析装置1は、測定装置2と情報処理装置3を備える。測定装置2は、捕捉体及び標識物質が結合している免疫細胞を含む試料をフローサイトメトリ法により光学的に測定する。情報処理装置3は、測定装置2による測定結果を解析し、解析結果を表示部320に表示する。本実施形態はこの例のみに限定されず、例えば、測定装置2及び情報処理装置3が一体的に構成された装置であってもよい。
実施形態1の解析処理のステップS53に代えて、CPU301は、ステップS52で抽出された測定データについて、X軸に蛍光シグナル面積値を割り当て、Y軸に細胞のサイズを割り当てた2Dスキャッタグラムを作成してもよい(図11d)。CPU301は、2Dスキャッタグラム中のR4と表示された所定の領域(以下、「所定の範囲(R4)」と呼ぶ)にある測定データ数を計数してもよい。範囲(R4)は、蛍光シグナル面積値(X軸)においてCD28の局在を反映する所定の範囲でゲーティングし、かつ細胞のサイズ(Y軸)において細胞単独の大きさを反映する所定の範囲でゲーティングする。範囲(R4)における測定データを計数することにより、免疫刺激に対して応答性の免疫細胞の数が計数される。この実施形態において、範囲(R4)にあるドットに対応する細胞は、CD28の局在が生成されている傾向がある。よって、範囲(R4)内のドットを計数することにより、CD28の局在が生成した免疫細胞が計数される。
測定データの解析において、2Dスキャッタグラムを用いずに、免疫細胞における標的分子の局在に関する分析を行ってもよい。例えば、実施形態1の解析処理のステップS53に代えて、CPU301は、ステップS52で抽出された細胞の測定データについて、X軸に蛍光シグナル面積値の階級値を割り当て、Y軸に度数を割り当てたヒストグラムを作成して、表示部320に表示してもよい。実施形態1の解析処理のステップS54に代えて、CPU301は、このヒストグラムから、ステップS52で抽出された測定データの、蛍光シグナル面積値の平均値、中央値及び最頻値の少なくとも1つを算出してもよい。得られた値を用いて、さらに解析してもよい。例えば、被検者の全血由来の試料から得た蛍光シグナル面積値の最頻値と、健常者の全血由来の試料から得た蛍光シグナル面積値の標準化された最頻値とを比較してもよい。この解析において、被検者の上記最頻値を示す階級値が、健常者の標準化された最頻値を示す階級値よりも小さい場合、被検者の全血中の免疫細胞では、健常者の全血中の免疫細胞よりも、CD28の局在が生成していると判定できる。
実施形態1〜3の解析処理では、撮像された画像データに基づいた解析を例示したが、画像データ以外の測定データから、免疫細胞における標的分子の局在に関する分析を行うことができる。撮像部203の変形例として、例えば図12を参照して、撮像部203は、光源203a、203bと、フローセル203cと、集光レンズ203d、203e、203jと、ダイクロックミラー203kと、蛍光受光部203lと、前方散乱受光部203mとを備えた、光学シグナルを検出する検出部であってもよい。
被検者から採取した全血(末梢血)に免疫刺激因子として抗CD28抗体を添加することにより、全血中の免疫細胞(T細胞)を刺激して、該免疫細胞上のCD28の局在に関する情報を取得した。対照として、免疫刺激因子を添加していない全血中の免疫細胞について測定した。
(1) 刺激した免疫細胞の測定
5名の被検者(健常ボランティア)から全血を採取した。各被検者の全血(200μL)をチューブに分取し、マウス抗ヒトCD28抗体(#302914 Clone:CD28.2:BioLegend社)を添加して穏やかに撹拌した。抗体を含む全血を200×gで2分間、室温で遠心分離してバフィーコートを形成した。バフィーコートをCO2インキュベーター内(37℃)で約2〜120分間静置することにより、分析対象の免疫細胞と他の白血球とを接触した後、4% PFAを含む固定液を添加して、免疫細胞の細胞膜を固定化処理した。抗ヒトCD28抗体の添加から4% PFAの添加までの時間は約5〜125分であった。次に、溶血剤(0.4〜0.8%塩化アンモニウム)を添加して、溶血処理を行った。その後、PE標識ヤギ抗マウスIgG抗体(#405307:BioLegend社)、APC標識マウス抗ヒトCD3抗体(#300411 Clone: UCHT1:BioLegend社)及びHoechst(H342:同仁化学)を用いて細胞及び細胞核を染色した。染色した細胞を1% BSA/PBSに懸濁し、イメージングフローサイトメータ(ImageStreamX MarkII Imaging Flow Cytometer:Amnis社)を用いて測定した。
マウス抗ヒトCD28抗体を添加しなかったこと以外は上記(1)と同様にして、上記の健常ボランティアの全血(200μL)に含まれる免疫細胞を処理して、IFCを用いて測定した。
IFCによる測定で得られた、刺激した免疫細胞を含む全血の測定データと、刺激しなかった免疫細胞を含む全血の測定データの多重パラメータ解析を行った。簡単には、IFCで取得した個々の細胞の透過光画像及び蛍光画像(CD28)に基づいて画像解析し、細胞のサイズ、細胞のアスペクト比、蛍光シグナル面積値及び総蛍光シグナル強度を取得した。まず、個々の細胞の透過光画像のデータに基づいて、X軸に細胞サイズをとり、Y軸に細胞のアスペクト比をとった2Dスキャッタグラムを作成した(図示せず)。赤血球ゴーストなどのデブリスの測定データを除くため、この2Dスキャッタグラムの所定の範囲(Cells)内にあるドットを選択し、選択した各ドットに対応する測定データを、細胞の測定データとして抽出した。
図16Aから分かるように、抗CD28抗体による免疫刺激を与えなかったT細胞では、CD28分子は細胞膜上に一様に分布した。これに対して、図16Bから分かるように、免疫刺激を与えたT細胞では、CD28分子は、細胞膜上の一部に局在していた。CD28の蛍光画像における、この局在を示した部分が、形成された免疫シナプスを示すと考えられる。図15A及びBの2Dスキャッタグラムにおいて、蛍光シグナル面積値が小さい値を示す細胞は、免疫シナプスを形成している傾向がある。よって、蛍光シグナル面積値が小さい領域をゲーティングして、免疫刺激応答性を示す免疫細胞が出現する領域(範囲(IS area2))を示した。図15A(刺激しなかった免疫細胞)と図15B(刺激した免疫細胞)とを比較すると、図15Bにおいて、ゲーティングした領域(範囲(IS area2))に細胞が顕著に出現していることが分かる。
試料として全血を用いる本実施形態の方法と、試料として全血から単離したPBMC(末梢血単核細胞)を用いる従来法とを、CD28局在陽性率を指標として比較した。
(1) 全血中の免疫細胞の測定
健常ボランティアから全血を採取した。採取した全血の一部(200μL)を用いて、実施例1と同様にして、抗ヒトCD28抗体で刺激した免疫細胞及び刺激していない免疫細胞の測定を行った。
上記の全血の一部(200μL)を取り、フィコールを用いる密度勾配遠心法によりPBMCを単離した。単離したPBMCをチューブに分取し、実施例1と同じ抗ヒトCD28抗体を添加し、200×gで2分間、室温で遠心分離して細胞を沈降させた。細胞をCO2インキュベーター内(37℃)で約2〜120分間静置することにより免疫細胞とチューブの壁面とを接触した後、4% PFAを含む固定液を添加して、免疫細胞の細胞膜を固定化処理した。次に、実施例1と同じ溶血剤を添加して、溶血処理を行った。その後、実施例1と同じPE標識ヤギ抗マウスIgG抗体、APC標識マウス抗ヒトCD3抗体及びHoechstを用いて細胞及び細胞核を染色した。実施例1と同様にして、染色した細胞をIFCで測定した。免疫刺激しなかった細胞の測定は、マウス抗ヒトCD28抗体を添加しなかったこと以外は上記と同様にして、全血から単離したPBMCを処理して、IFCを用いて測定した。
実施例1と同様にして、IFCによる測定で得られたデータの多重パラメータ解析を行った。全血又は単離したPBMC中のT細胞に対する、CD28が局在したT細胞の割合(CD28局在陽性率)を算出した。結果を図18に示す。
図18より、刺激なしの場合、PBMC及び全血におけるT細胞のCD28局在陽性率は同程度の値を示した。一方、刺激ありの場合、PBMCにおけるT細胞のCD28局在陽性率は、全血よりも顕著に低い値を示した。これは、全血からPBMCを分離及び単離する工程に起因する免疫細胞の質及び機能の変化を示唆すると考えられる。本実施形態の方法では、PBMCを分離及び単離する工程を行わないので、当該工程に起因する質及び機能への影響が無くなる。よって、本実施形態の方法は、PBMCを用いる従来法よりも測定の精度が向上することが示された。
試料として全血を用いる本実施形態の方法と、試料として全血から単離したPBMC(末梢血単核細胞)を用いる従来法とを、ノイズシグナルを指標として比較した。
実施例1と同様にして、5名の被検者から採取した全血を用いて、抗CD28抗体を添加せずに全血中のT細胞を測定し、CD28局在陽性率を算出した。また、実施例2と同様にして、4名の被検者から採取した全血からPBMCを単離し、抗CD28抗体を添加せずにPBMC中のT細胞を測定し、CD28局在陽性率を算出した。結果を図19に示す。
図19より、刺激なしの条件での全血及びPBMCにおけるT細胞のCD28局在陽性率を比較すると、PMBCにおいてデータのばらつきやノイズシグナルが見られた。これは、全血からPBMCを分離及び単離する工程が、免疫細胞の質及び機能に影響することを示唆すると考えられる。一方、全血を用いる本実施形態の方法では、PBMCを分離及び単離する工程を行わないので、当該工程に起因するデータのばらつきやノイズシグナルが低減された。よって、本実施形態の方法は、PBMCを用いる従来法よりも測定の精度が向上することが示された。
分析対象の免疫細胞としてT細胞クローンを用いて、本実施形態の方法及び従来法による測定を行い、T細胞クローンのCD28局在陽性率を比較した。
5×106 cellsのCD4陽性細胞(ST-70026:Stemcell Technologies社)及びCD8陽性細胞(0508-100:フナコシ、PB08C-1:コスモバイオ)を、1μg/mL LEAF(商標)精製抗ヒトCD3抗体(317315:Biolegend社)、10 ng/mL 組換え型ヒトIL-2(rhIL-2)(202-IL-500:R&D systems社)、0.2μg/mL フィトヘマグルチニン-L(PHA)(L4144-5MG:SIGMA社)及び2%ヒト血清(H4522-100ML:SIGMA社)を含むYssel's培地(400102:Gemini Bio-Products社)に播種し、1×106 cellsのPBMC及び1×104 cellsのJY細胞と14日間、共培養した。14日後、T細胞を限界希釈法により0.3、1.0及び3.0 cells/wellとなるように96ウェルプレートに播種し、コロニーが形成されるまで、1×104 cellsのPBMCと共培養した。コロニーが形成されたウェルからT細胞を回収して、T細胞クローンを樹立した。
(1) 本実施形態の方法によるT細胞クローンの測定
Hoechstで標識したT細胞クローン(5×105 cells/100μL)を、健常ボランティアから採取した全血(200μL)に添加し、そして実施例1と同じ抗ヒトCD28抗体を添加した。実施例1と同様にして、バフィーコートを形成した。バフィーコートをCO2インキュベーター内(37℃)で約2〜120分間静置することにより、T細胞クローンを含む免疫細胞と他の白血球とを接触した後、4% PFAを含む固定液を添加して、免疫細胞の細胞膜を固定化処理した。次に、実施例1と同じ溶血剤を添加して、溶血処理を行った。その後、実施例1と同じPE標識ヤギ抗マウスIgG抗体、APC標識マウス抗ヒトCD3抗体を用いて細胞を染色した。実施例1と同様にして、染色した細胞をIFCで測定した。免疫刺激しなかったT細胞クローンの測定は、マウス抗ヒトCD28抗体を添加しなかったこと以外は上記と同様にして、全血に添加したT細胞クローンをIFCで測定した。
Hoechstで標識したT細胞クローン(5×105 cells/100μL)を48ウェルプレートに播種し、実施例1と同じ抗ヒトCD28抗体を添加した。200×gで2分間、室温で遠心分離して細胞をプレート内に沈降させた。細胞をCO2インキュベーター内(37℃)で約2〜120分間静置することにより免疫細胞とプレートの底面とを接触した後、4% PFAを含む固定液を添加して、免疫細胞の細胞膜を固定化処理した。次に、実施例1と同じ溶血剤を添加して、溶血処理を行った。その後、実施例1と同じPE標識ヤギ抗マウスIgG抗体、APC標識マウス抗ヒトCD3抗体及びHoechstを用いて細胞及び細胞核を染色した。実施例1と同様にして、染色した細胞をIFCで測定した。免疫刺激しなかった細胞の測定は、マウス抗ヒトCD28抗体を添加しなかったこと以外は上記と同様にして、48ウェルプレートに播種したT細胞クローンをIFCで測定した。
実施例1と同様にして、IFCによる測定で得られたデータの多重パラメータ解析を行った。全血又はプレートに添加したT細胞クローンに対する、CD28が局在したT細胞クローンの割合(CD28局在陽性率)を算出した。結果を図20A及びBに示す。図中、「P」は、プレートを用いた従来法の条件を示し、「B」は、全血を用いた本実施形態の方法の条件を示す。
図20A及びBから分かるように、T細胞クローンのCD28局在陽性率は、全血中にT細胞クローンを添加した場合と、プレート中にT細胞クローンを播種した場合との間で顕著な差は認められなかった。これまで、全血に含まれる免疫細胞以外の血球成分は、免疫刺激による標的分子の局在及び測定に影響すると考えられてきた。しかし、本実施形態の方法では、全血中で免疫刺激しても、従来法と同様に、免疫刺激応答性を示す免疫細胞(IS形成能を示す免疫細胞)を検出できることが示された。
2種類の溶血剤を用いて、本実施形態の方法及び従来法による測定を行い、免疫細胞のCD28局在陽性率を比較した。分析対象の免疫細胞として、実施例4のT細胞クローンを用いた。
(1) 本実施形態の方法によるT細胞クローンの測定
溶血剤1(0.8%塩化アンモニウム)又は溶血剤2(0.41%塩化アンモニウム及び0.2%界面活性剤)を用いたこと以外は、実施例4と同様にして、全血に添加したT細胞クローンをIFCで測定した。溶血剤1及び2はいずれも、溶血用試薬として市販されている製品であった。
実施例4と同様にして、48ウェルプレートに播種したT細胞クローンをIFCで測定した。
実施例1と同様にして、IFCによる測定で得られたデータの多重パラメータ解析を行った。全血又はプレートに添加したT細胞クローンに対する、CD28が局在したT細胞クローンの割合(CD28局在陽性率)を算出した。結果を図21に示す。図中、「P」は、プレートを用いた従来法の条件を示し、「B」は、全血を用いた本実施形態の方法の条件を示す。また、「1」は、溶血剤1を用いたことを示し、「2」は、溶血剤2を用いたことを示す。
図21から分かるように、溶血剤1及び2のいずれを用いた場合も、T細胞クローンのCD28局在陽性率に顕著な差は認められなかった。本実施形態の方法では、市販の溶血用試薬を用いれば、免疫刺激応答性を示す免疫細胞(IS形成能を示す免疫細胞)を検出できることが示された。
これまでの実施例では、免疫刺激した免疫細胞と、全血の遠心分離により形成した赤血球層とを接触していたが、他の接触方法でも本実施形態の方法を実施できるかを検討した。分析対象の免疫細胞として、実施例4のT細胞クローンを用いた。
(1) 遠心分離による接触
Hoechstで標識したT細胞クローン(5×105 cells/100μL)を、健常ボランティアから採取した全血(200μL)に添加し、そして実施例1と同じ抗ヒトCD28抗体を添加した。実施例1と同様にして、遠心分離によりバフィーコートを形成した。バフィーコートをCO2インキュベーター内(37℃)で約2〜120分間静置することにより、T細胞クローンを含む免疫細胞と他の白血球とを接触した。また、抗ヒトCD28抗体を添加しなかったこと以外は、上記と同様にして、刺激していないT細胞クローンを含む免疫細胞と赤血球層とを接触した。
Hoechstで標識したT細胞クローン(5×105 cells/100μL)を、健常ボランティアから採取した全血(200μL)に添加し、そして実施例1と同じ抗ヒトCD28抗体を添加した。刺激したT細胞クローンを含む全血を収容したチューブを、ローテーターにセットして、30 rpmで約2〜120分間、CO2インキュベーター内(37℃)で転倒混和した。
Hoechstで標識したT細胞クローン(5×105 cells/100μL)を、健常ボランティアから採取した全血(200μL)に添加し、そして実施例1と同じ抗ヒトCD28抗体を添加した。刺激したT細胞クローンを含む全血を収容したチューブをCO2インキュベーター内(37℃)で約2〜120分間静置した。
接触面の提供後、T細胞クローンを含む全血に4% PFAを含む固定液を添加して、免疫細胞の細胞膜を固定化処理した。次に、実施例1と同じ溶血剤を添加して、溶血処理を行った。その後、実施例1と同じPE標識ヤギ抗マウスIgG抗体、APC標識マウス抗ヒトCD3抗体及びHoechstを用いて細胞及び細胞核を染色した。実施例1と同様にして、染色した細胞をIFCで測定した。
実施例1と同様にして、IFCによる測定で得られたデータの多重パラメータ解析を行った。全血又はプレートに添加したT細胞クローンに対する、CD28が局在したT細胞クローンの割合(CD28局在陽性率)を算出した。結果を図22に示す。
図22から分かるように、遠心分離、ローテーション及び静置のいずれの接触方法によっても、CD28局在陽性率は、刺激しなかったT細胞クローンに比べて、顕著に高い値を示した。ローテーション及び静置によってもCD28局在陽性率は増加したことから、全血を収容するチューブの内壁面との接触も、免疫細胞への免疫刺激に寄与していることが示唆される。本実施形態の方法では、いずれの接触方法によっても、免疫刺激応答性を示す免疫細胞(IS形成能を示す免疫細胞)を検出できることが示された。
被験者から採取した全血(末梢血)に、免疫刺激因子を有する他家細胞(ヒトB細胞)を添加することにより、全血中の免疫細胞(T細胞及びNK細胞)を刺激して、該免疫細胞の構造タンパク質であるFアクチンの局在に関する情報を取得した。対照として、他家細胞を添加していない全血中の免疫細胞について測定した。
(1) 免疫刺激した免疫細胞の測定
健常人ボランティア血(200μL)をチューブに分取し、ここに他家細胞としてヒトB細胞(理化学研究所、製品番号HEV0032)を添加した。得られた全血試料を遠心分離して、バフィーコートを形成させた。10分以上の反応後、4%PFAを含む固定液で試料中の免疫細胞の細胞膜を固定化処理した。次に、固定化した細胞に溶血剤を添加し、溶血処理を行った。その後、Alexa488-マウス抗ヒトCD3抗体、PE-Cy7-マウス抗ヒトCD56抗体、及びAlexa674-ファロイジンを用いて細胞を染色した。染色した細胞をIFC(ImageStreamX MarkII Imaging Flow Cytometer:Amnis社)を用いて測定した。
健常人ボランティア血(200μL)をチューブに分取し、遠心分離によりバフィーコートを形成させた。2分以上の反応後、4%PFAを含む固定液で試料中の免疫細胞の細胞膜を固定化処理した。次に、固定化した細胞に溶血剤を添加し、溶血処理を行った。その後、Alexa488-マウス抗ヒトCD3抗体、PE-Cy7-マウス抗ヒトCD56抗体、及びAlexa674-ファロイジンを用いて細胞を染色した。染色した細胞をIFC(ImageStreamX MarkII Imaging Flow Cytometer:Amnis社)を用いて測定した。
IFCによる測定で得られた、刺激した免疫細胞を含む全血の測定データと、刺激しなかった免疫細胞を含む全血の測定データの多重パラメータ解析を行った。簡単には、IFCで取得した個々の細胞の透過光画像及び蛍光画像(CD3、CD56及びFアクチン)に基づいて画像解析し、細胞のサイズ、細胞のアスペクト比、蛍光シグナル面積値及び総蛍光シグナル強度を取得した。まず、個々の細胞の透過光画像のデータに基づいて、X軸に細胞サイズをとり、Y軸に細胞のアスペクト比をとった2Dスキャッタグラムを作成した(図示せず)。赤血球ゴーストなどのデブリスの測定データを除くため、この2Dスキャッタグラムの所定の範囲(Cells)内にあるドットを選択し、選択した各ドットに対応する測定データを、細胞の測定データとして抽出した。

ヒトB細胞株を含む免疫刺激因子との接触を与えなかったT細胞及びNK細胞では、Fアクチン分子は細胞膜上に一様に分布した。これに対して、当該免疫刺激を与えた場合では、Fアクチン分子はT細胞及びNK細胞の膜上の一部に局在していることが分かった。この局在を示す部分が、形成された免疫シナプスを示すと考えられる(図25参照)。2Dスキャッタグラムでは、Y軸に細胞におけるFアクチン蛍光シグナルの面積値を反映する情報を、X軸に細胞におけるFアクチンの総蛍光シグナル値を反映する情報を示す(図24参照)。ここで、局在面積が小さい値を示す細胞は、ISが形成されている傾向があるため、この領域をゲーティングして免疫刺激応答性を示す免疫細胞が出現する領域を示した。図24を参照して、刺激なし及び刺激ありの2Dスキャッタグラムを比較すると、刺激ありの場合において、ゲーティングエリアに細胞が顕著に出現していることが分かる。図26A及びBより、刺激なしの場合、全血中のT細胞及びNK細胞のIS陽性率は非常に低い値を示した。刺激なしの場合に比べて、刺激ありの場合では、全血中のT細胞及びNK細胞のIS陽性率の値は有意に増加した。

Claims (25)
- 免疫細胞を含む全血と免疫刺激因子とをインビトロで混合し、前記免疫細胞の標的分子を標識する工程と、
前記標識を検出することにより、前記免疫細胞における前記標的分子の局在に関する情報を取得する工程と
を含む、免疫細胞の分析方法。 - 前記標的分子を標識する工程が、前記免疫細胞と、前記免疫細胞とは異なる物質とを接触する工程を含む請求項1に記載の方法。
- 前記接触工程が、前記免疫細胞を含む全血及び前記免疫刺激因子の混合物を、遠心分離、撹拌、又は静置することにより行われる請求項2に記載の方法。
- 前記免疫細胞とは異なる物質が、前記標的分子を標識される免疫細胞以外の血球、又は前記全血を収容する容器の内表面である請求項2又は3に記載の方法。
- 前記接触工程が、前記免疫細胞を含む全血及び前記免疫刺激因子の混合物を遠心分離して、前記免疫細胞を含む層と、赤血球を含む層とを形成することにより行われる請求項2〜4のいずれか1項に記載の方法。
- 前記標的分子を標識する工程と、前記標的分子の局在に関する情報を取得する工程との間に、前記免疫細胞を固定する工程を含む請求項1〜5のいずれか1項に記載の方法。
- 前記標的分子を標識する工程と、前記標的分子の局在に関する情報を取得する工程との間に、前記全血に含まれる赤血球を溶解する工程を含む請求項1〜6のいずれか1項に記載の方法。
- 前記赤血球を溶解する工程が、前記免疫細胞を固定する工程と、前記標的分子の局在に関する情報を取得する工程との間に行われる請求項6を引用する請求項7に記載の方法。
- 前記標的分子の局在に関する情報を取得する工程において、前記免疫細胞の前記標的分子と、前記標的分子に結合する捕捉体と、検出可能な光学シグナルを生じうる標識物質とを結合することにより、前記標的分子を標識する請求項1〜8のいずれか1項に記載の方法。
- 前記免疫刺激因子が、前記標的分子に結合する捕捉体として共用される請求項9に記載の方法。
- 前記標識物質が、前記捕捉体に直接的又は間接的に結合することにより、前記標的分子を標識する請求項9又は10に記載の方法。
- 前記標識物質から生じた光学シグナルを検出し、検出した光学シグナルに基づいて、前記免疫細胞における前記標的分子の局在に関する情報を取得する請求項9〜11のいずれか1項に記載の方法。
- 前記光学シグナルに基づいて、前記免疫細胞における標識された標的分子の分布を反映する値を取得し、取得した値に基づいて、前記標的分子の局在が生成している免疫細胞を計数する請求項12に記載の方法。
- 前記全血に含まれる免疫細胞が、T細胞サブセット又はNK細胞サブセットである請求項1〜13のいずれか1項に記載の方法。
- 前記標的分子の局在が生成している免疫細胞の数に基づいて、前記T細胞サブセット又は前記NK細胞サブセットの免疫刺激応答性の指標を取得する請求項14に記載の方法。
- 前記全血に含まれる免疫細胞の総数を計数し、前記標的分子の局在が生成している免疫細胞の数と、前記総数に基づいて、前記T細胞サブセット又は前記NK細胞サブセットの免疫刺激応答性の指標を取得する請求項15に記載の方法。
- 前記標的分子が、表面抗原、及び構造タンパク質の重合体から選択される少なくとも1つである請求項1〜16のいずれか1項に記載の方法。
- 前記表面抗原が、免疫シナプスを構成する分子である請求項17に記載の方法。
- 前記表面抗原が、CD28、CD3、TCRα/β、CD40L、OX40、CTLA4、PD-1及びICOSから選択される少なくとも1つであり、前記構造タンパク質の重合体がFアクチンである請求項17又は18に記載の方法。
- 前記免疫刺激因子が、免疫刺激抗体、免疫刺激ペプチド、主要組織適合性分子(MHC分子)、及びMHC分子と抗原ペプチドとの複合体からなる群より選択される少なくとも1つを含む請求項1〜19のいずれか1項に記載の方法。
- 前記免疫刺激因子が、他家細胞、抗原提示細胞、及び腫瘍細胞からなる群より選択される少なくとも1つの細胞上に存在する免疫刺激因子を含む請求項1〜19のいずれか1項に記載の方法。
- 前記免疫刺激因子による前記免疫細胞の刺激時間が、6時間未満である請求項1〜21のいずれか1項に記載の方法。
- 免疫細胞の分析方法であって、
標識された標的分子を有する免疫細胞の前記標識を検出することにより、前記免疫細胞における前記標的分子の局在に関する情報を取得する工程を含み、
前記標識された標的分子を含む免疫細胞が、免疫細胞を含む全血と免疫刺激因子とをインビトロで混合し、前記全血に含まれる免疫細胞の標的分子を標識することにより調製された細胞である、
