Abstract
Description
本発明は、LAG3+ Tregによる作用メカニズムを解明すると共に、この知見に基づく新規なB細胞活性化の抑制剤および自己免疫疾患の治療剤を提供することを課題とする。
〔1〕TGF-beta3を含むB細胞活性化の抑制剤。
〔2〕前記B細胞は自己反応性B細胞である〔1〕に記載の抑制剤。
〔3〕前記TGF-beta3はB細胞による抗体産生を抑制する〔1〕又は〔2〕に記載の抑制剤。
〔4〕〔1〕〜〔3〕のいずれかに記載の抑制剤を含む自己免疫疾患の治療剤。
〔5〕TGF-beta3と抗体又は抗体断片とが連結された分子を含む自己免疫疾患の治療剤。
〔6〕前記抗体又は抗体断片はB細胞を認識する可変領域を含む〔5〕に記載の治療剤。
〔7〕前記可変領域はCD19、CD20、CD40、CD22、IL21R、BAFF-R、BCMA、TACI、CD27、又はCD138に含まれるいずれか1つまたは複数をマーカーとして前記B細胞を認識する〔6〕に記載の治療剤。
〔8〕TGF-beta3を認識する第一の可変領域と、B細胞を認識する第二の可変領域とを含む多重特異性抗体を含む自己免疫疾患の治療剤。
〔9〕前記自己免疫疾患は全身性エリテマトーデス、天疱瘡、多発性硬化症、視神経脊髄炎、ANCA関連血管炎、関節リウマチ、移植臓器拒絶、シェーグレン症候群、若年性皮膚筋炎、重症筋無力症、又はバセドウ病、橋本病等の自己免疫性甲状腺疾患のいずれかである〔4〕〜〔8〕のいずれかに記載の治療剤。
〔10〕〔4〕〜〔8〕のいずれかの治療剤を含む医療用キット。
また、本発明は以下にも関する。
〔11〕TGF-beta3;TGF-beta3と抗体若しくは抗体断片とが連結された分子;又はTGF-beta3を認識する第一の可変領域と、B細胞を認識する第二の可変領域とを含む多重特異性抗体を投与する工程を含む、B細胞活性化の抑制方法。
〔12〕TGF-beta3;TGF-beta3と抗体若しくは抗体断片とが連結された分子;又はTGF-beta3を認識する第一の可変領域と、B細胞を認識する第二の可変領域とを含む多重特異性抗体を投与する工程を含む、自己免疫疾患の治療方法。
〔13〕B細胞活性化の抑制において用いるための、TGF-beta3;TGF-beta3と抗体若しくは抗体断片とが連結された分子;又はTGF-beta3を認識する第一の可変領域と、B細胞を認識する第二の可変領域とを含む多重特異性抗体。
〔14〕自己免疫疾患の治療において用いるための、TGF-beta3;TGF-beta3と抗体若しくは抗体断片とが連結された分子;又はTGF-beta3を認識する第一の可変領域と、B細胞を認識する第二の可変領域とを含む多重特異性抗体。

マウス
C57BL/6(B6)、C57BL/6-Faslpr/lpr(B6/lpr)、C57BL/6-FasLgld/gld(B6/gld)、MRL-Faslpr/lpr(MRL/lpr)、およびMRL-Fas+/+(MRL/+)マウスは、日本エスエルシー株式会社から購入した。B6リコンビナーゼ活性化遺伝子(Rag)-1欠損(Rag1KO)マウス、floxed-Prdm1(Prdm1fl/fl)マウス、Il10欠損(IL-10KO)マウス、T細胞受容体(TCR)トランスジェニックOT-IIマウス(MHCクラスII I-Abについてはトリ卵白アルブミン(アミノ酸残基323〜339)に特異的)、およびTEaマウス(I-AbについてはMHCクラスII I-Eα分子由来のEαペプチド(アミノ酸残基52〜68)に特異的)は、ジャクソン研究所から購入した。Floxed-Stat3(Stat3fl/fl)マウスは、株式会社オリエンタルバイオサービス(日本)から購入した。Rag1KOマウスは、滅菌濾過空気下のマイクロアイソレーターケージに収容した。B6-Pdcd1欠損(PD-1KO)マウス(非特許文献47)は、RIKEN BRC(日本)から購入した。Floxed Egr2(Egr2fl/fl)マウスは、Patrick Charnay氏(INSERM、フランス)より供与された(非特許文献48)。Egr2 コンディショナルノックアウト(CKO)マウス(Egr2fl/fl CD4-Cre+)、Prdm1 CKOマウス(Prdm1fl/fl CD4-Cre+)、およびSTAT3 CKO(Stat3fl/fl CD4-Cre+)は、Egr2fl/flマウス、Prdm1fl/flマウス、またはStat3fl/flマウスをB6バックグラウンドのCD4-Creトランスジェニックマウスとそれぞれ交配することによって作製した。CD4-Creトランスジェニックマウス(4196系)、およびStat1欠損(STAT1KO)マウスは、Taconicから購入した。全ての実験について、7週齢以上の同週齢同性のマウスを使用した。全ての動物実験は、東京大学動物実験倫理委員会によって承認された。
以下の試薬は、BD Pharmingenから購入した; CD3εに対する精製モノクローナル抗体(mAb)(145-2C11)、FasLブロッキング(MFL3)、抗CD40(3/23)、Fcブロック(抗CD16/CD32)、FITC抗CD45RB(16A)、FITC抗Fas(Jo2)、PE抗CD45RB(16A)、APC-Cy7抗CD45RB(16A)、PE抗LAG3(C9B7W)、APC抗LAG-3(C9B7W)、FITC抗IgG1(A85-1)、APC抗IgG1(A85-1)、FITC抗GL7(Ly-77)、FITC抗CD25(PC61)、PE抗CD25(PC61)、APC抗CD25(PC61)、APC-Cy7抗CD25(PC61)、APC抗CD4(L3T4)、APC-Cy7抗CD4(L3T4)、PE抗CXCR5(2G8)、APC-Cy7抗B220(RA3-6B2)、PE抗PD-1(J43);CD8a(53-6.7)、CD19(1D3)、CD11c(HL3)、CD45RB(16A)、CD25(7D4)、およびCXCR5(2G8)に対するビオチン化mAb;ストレプトアビジン(SA)-FITC抗体(Ab)、SA-APC、ならびにSA-APC-Cy7。Alexa Fluor 488 抗LAG3 mAb(C9B7W)およびFITC抗PD-L1 mAb(MIH6)は、AbD Serotecから購入した。Qdot605抗CD4 mAb(RM4-5)およびSA-Qdot605は、Invitrogenから購入した。PE抗PD-L1 mAb(MIH5)、PE抗Egr2 mAb(erongr2)、PE抗PD-L1 mAb(10F. 9G2)、およびAPC抗B220 mAb(RA3-6B2)は、eBioscienceから購入した。NP(13)-OVAおよびNP(9)-BSAは、Biosearch Technologiesから購入した。SAコンジュゲートマイクロビーズは、Miltenyi Biotecから購入した。FITC抗マウスIgG Abは、Sigmaから購入した。抗PD-L1ブロッキングmAb(10F.9G2)は、Biolegendから購入した。Alexa 488 Fluor抗GFP mAbは、Medical & Biological Laboratoriesから購入した。組換えTGF-beta1(rTGF-beta1)およびrTGF-beta3はMiltenyi Biotecから購入し、rTGF-beta2、抗TGF-beta3ブロッキングポリクローナルAb(MAB234)、およびrIL-27は、R&D Systemsから購入した。
ヒト由来細胞を用いた試験に関しては、以下の抗ヒトmAbを使用した:V450抗hCD4(RPA-T4)、V500抗hCD4(RPA-T4)(どちらもBD Biosciences製);PerCP-Cy5.5抗ヒトCD3(UCHT1)、Brilliant Violet 421抗hCD25(BC96)、APC抗hCD19(HIB19)、PerCP-Cy5.5抗hCD4(OKT4)、Alexa Fluor 647抗hCD197(G043H7)、APC-Cy7抗ヒトCD45RA(HI100)(いずれもBioLegend製); Alexa Fluor 488抗hCD25(BC96)、およびPE-Cy7抗hCD127(eBioRDR5)(いずれもeBioscience製)。PE抗hLAG3 ポリクローナルAbはR&D Systemsから購入した。
T細胞およびB細胞は、10% FBS、100μg/ml L-グルタミン、100 U/mlペニシリン、100μg/mlストレプトマイシン、および50μM 2-メルカプトエタノール(全てSigmaから購入した)を添加したRPMI-1640培地中で培養した。
ゲノムEgr2遺伝子座全体を包含する細菌人工染色体(BAC)クローンRP23-88D4は、BACライブラリ(Invitrogen)から入手した。Red/ET組み換え系を用いてEgr2エキソン1の開始コドンにSV40 polyA配列と共に高感度緑色蛍光タンパク質遺伝子(eGFP)を挿入して、このクローンを改変した。PI-SceIを用いて切断した直鎖状Egr2-eGFPコンストラクトを、B6マウス由来の受精した接合子の前核に注入し、偽妊娠雌に移した。以下を用いた尾部より抽出したDNAのPCRによって、出生仔の遺伝子導入を確認した:

SV40ポリA特異的プライマー:リバース5'-TGAGTTTGGACAAACCACAACTAGA-3'(配列番号:8)
(PCR産物のサイズ:2.1 kb)。F1ヘテロ接合体トランスジェニック雄性マウスと野生型(WT)雌性マウスを交配させることにより、マウスを作製した。
マウス脾臓は細切し、IV型コラゲナーゼ(Sigma-Aldrich)で処理した。ACK(ammonium chloride with potassium)溶解緩衝液中での低浸透圧ショックにより赤血球を溶解させた後、すぐに等浸透圧に回復させた。FcRブロッキング抗体(抗マウスCD16/CD32 mAb)の存在下、2%FCSを含む氷冷PBS中で表面染色を行った。始めに、高純度のCD4+ T細胞を得るために、抗B220 mAb、抗CD19 mAb、抗CD11c mAb、および抗CD8a mAbを用いたMACS(magnetic-activated cell sorting;Miltenyi Biotec)によるネガティブセレクションによって、単細胞懸濁液を精製した。続いて、高純度のCD4+CD25-CD45RBlowLAG3+ T細胞を得るために、抗CD45RB mAbを用いてCD45RBhigh細胞を除去した。CD4+CD25-CD45RBlowLAG3+ T細胞(LAG3+ Treg)、CD4+CD25+ T細胞(CD25+ Treg)、CD4+CD25-LAG3- ヘルパーT細胞(Th細胞)、またはCD4+CD25-CD62LhiCD44low(ナイーブT)を単離するために、FcRブロッキングした細胞をCD4、CD25、CD45RB、およびLAG3に特異的なmAbで染色した。細胞内抗Egr2染色用の細胞は、製造元のプロトールに従ってFoxp3 Staining Buffer Set(eBioscience)を用いて染色した。MACS選別した細胞およびFACS(FACSVantage SE(Becton-Dickinson)またはMoFlo XDP(Beckman Coulter))選別した細胞の純度は、それぞれ90%超および99%超であった。
NP(13)-OVA(Biosearch Technologies)のPBS溶液とalum(Pierce)を1:1比で30分間4℃で混合することによって、NP-OVAのalum溶液を調製した。NP-OVA/alumによる免疫は、腹腔内注射によって実施した。
FACSで精製したB6マウス由来のLAG3+ Treg 2×105個を、細胞移入の1日前に100μg NP-OVA/alumで予備免疫した、またはしていない10週齢Egr2CKOマウスに静脈内注射した。脾臓におけるTFHおよびGCBの分化は、細胞移入の7日後にFACSにより解析した。血清中の抗BSA-NP抗体レベルは、上述のとおり、免疫の7日後にELISAにより測定した。
製造元のプロトールに従ってB細胞単離キット(Miltenyi Biotec)を用いたMACSによるネガティブセレクションによって、脾臓B細胞を精製した。MACS選別したB細胞の純度は、B220染色に関して>95%陽性であった。B細胞増殖アッセイのために、B細胞を5μM 5-(および6-)カルボキシフルオレセイン二酢酸サクシニミジルエステル(CFSE; Dojindo)で10分間、37℃で標識した後、rTGF-beta1、rTGF-beta2、またはrTGF-beta3の存在下または非存在下、10μg/ml 抗IgM F(ab)'2(Jackson ImmunoResearch Laboratories)で72時間刺激した。細胞を、抗B220 mAb、7-アミノ-アクチノマイシンD(7-AAD)(Biolegend)およびPE抗CD40 mAbで染色した。7-AAD陰性のCFSE希釈B220+CD40+ 生存B細胞および7-AAD陽性のB220+ 死細胞のパーセンテージを、FACSにより評価した。
96ウェル平底プレートのウェルを、100μl/ウェルのPBS中の10μg/ml 抗CD3 mAbでコーティングし、4℃で一晩インキュベートした。ウェルを洗浄した後、コーティングされたウェルに、MACSで精製したB細胞およびFACSで精製した各T細胞サブセット(LAG3+ Treg、CD25+ Treg、またはCD4+CD25-CD44loCD62Lhi ナイーブT細胞)または後述のIL-27処理CD4+ T細胞を1×105個/ウェルの密度で加え、rTGF-beta3(1 ng/ml)添加または非添加の10μg/ml 抗CD40 mAb(3/23)+10μg/ml rIL-4(Cell Signaling Technology)含有RPMI培地中で培養した。3日目にアポトーシスを受けているB細胞を、また7日目の培養上清における総IgG産生量を、製造元のプロトールに従ってAnnexin V Apoptosis Detection Kit(BD Pharmingen)およびマウスIgG ELISA Quantitation Set(Bethyl Laboratories)をそれぞれ用いて測定した。
MACSで精製したB6マウスまたはPD-1KOマウス由来のB細胞2×105個、および、FACSで精製したOT-IIマウスまたはPD-1KO OT-IIマウス由来のTh細胞2×105個を、FACSで精製したB6、Egr2CKO、B6/lpr、B6/gld、もしくはIL-10KOマウス由来のLAG3+ Treg 1×105個と組み合わせてまたは組み合わせずに、Rag1KOマウスに静脈内注射した。対照マウスにはPBSを投与した。続いて細胞移入の24時間後、マウスを100μg NP-OVA/alumで免疫した。初回免疫の14日後、マウスを50μg NP-OVA/alumで再度免疫した。示されている場合には細胞移入の翌日に、抗FasLブロッキング抗体(200μg/マウス)もしくは抗TGF-beta3ブロッキング抗体(100μg/マウス)を週1回の間隔で、または抗PD-L1ブロッキング抗体(200μg/マウス)を3日毎に、マウスに静脈内注射した。再免疫の7日後に、血清中抗NP抗体レベルをELISAで解析し、脾細胞をFACSにより解析した。
抗NP IgG抗体レベルを、インビトロまたはインビボ抗体産生アッセイにおける捕捉抗原としてNP(9)-BSA(Biosearch Technologies)を用いたELISAによって、それぞれ定量した。ELISAプレートは、製造元のプロトールに従ってImmuno-Tek ELISA construction system(Zepto Metrix)を用いて、調製した。試料血清または培地とのインキュベーションの後、HRPコンジュゲートヤギ抗マウスIgG1およびTMB基質を用いてプレートを発色させた。NP(13)-OVA免疫B6マウス由来の連続希釈したプール血清を、各プレートにおける対照として含めた。抗NP IgG1抗体の濃度は、プール血清から構築された検量線との比較によって推定した。
FACSで精製したOT-IIマウス由来のTh細胞2×105個を、FACSで精製したB6マウス由来のLAG3+ Treg 1×105個と組み合わせてまたは組み合わせずに、TEaマウスに静脈内注射した。対照マウスにはPBSを投与した。続いて細胞移入の24時間後、マウスを100μg NP-OVA/alumで免疫した。血清中の抗BSA-NP抗体レベルは、上述のとおり、免疫の14日後にELISAにより解析した。
8週齢のMRL/lpr マウスを、個別の処理群に無作為に割り当てた。処理群における10週齢のMRL/lprマウスに、MRL/+マウスから得たLAG3+ Treg、LAG3- T細胞、CD25+ Treg、またはナイーブT細胞(それぞれ細胞1×105個)を、静脈内注射した。3回注射群(MRL/+ LAG3+ Treg×3、MRL/+ CD25+ Treg×3、およびMRL/lpr LAG3+ Treg×3)には、1×105個のLAG3+ Tregを週1回の間隔で静脈内養子移入した(それぞれ10、11、および12、または、13、14、および15週齢;マウスは、初回注射の時点で10週齢であった)。各T細胞サブセットをまずMACSによって濃縮し、次に、上述のようなFACS(CD4、CD25、CD45RB、およびLAG3の発現に基づく)によって選別した。対照群のマウスにはPBSを投与した。示されている場合には最初の細胞移入の翌日に、抗TGF-beta3ブロッキング抗体(100μg/マウス)を週1回の間隔でマウスに静脈内注射した。マウスを18週齢で屠殺し、病理学的な変化を調査した。抗dsDNA抗体は、製造元のプロトールに従ってマウス抗ds DNA ELISA Kit(Shibayagi)を用い、13および18週齢で測定した。
尿中タンパクはディップスティック(Albustix, Bayer)を用いて週1回の間隔で半定量的に評価した(0=なし;1=30〜100 mg/dl;2=100〜300 mg/dl;3=300〜1,000 mg/dl;4 ≧ 1,000 mg/dl)。
MRL/lprマウスを18週齢で屠殺した。腎臓病理を、非特許文献49に記載のように、糸球体の炎症、増殖、半月体形成、および壊死について標準的な方法でグレード付けした。糸球体評価は、実験条件を知らされていない評価者が切片ごとに少なくとも80の糸球体を検査することによって実施した。間質および尿細管の変化にも留意した。これらの特徴の各々に、0から4までのスコア(0は「損傷なし」を表し、4は「重症」を表す)を割り当てた。
B6およびOT-IIマウスを、100μg NP-OVA/alumで免疫した。最初の免疫の3週間後にマウスを100μg NP-OVA/alumで再度免疫した。免疫後のB6マウス由来のMACSで精製した2×105個のB細胞および免疫後のOT-II マウス由来のFACSで精製した1×105個のCD4+CD25-LAG3-ヘルパーT細胞(Th細胞)を、免疫していないOT-IIマウス由来のFACSで精製した1×105個のLAG3+ Tregと組み合わせてまたは組み合わせずに、10μg/ml抗PD-L1ブロッキングmAb(10F.9G2)または10μg/ml抗FasLブロッキングmAb(MFL3)の存在下または非存在下で、再免疫の7日後に96ウェル丸底プレートに播種した。培養上清を3週間目に回収し、上述のように抗NP抗体レベルをELISAで分析した。
T細胞の総RNAをRNeasy Micro Kit(Qiagen)を用いて調製した。非特許文献14に記載のように、RNAをcDNAに逆転写して、定量的リアルタイムPCR分析を行った。対照のマウスbetaアクチンまたはヒトGAPDHの存在量を基準として、相対的なRNA存在量を測定した。
B6マウス由来のCD4+CD25+、CD4+CD25-CD45RBlowLAG3+、およびCD4+CD25-CD45RBhighLAG3- FACS精製T細胞の総RNAを回収し、次いで非特許文献14に記載の方法でAffymetrixマイクロアレイ分析のための試料を調製した。マイクロアレイデータはBioconductor(バージョン19)および統計処理ソフトウェアRおよびGeneSpring GXバージョン7.3.1(Silicon Genetics)を用いて分析した。すべてのマイクロアレイデータは、ArrayExpressデータベース(http://www.ebi.ac.uk/arrayexpress)にアクセッション番号E-MEXP-1343として保存されており、これは本発明者らの研究室での以前の研究(非特許文献14)と同一である。
各T細胞サブセットを、抗CD3 mAbまたは抗CD3 mb/抗CD28 mAbをコーティングしたウェルに、それぞれTGF-beta1および2の測定用に無血清X-Vivo-20培地(Lonza)に、またはTGF-beta3用に上記のRPMI培地に、3×105細胞/ウェルで撒いた。37℃で72時間インキュベートした後、上清を回収して、-80℃で保存した。上清中のTGF-beta1、2、および3レベルは、それぞれTGF-beta1 Emax ImmunoAssay System(Promega)、TGF-beta2 Quantikine ELISA Kit(R&D Systems)、およびTGF-beta3 ELISA Kit(Mybiosource)によって、製造業者のプロトコールに従い測定した。10%FBSを添加したRPMI培地におけるTGF-beta3レベルは、TGF-beta3 ELISA Kitの検出限界以下であった。
MACSで精製したB6マウス由来のB細胞を0.75μM CpG-ODN(ENZO Life Science)で72時間、最後の16時間はrTGF-beta3(20 ng/ml)を添加または非添加で前処理し、次にRPMI培地中、10μg/mlの抗CD40 mAb、10μg/mlのrIL-4、または10μg/mlの抗IgM F(ab)'2により刺激した。刺激の後、細胞をLysis Buffer(50mM Tris-HCl、0.15 M NaCl、1%Triton X-100、1 mM EDTA)で処理し、2×Laemmli Buffer(BioRad)で95℃、5分間変性させた後、Mini-PROTEAN TGXプレキャストゲル(BioRad)で電気泳動により分離した。細胞溶解物の総タンパク質濃度は、BCA Protein Assayキット(Pierce)を用いて決定した。電気泳動後のゲルはポリフッ化ビニリデン(PVDF)メンブレン上にブロットし、5%BSAでブロッキングした後、リン酸化または総STAT6、NF-κB p65、またはSyk(いずれもCell Signaling Technologyから購入)に対する抗体を反応させ、さらに抗ウサギIgG-HRP(Invitrogen)に対する二次抗体でプローブした。メンブレンはECL Prime基質(GE Healthcare)で発光させた。
上記のMACSで選別したCD4+ T細胞を、FACSによりCD4+CD25-CD62L+CD44-ナイーブT細胞として精製し、2μg/ml抗CD3 mAbおよび2μg/ml抗CD28 mAbをコーティングした96ウェルプレートに、上記のRPMI培地100μl中3×105の密度で播種した。各エフェクター細胞分化のためのサイトカインは以下の通りである:Th0、抗IFN-γ(10μg/ml; XMG1.2)、および抗IL-4(10μg/ml; 11B11);Th0、抗IFN-γ(10μg/ml;XMG1.2(BD Pharmingen))、および抗IL-4(10μg/ml;11B11(BD Pharmingen));Th1、IL-12(10 ng/ml(R&D Systems))、IL-2(50μg/ml(R&D Systems))、および抗IL-4(10μg/ml;11B11);Th17、TGF-beta1(1ng/ml)、IL-6(50 ng/ml(BioLegend))、IL-23(50 ng/ml(R&D Systems))、抗IFN-γ(10μg/ml;XMG1.2)、および抗IL-4(10μg/ml;11B11);誘導性Treg、TGF-beta1(5ng/ml)および抗IL-4(10μg/ml;11B11)。培養上清を5日目に回収し、TGF-beta3レベルを上記のようにELISAで測定した。
CD4+CD62L+ T Cell Isolation Kit(Miltenyi Biotec)を用いて製造業者のプロトコールに従い、MACSで精製したナイーブCD4+ T細胞を、25ng/ml IL-27を加えた上記のRPMI培養培地中、2μg/ml抗CD3 mAbおよび1μg/ml抗CD28 mAbでコ−ティングした24または96ウェルプレートにおいて、インビトロで5日間刺激した。IL-27で処理したナイーブT細胞を次いで、CD4発現についてフローサイトメトリーで選別し、各アッセイに用いた。ELISAによる培養上清中のTGF-beta3レベルの決定のため、IL-27処理したT細胞を、PMA(50 ng/ml; Sigma)およびイオノマイシン(1μg/ml; Sigma)で最後の4時間刺激した。
マウスTGF-beta3の全長断片を、Tgfb3 cDNA(NM_009368)を含むOmicsLink(商標)Expression-Ready ORFクローニングベクター(GeneCopoeia)から単離した。Tgfb3 cDNAをEcoRIサイトを用いて、CAG(サイトメガロウイルス即時型初期エンハンサー/ニワトリbetaアクチンハイブリッド)プロモーターを有するpCAGGSベクター(非特許文献50)にサブクローニングし、pCAGGS-Tgfb3とした。組み換えプラスミドを次いで、大腸菌(Escherichia coli)JM109のコンピテント細胞内に形質転換し、EndFree Plasmid Giga Kit(Qiagen)を用いて製造業者のプロトコールに従い、プラスミド精製カラムにて精製した。精製したプラスミドDNAは、使用の直前に、滅菌したPBS(pH 7.4)で1μg/μlに希釈した。
MRL/lprマウスに、滅菌したPBS(pH 7.4)に溶かした100μgのプラスミドDNA(pCAGGs-Tgfb3または対照pCAGGS)を、4週間の間隔を置いて二回、静脈内注射した。週1回の間隔でタンパク尿を半定量的に測定し、最後の投与の6週間後に腎臓病理を上記のように評価した。
ヒトの試料はすべて、インフォームドコンセントの元で入手した。ヒトリサーチプロジェクトのためのプロトコールは、東京大学の倫理委員会によって承認されている。健康なヒトおよびSLE患者由来のPBMCを、Ficoll-Paque(Amersham Pharmacia Biotech)グラジエントによって単離した。細胞を洗浄した後、それらを示したmAbで、4℃にて20分間染色した。mAbの非特異的結合を防ぐために、標識したmAbで染色する前にHuman Fc Receptor Binding Inhibitor(eBioscience)を添加した。死細胞は、7-AADによって除いた。蛍光陽性細胞をMoflo XDPセルソーターで分析した。5つの異なる亜集団は以下の通りである:(ナイーブT)CD4+CD25-CD45RA+CCR7+細胞;(CD25+ Treg)CD4+CD25+CD127dimCD45RA-細胞;(LAG3+ Treg)CD4+CD25-CD45RA-LAG3+細胞;(Tfh)CD3+CD19-CD4+CD25-LAG3-CXCR5+CD45RA-細胞;(B細胞)CD3-CD19+細胞。
プレートに結合した5μg/mlの抗CD3 mAbで72時間刺激したヒトナイーブTおよびCD25+ TregおよびLAG3+ Tregから単離した総RNAを、上記のようにEGR2、IL10、IFNG、およびFOXP3のmRNA発現について分析した。TGFB3 mRNA発現は、各T細胞サブセット由来の刺激していない細胞を用いて決定した。
サイトカイン分析のために、ヒトナイーブT、CD25+ Treg、およびLAG3+ Tregを、5μg/mlの抗CD3 mAb /抗CD28 mAbでコーティングした96ウェル平底プレートに、10% FBS、100μg/ml L-グルタミン、100 U/mlペニシリン、100μg/mlストレプトマイシン、および50μM 2-メルカプトエタノールを添加したRPMI-1640培地中、2×104細胞/ウェルで撒いた。培養上清を3日目に回収し、製造業者のプロトコールに従いOptEIA Human IL-10 ELISA Kit II(BD Biosciences)を用いたELISAにより、培養上清中IL-10レベルを測定した。
上記のRPMI-1640培地を共培養に用いた。FACSで精製した1×105個のヒトB細胞およびFACSで精製した5×104個のヒトTFH細胞を、FACSで精製した1×105個のヒトCD25+ TregまたはLAG3+ Tregと共にまたは伴わずに、2μg/ml組み換えブドウ球菌エンテロトキシンB(Toxin Technology)の存在下で、丸底96ウェルプレートに播種した。培養上清を12日目に回収し、総IgGレベルを、製造業者のプロトコールに従いHuman IgG Quantitation Setキット(Bethyl Laboratories)を用いたELISAにより分析した。
統計的有意性、正規分布、および群間のF検定を、GraphPad Prismバージョン5.03(GraphPad Software, Inc.)を用いて分析した。定量的組織学およびタンパク尿の進行を、Mann-WhitneyのU検定により分析した。他のすべての統計的差異は、Studentの両側t検定を用いて決定した。分散が同等でない場合、Student のt検定にWelchの補正を適用した。すべての試験について、P<0.05を統計的に有意な差とみなした。図のすべてのデータは、平均±s.dで表される。試料のサイズは、以前の研究で典型的に用いた数に基づいて推定した。試料サイズをあらかじめ決めるために統計的方法は使用しなかった。分析から除外した試料や動物はなかった。MRL/lprマウスにおける養子細胞移入試験を除いて、動物の無作為化は行わなかった。組織学的分析以外、動物実験は盲検方式で実施しなかった。

T細胞におけるEgr2の役割を明らかにするために、本発明者らはT細胞特異的なEgr2コンディショナルノックアウト(CKO)マウス(Egr2fl/fl CD4-Cre+)を作出した。Egr2 CKOマウスは、CD4+CD25-CXCR5+PD-1+ TFHおよびB220+GL-7+Fas+ 濾胞B細胞 (GCB)の比率の顕著な増加を示した(図1a)。また、Egr2 CKOマウスでは、4-ヒドロキシ-3-ニトロフェニルアセチル(NP)-オボアルブミン(NP-OVA)の単回免疫後のNP特異的抗体の産生が増強されていた(図1bおよび図8a)。野生型(WT)LAG3+ Tregを移入すると、TFHおよびGCBの自発的分化が顕著に抑制され(図1a)、過剰な抗体産生が阻害された(図1b)ことから、LAG3+ TregはインビボでB細胞応答を抑制できることが示唆された。インビトロでのT細胞/B細胞共培養系において、抗CD3抗体で刺激したWT LAG3+ Tregは、抗IgM抗体で刺激したB細胞の生存率、および抗CD40抗体/IL-4で刺激したB細胞による総IgG産生量を、CD25+ Tregよりも効果的に低減した(図1c、d)。インビボでのB細胞応答を評価するために、WTマウス由来のB細胞、およびOVA特異的OT-II T細胞受容体(TCR)トランスジェニックマウス由来のCD4+CD25-LAG3-ヘルパーT細胞(Th細胞)を、組み換え活性化遺伝子1欠損(Rag1KO)マウスに移入し、その後、NP-OVAを2回免疫した。WT LAG3+ Tregの共移入は、NP特異的抗体応答ならびにTFHおよびGCBの発生を著しく効果的に抑制した(図1e,fおよび図8b)。Egr2CKOマウスにおいて増強されたGCBの発生および抗体産生は、B細胞制御におけるEgr2の極めて重要な役割を示唆しており、Egr2欠損LAG3+ Treg(図8c)は、インビボでのB細胞による抗体産生ならびにTFHおよびGCBの発生を抑制することができなかった(図1e、f)。このように、LAG3+ TregによるB細胞応答の抑制には、LAG3+ TregにおけるEgr2の発現が必要であることが明らかとなった。Egr2プロモーターの制御下で緑色蛍光タンパク質(GFP)を発現したトランスジェニックマウス(Egr2-GFPマウス、図9a)において、CD4+ T細胞におけるGFPの発現は、Egr2タンパク質の発現と相関していた(図9b)。Egr2の重要性は、Egr2-GFPマウス由来のCD4+CD25-Egr2-GFP+細胞も、LAG3+ Tregと同様にインビボでB細胞抑制活性を示したという知見によって確認された(図9c)。本発明者らは次に、LAG3+ Tregを介した抗体産生の抑制が、Rag1KOマウスのようなリンパ球減少状態の下のみならず、より生理学的な非リンパ球減少状態の下でも誘導されるかどうかを検証した。Eαペプチド特異的TCRトランスジェニックTEaマウスに、WT B細胞およびOT-II Th細胞を養子移入し、その後、NP-OVAを1回免疫した。共移入WT LAG3+ Tregは、非リンパ球減少TEaマウスにおいてNP特異的な抗体産生を効果的に抑制した(図1gおよび図8d)。
本発明者らは、ループス様病態を呈する、Fas変異を有するMRL-Faslpr/lpr(MRL/lpr)マウス(非特許文献17)における病態の進行を、LAG3+ Tregが阻害できるかどうかを調べた。MRL/lprマウスに、Fas変異を有さないMRL-Fas+/+(MRL/+)マウスに由来する様々なT細胞サブセットを養子移入したところ、LAG3+ Tregはタンパク尿の進行を顕著に遅延させたが、CD25+ Tregは遅延させなかった(図2a)。さらに、LAG3+ Tregを3回移入することにより、タンパク尿の進行がほぼ完全に抑制された。抗ds-DNA抗体力価および糸球体病変スコアの増加も、LAG3+ Tregの単回移入によって阻害された(図2b〜dおよび図10a)。対照的に、既報(非特許文献18)と一致して、MRL/+マウス由来のCD25+ TregをMRL/lprマウスに3回移入しても疾患の進行は変化しなかった(図10b〜d)。さらに、MRL/lprマウス由来のLAG3+ TregをMRL/lprマウスに養子細胞移入した場合も治療効果を及ぼさなかった(図10b〜d)。また、顕性タンパク尿発症後のMRL/lprマウスにMRL/+ LAG3+ Tregを3回注射すると、当該マウスのループス様病態が改善された(図10e、f)。
次に、本発明者らは、LAG3+ TregによるB細胞抑制がIL-10またはTGF-betaファミリーメンバーによって媒介されるかどうかを調べた。上記のように、LAG3はTr1細胞の特異的な細胞表面マーカーの1つであると考えられている(非特許文献16)。本発明者らは、LAG3+ Tregが大量のIL-10を産生し(非特許文献14)、Egr2がB lymphocyte-induced maturation protein-1(Blimp-1)(Prdm1遺伝子によってコードされる)を通じてCD4+ T細胞におけるIL-27誘導性のIL-10産生を媒介すること(非特許文献19)をこれまでに報告した。予想通り、T細胞特異的なPrdm1CKOマウス(Prdm1fl/fl CD4-Cre+)由来のLAG3+ TregにおけるIL-10発現レベルは、WTマウス由来の場合と比べて顕著に低減された(図12a)。Prdm1CKOおよびIL-10欠損(IL-10KO)マウスに由来するLAG3+ Tregも、インビボでのNP特異的抗体応答を効果的に抑制したことから(図12b)、IL-10がLAG3+ TregによるB細胞抑制にとって重要ではない可能性があることが示唆された。LAG3+ Tregのマイクロアレイ分析(非特許文献14)および定量的リアルタイムPCRの結果から、TGF-beta1または2ではなく、TGF-beta3の発現の顕著な増加が明らかにされた(図3a、b)。T細胞受容体(TCR)刺激は、LAG3+ Tregの培養上清において、TGF-beta1または2ではなく、TGF-beta3の大量の産生を誘導した(図3c〜e)。対照的に、CD25+ Tregは、これらの条件下で少量のTGF-beta1を産生しただけであった。TGF-beta3は、抗IgM抗体で刺激したB細胞の増殖およびCD40発現を顕著に抑制し(図3f)、B細胞死を強力に誘導し(図3g)、総IgG産生を抑制した(図3h)。TGF-beta3は、TGF-beta1がB細胞の機能を強力に抑制したという以前の所見(非特許文献20)に合致して、TGF-beta1および2と類似の効果を生み出した(図13a、b)。シグナル伝達に関して、TGF-beta3の添加は、活性化B細胞においてsignal transducer and activator of transcription(STAT)6、Syk、およびNF-κB p65のリン酸化を顕著に低減させた(図3i〜k)。ヘルパーT細胞によって産生されたIL-4はB細胞の増殖および生存を増強し、同時にSTAT6の活性化を介して免疫グロブリンの分泌およびアイソタイプ・スイッチングを促進する(非特許文献21)。チロシンキナーゼSykの活性化は、B細胞受容体(BCR)刺激に応じた細胞内シグナル伝達にとって重要である(非特許文献22)。抗原で刺激されたB細胞による特異的な抗体産生に必要なCD40活性化は、NF-κB p65のリン酸化を誘導する(非特許文献23)。それゆえ、TGF-beta3はB細胞の機能にとって重要な複数の経路を阻害する。TGF-beta3は、IL-23依存的に分化しているTh17細胞によっても産生される(非特許文献24)ため、TGF-beta3の産生はLAG3+ Tregによるものに限定されない。本発明者らは、Th17細胞に加えてTh1細胞がTGF-beta3を産生することを見出した(図3l)。しかしながら、LAG3+ TregはTh1細胞およびTh17細胞よりも顕著に多量のTGF-beta3を産生した。
Programmed death-1(PD-1)遺伝子の変異体がSLEの疾患感受性と関連付けられている(非特許文献1)。PD-1は、T細胞およびB細胞の両方に負の共刺激シグナルを付与し(非特許文献25、26)、PD-1欠損(PD-1KO)マウスはループス様の疾患を発症する(非特許文献27)。さらに、PD-1およびLAG3は自己免疫および腫瘍免疫を相乗的に制御する(非特許文献28、29)。TGF-beta3とPD-1との間の潜在的な協調について調べるため、本発明者らは、B6マウス、PD-1KOマウス、Fasを欠損したB6/lprマウス、およびFasLを欠損したB6/gldマウスに由来する、抗IgM抗体で刺激したB細胞にrTGF-beta3を加えた。PD-1KOマウス由来のB細胞はTGF-beta3誘導性の細胞分裂阻害に耐性であったが、B6/lprマウスまたはB6/gldマウスに由来するB細胞は耐性を示さなかった(図5a)。リン酸化されたSTAT6がB細胞株において抗アポトーシスBcl-xLの発現を誘導することが知られているという既報(非特許文献30)に合致して、TGF-beta3は、WTマウス由来の活性化細胞においてBcl-xLおよびBcl-2a1の発現を抑制したが、PD-1KO由来のB細胞においては抑制しなかった(図5b)。TGF-beta3およびPD-1による協調的なB細胞抑制は、LAG3+ Tregを介した抑制における、B細胞上でのPD-1発現の重要性を強く支持している。抗PD-L1ブロッキング抗体の添加によって、インビトロでのLAG3+ Tregを介した抗体産生抑制が阻害された(図5c)。抗NP抗体の産生は、PD-1KO B細胞およびWT OT-II Th細胞を移入しOVA−NPで2回免疫を行ったRag1KOマウスでのWT LAG3+ Tregの共移入によって抑制されなかった。しかしながら、WT B細胞およびPD-1KO OT-II Th細胞を移入したRag1KOマウスは、LAG3+ Tregにより抑制された(図5d)。これらの結果は、LAG3+ TregによるB細胞抑制のためにB細胞上でのPD-1の発現が必要とされることを支持するものである。CD4+CD25-Egr2 + T細胞は、PD-1のリガンドPD-L1、およびLAG3の両方を共発現した(図14a、b)。しかしながら、PD-L1およびPD-L2は、GCBを含むさまざまな細胞種に発現している(非特許文献31)ので、LAG3+ Treg上のPD-L1は、B細胞上のPD-1に対する唯一のリガンドではない可能性もある。
IL-27は、抗原提示細胞(APC)によって産生されるサイトカインのIL-12/IL-23ヘテロ二量体ファミリーの一員である。IL-27は、IL-10産生Tr1細胞の分化因子として特定された(非特許文献32)。本発明者らは、IL-27がCD4+ T細胞におけるEgr2の発現を誘導し、Egr2がBlimp-1媒介性のIL-10の産生に必要とされることを以前に報告した(非特許文献19)。IL-27処理は、Egr2および、LAG3だけでなく(図6a)、TGF-beta3 mRNAおよび、TGF-beta3タンパク質産生も誘導した(図6b、c)。Egr2欠損CD4+ T細胞は、IL-27誘導性のTGF-beta3産生の大幅な低下を示し(図6c)、これにより、TGF-beta3の誘導に対するEgr2の重要性が明らかになった。CD4+ T細胞における特異的STATタンパク質活性化は、ヘルパーT細胞系統の分化に関連している。IL-27を介したIL-10の誘導にはSTAT1およびSTAT3の両方が必要になる(非特許文献32)が、本発明者らは、IL-27を介したEgr2誘導がSTAT3に依存することを以前に見出した(非特許文献19)。IL-27媒介性のTGF-beta3誘導は、STAT1欠損CD4+ T細胞ではなく、STAT3欠損CD4+ T細胞で損なわれ(図6c)、これにより、IL-27によるEgr2とTGF-beta3誘導がSTAT3依存的であることが実証された。さらに、IL-27で処理されたCD4+ T細胞は、B細胞抗体産生を顕著に抑制した(図6d)。対照的に、IL-27で処理されたEgr2欠損CD4+ T細胞は、B細胞による抗体産生を抑制することができなかった。このように、IL-27誘導性のTGF-beta3産生細胞は、Egr2依存的に液性免疫に対する抑制活性を示す。
本発明者らは、健常ドナーの末梢血単核細胞(PBMC)に由来するCD4+ T細胞においてCD4+CD25-CD45RA-LAG3+ T細胞を同定した(図7a)。マウスLAG3+ Tregと同様に、ヒトCD4+CD25-CD45RA-LAG3+ T細胞は、TCR刺激に応じてEGR2、IL10、およびIFNGを発現し(図7b)、大量のIL-10を産生した(図7c)。ヒトCD4+CD25-CD45RA-LAG3+ T細胞の制御活性は、それらがB細胞およびTFH細胞と共培養された場合に、CD4+CD25+CD127lowCD45RA-活性化Treg(非特許文献33)と比べて効果的に抗体産生を抑制したという測定結果によって確認された(図7d)。ヒトCD4+CD25-CD45RA-LAG3+ T細胞は培養後に高レベルのTGF-beta3を発現し(図7e)、それらがマウスLAG3+ Tregと同一の機構を通じてB細胞を抑制することを示唆していた。それゆえ、本発明者らは、CD4+CD25-CD45RA-LAG3+ T細胞集団を、マウスLAG3+ Tregに対するヒトの対応物と考える。本発明者らは次に、LAG3+ Tregがヒトの全身性自己免疫疾患において減少されうるかどうかを評価した。血中のLAG3+ Tregの割合は、健常ドナーと比べてSLE患者の末梢血において顕著に低かった(図7f)。これらの所見から、SLE患者において抗体抑制能を有するT細胞集団をモニターするためにLAG3発現が使用されうることが示唆される。
TGF-beta3とTGF-beta1は一般的には共通のシグナル伝達分子を介して、少なくともin vitroでは同様の生物活性を示すと理解されている。しかし、in vivoでは主にノックアウトマウスの成績から、表現が異なることが報告されており、この違いは発現の時期や場所によって生じている、或いは、本来致死であるTGF-beta1 KOマウスにTGF-beta3をノックインしたマウスでは、rescueされるものの異なる表現も見られることから、生体においては異なる生物活性を示す可能性も示唆されている(非特許文献51)。
また免疫系の報告では、実験的自己免疫性脳脊髄炎(EAE)モデルで、TGF-beta3が病原性のT細胞(Th17細胞)を誘導して、病態を悪化させるという、今回の実験結果とは逆の報告もある(非特許文献52)。同文献では、TGF-beta1にはこの作用が無いことが示されいる。同様に自己免疫疾患の悪化にTGF-beta3が関与していることがTRIM28 KOマウスでも報告されている(特許文献52)。このマウスは自然発症の自己免疫疾患で短命であり、EAEも重症化するが、これにTGF-beta3のderepressionが関与しているとされている。
その結果、TGF-beta3発現プラスミドは脾臓重量(21週齢)、経時的なタンパク尿の増加をコントロールプラスミドに比べて有意に抑制し、その効果はTGF-beta1発現プラスミドより強い傾向を示した(図15a, b)。また、TGF-beta3発現プラスミドは腎臓へのIgGの沈着を抑制し、加えて細胞の浸潤と増殖・半月体形成・細胞死等を指標した糸球体傷害もコントロールプラスミド、TGF-beta1発現プラスミドに比べて有意に軽減していることが明らかになった(図16a, b)。さらにTGF-beta1発現プラスミドはリンパ節、脾臓など二次リンパ組織胚中心に存在する、CD4/CXCR5/PD-1陽性CD25陰性の濾胞性ヘルパーT細胞(TFH)の割合を有意に増加させたのに対し、TGF-beta3発現プラスミドはこれを有意に抑制した(図17a, b)。
以上の結果から、MRL/lprマウスモデルにおけるTGF-beta1とTGF-beta3の治療効果の違いは、抗体産生を伴う免疫反応に重要な濾胞性ヘルパーT細胞の抑制能の有無に依存している可能性が示唆された。すなわち、TGF-beta3は濾胞性ヘルパーT細胞の抑制というTGF-beta1にはない作用により、SLEなどの自己免疫疾患治療に有用である可能性がある。
本研究の結果から、LAG3+ Tregが、ループス様病態を呈するMRL/lprマウスにおけるGCBおよびTFHの発生、抗体産生、ならびに疾患の進行を抑制することが実証された。Egr2依存的かつFas依存的にLAG3+ Tregによって産生されるTGF-beta3は、液性免疫の抑制において決定的な役割を果たす。LAG3+ TregはCD25+ Tregよりもはるかに高いレベルのIL-10を産生する(非特許文献14)ので、LAG3+ Tregは制御性サイトカインの強力な産生体でもある。TGF-beta3の炎症促進性の役割は、TGF-beta3が病原性Th17細胞を効果的に誘導するという知見によって以前に実証された(非特許文献24、34)。本発明者らの結果は、自己免疫の制御におけるTGF-beta3の、これまで認識されていなかった役割を明らかにするものであった。TGF-beta1も炎症促進性および抗炎症性の両方の効果を及ぼす(非特許文献35〜37)。特に、TGF-beta1はB細胞アポトーシスを誘導し、活性化したヒト扁桃腺B細胞からの免疫グロブリン産生を低減する(非特許文献38、39)。CD25+ TregおよびTh3制御性細胞(非特許文献40、41)は強力なTGF-beta1産生源であり、CD25+ TregはTGF-beta1を通じてB細胞の免疫グロブリン合成を抑制することが示されている(非特許文献42)。しかしながら、CD25+ Tregを含むCD4+ T細胞により産生されるTGF-beta1の量は比較的限られており(図3e)、免疫抑制に関連するTGF-beta1産生源を明確にすることは困難であった。いくつかの系において、TGF-beta1およびTGF-beta3が明らかなアイソフォーム特異的生物活性を示すことが実証されているが、TGF-beta1およびTGF-beta3はB細胞応答については同等な抑制活性を示した(図13a、b)。ヘルパーT細胞の発生に関して、TGF-beta3は、病原性Th17細胞の発生中にTh17細胞によって自発的に産生される(非特許文献24)。本発明者らの実施においては、Th17細胞だけでなく、Th1細胞も、かなりの量のTGF-beta3を産生したが、LAG3+ TregはTh1およびTh17細胞よりもさらに多くの量のTGF-beta3を産生した(図3l)。よって、LAG3+ Tregにより産生される大量のTGF-beta3は、免疫寛容の発生および維持において重要な役割を果たす。
本発明者らは、TGF-beta3によるB細胞活性抑制の知見に基づき、TGF-beta3を用いた新たな機能分子の創製を検討した。機能分子は、例えば TGF-beta3を抗体のFc領域とアミノ酸で連結し、さらに該抗体はB細胞を認識する可変領域を含む構成が考えられる。抗体又は抗体断片との連結はTBP(Latent-TGFbeta-binding protein)-1,3および4の第3番目のTBdomain(TGFbeta binding domain)を用いた連結、酵素を用いた連結、化学的な修飾反応を用いた連結等で実施することも出来る。
その他の手法としては、一の可変領域がTGF-beta3を認識し、他の可変領域がB細胞を認識する多重特異性抗体の構成をとることもできる。また、TGF-beta3と抗体のFc断片とが連結し、さらに該TGF-beta3にB細胞を認識する可変領域断片が連結した融合タンパク質の構成をとることもできる。
B細胞は望ましくは自己反応性B細胞であり、また可変領域によるB細胞の認識は、例えばCD19、CD20、CD40、CD22、IL21R、BAFF-R、BCMA、TACI、CD27、CD138等のB細胞表面マーカーとすることができる。
遺伝子構築
マウスTGFbeta3遺伝子は、データベースに登録されているアミノ酸配列(Uniprot No. P17125 )の24番目から410番目を使用し、Latent TGFbeta3として発現するために、N末端側にマウスIg kappa鎖のシグナル配列を付加し、直後に精製を簡便に行う目的でFLAGタグを融合させた。また、Latent TGFbeta3については、発現量を改善する目的で公開論文(Molecular Therapy (2010) 18:12, 2104-2111)の情報に従って、データベースに登録されているアミノ酸配列(Uniprot No. P17125 )の25番目のCysをSerに置換した。野生型のTGFbeta3遺伝子はOmicsLink(商標)Expression-Ready ORFクローニングベクター(GeneCopoeia)に挿入されている遺伝子配列を使用した。シグナル配列を除いたLatent TGFbeta3のアミノ酸配列を配列番号11に記載した。
TGFbeta3改変抗体分子は、マウスCD19を認識する抗体を片腕に有する分子形であり、C末端にTGFbeta3を連結するためのTB3ドメイン(TBP(Latent-TGFbeta-binding protein)-1,3および4の第3番目のTBdomain(TGFbeta binding domain))を連結して調製した。また、比較対象としてTGFbeta3を連結するTB3ドメインを持たない抗体も調製した。これらの改変抗体分子は、抗CD19抗体の1個のFabがFcに連結した構造となる。
マウスCD19を認識する抗体1D3の抗体可変領域遺伝子は、ATCCで販売されているラットハイブリドーマ1D3 (ATCC(登録商標)HB-305TM)から定法に従ってクローニングした。クローニングしたL鎖可変領域は、マウスkappa鎖定常領域と連結した。クローニングしたH鎖可変領域は以下に示す改変定常領域配列と連結した。H鎖定常領域としては、まず、Fc受容体への結合を欠失するようにアミノ酸変異を導入したFc領域mF18(EUナンバリングで表される235位のProがLysに置換されたアミノ酸変異および239位のSerがLysに置換されたアミノ酸変異を有する)に対し、Fc領域のヘテロ化を促進するためにCH3ドメインD399Rを導入した。さらに、C末端側に検出および精製に使用するV5タグおよびlatent TGFbeta3をコンジュゲートするためにマウスLTBP1タンパク質の3番目のTBドメイン(TGFbeta binding domain)の配列(TB3)を連結した(配列番号:9)。Fc領域のみを有するH鎖定常領域にも、上述のFc受容体非結合のための変異と、ヘテロ化促進変異としてK409Eを導入した(配列番号:10)。Fc受容体非結合変異の詳細は公開特許WO 2012-132067に、ヘテロ化促進変異の詳細は公開特許WO2015-046467に記載されている。
これらの遺伝子は哺乳類細胞一過性発現用ベクターpBEF-OriP(Wako)にクローニングした。得られたプラスミドはプラスミド調製キット(Macherey-Nagel, NucleoBond Xtra Maxi)でトランスフェクショングレードの試料を調製し発現実験に使用した。発現したタンパク質の遺伝子名、タンパク質名の一覧を表1に示した。
これらのタンパク質の発現にはExpi293-F細胞(Life technologies, A14527)とそのトランスフェクションキット(ExpiFectamine293 transfection kit)を使用した。Expi293-F細胞の培養はExpi293 medium(Life technologies)を培地に用い、37℃のCO2インキュベーター内で135 rpmで振盪して行った。プラスミドの導入はLife technologies社のプロトコールで実施したが、TGFbeta3および抗体のプラスミドに加えてpBEF-EBNA1を加えて行った。また、キットに付属するEnhancer溶液は使用しなかった。検討の結果、培養期間は5または7日間とした。培養終了後、培養液を遠心し、上清を0.45μmのフィルターでろ過した。得られた培養上清は液体窒素で凍結後、-80℃の冷凍庫内で保存した。
Latent TGFbeta3の発現はssIgK-FLAG-TGFbeta3(C25S)のコンストラクトを使用し、培養液30mlあたり、30 μgのプラスミドと2.4μgのpBEF-EBNA1を共発現した。培養期間は7日で最大となったので以降の実験では7日間培養で実施した。またトランスフェクションキットに付属するEnhancer1および2溶液を添加した場合、Pro体比率が高まったため使用していない。
AKTA avant(GE healthcare)を使用し、200mlの培養上清を50mM HEPES/150mM NaCl/1mM EDTA,pH7.8溶液で平衡化した2mlのFLAG-M2レジン(Sigma社Anti-FLAG M2 agarose affinity gel #A2220)をつめたカラムにアプライした。実験は低温室内で行い、流速1ml/min.でアプライした後、平衡化バッファーで洗浄した。溶出は0.1mg/ml FLAG peptide(Sigma社 #F3290)を含む平衡化バッファーで行った。Latent-TGFbeta3を含む画分はまとめて20mM Tris-HCl,pH8.0/1mM EDTA溶液に透析した。透析後の画分はBioRad社ENrich-Qカラム(10x100mm #780-0003)に吸着後、濃度勾配0-300mM/45CV のNaCl溶液で溶出した。Pro体との分離が良くない場合、さらにGE heathcare社RESOURCE-Q1mlカラム(#17-1177-01)でも精製を試みた。イオン交換カラムでの分離の後、サンプルを限外ろ過膜で濃縮し、GE healthcare社Superdex200increase 10/300ゲル濾過カラム(#28-9909-44)で精製した。Latent-TGFbeta3のゲル濾過クロマトグラムを図18に示す。以降の実験ではこのゲル濾過画分を濃縮して使用した。
表1で示した遺伝子を挿入したプラスミドを使用してTGFbeta3改変抗体分子(1D3-mF18-latent TGFbeta3)および改変抗体分子(1D3-mF18)を発現した。両分子ともに、抗体H鎖、L鎖は同じプラスミドを使用し、TGFbeta3改変抗体分子の発現は、TGFbeta3発現プラスミドを共発現させることで抗体C末端側にコンジュゲートさせた。コンジュゲート体の発現を最適化するために、発現条件の検討をおこなった。発現に使用したプラスミド量を表2に示す。TGFbeta3のプラスミド量比を抗体と等量から2倍量、5倍量まで増加させて検討した結果、コンジュゲート体が最大となるのは2倍量で、結合率が最大となるのは5倍量であった(図20)。今後の検討ではこの2倍量か5倍量のプラスミド比で実験を行った。また、培養期間はLatent体と同様に7日でコンジュゲート体量が最大となったため、7日間培養としている。
TGFbeta3と抗体のプラスミド量比2:1または5:1でSingle arm抗体と通常の抗体(2arm)の発現を実施した。得られた培養上清100 ml毎に、PBSで平衡化したantiV5-tagレジン(MBL社V5-tagged protein Purification gel #3316)1 mlを添加し、4℃で15時間振盪しレジンに抗体を吸着させた。このレジンをCentrifuge column,10ml(PIERCE社)に詰めて、カラム容積の10倍量のPBSで洗浄した。溶出はカラム容積量の2mg / ml V5 peptide in PBS (MBL社 V5-tag peptide #3315-205)で室温30分処理し溶出した。これを5回繰り返して溶出液とした。
この溶出液を限外ろ過膜(Amicon Ultra MWCO10K, Millipore社 #UFC901024または#UFC801024)で濃縮し、ゲル濾過カラムに供した。
ゲル濾過精製は精製装置AKTAavant (GE healthcare)で行い、カラムはSuperdex200increase10/300(GE healthcare, #28-9909-44)を使用した。平衡化バッファーはPBSを使用した。Single arm 1D3-mF18-latent TGFbeta3コンジュゲート体のゲル濾過クロマトグラムを図21に示す。
電気泳動(図22)の結果、先に溶出されるPeak1が、Latent-TGFbeta3がコンジュゲートされた分子であったので、これを分取して使用した。
調製したLatent TGFbeta3およびTGFbeta3改変抗体分子 (Single arm 1D3-mF18-latent TGFbeta3コンジュゲート体または TGFbeta3がコンジュゲートされていないSingle arm 1D3-mF18) の生理活性を、HEK-Blue_TGFb細胞 (hkb-tgfb, InvivoGen)を用いて確認した。酸処理またはプロテアーゼ処理によって放出されるmature型TGFbeta3の生理活性の評価方法については、Smadシグナルの入力を分泌型アルカリフォスファターゼ(SEAP)の活性を評価することで検証した。
Latent TGFbeta3またはTGFbeta3改変抗体分子に5μg/mLのPlasmin (P1867, Sigma)プロテアーゼを添加したサンプル、標準品として4mM HClで溶解したmature型TGFbeta3リコンビナントタンパク質(243-B3-010/CF, R&D systems)をflat-bottom 96-well plateに分注し、HEK-Blue_TGFb細胞懸濁液を各5×104 cells/wellとなるように添加して、37℃のCO2インキュベーターにて一晩(20hrs)培養した。翌日、20μLの細胞培養上清に対して180μLのQUANTI-Blue (rep-qb1, InvivoGen) 発色基質を添加し、37℃, CO2インキュベーターで1時間反応後、VersaMaxスペクトロメーター (Molecular devices) にてOD655nmで分泌型アルカリフォスファターゼ(SEAP)活性を測定した。
mature型TGFbeta3リコンビナントタンパク質のEC50が約0.2ng/mLに対して、Plasminプロテアーゼ処理したLatent TGFbeta3 およびTGFbeta3改変抗体分子(1D3-mF18-latent TGFbeta3)のEC50はそれぞれ約10ng/mL前後と生理活性を示した。一方でTGFbeta3がコンジュゲートされていない抗体分子(1D3-mF18)はPlasminプロテアーゼ処理をしても生理活性は全く認められなかった。(図23)
TGFbeta3改変抗体分子のB細胞結合特異性をFACS解析により検証した。日本チャールスリバーより購入した10-14週齢・オスのC57BL/6Nマウスから、脾臓を単離した。脾細胞を調製してFACSバッファーに再懸濁し、TGFbeta3改変抗体分子、T細胞・B細胞・NK細胞・単球・樹状細胞等のマーカーに対する蛍光標識抗体を反応させた。細胞表面マーカーは、B細胞への特異性に関してはB220 (BUV395, 563793, BD), CD138 (APC, 132506, Biolegend)、T細胞に関してはCD3e (BUV737, 564618, BD), CD4 (PE-CF594, 562285, BD), CD8 (PE-Cy7, 552877, BD), CD25 (APC, 102012, Biolegend)、lymphoid DC細胞・plasmacytoid DC細胞・myeloid DC細胞・macrophageに関してはCD11b (PE, 553311, BD), CD11c (APC, 550261, BD), Gr-1 (FITC, 553127, BD)、NK細胞に関してはCD11b (PE, 553311, BD), CD49b (DX5) (BV421, 563063, BD)をそれぞれ指標にそれぞれの抗原を認識する蛍光標識抗体を組み合わせた。その後、TGFbeta3改変抗体分子のV5タグを認識する抗体(rabbit anti-V5 Ab, V8137, Sigma), BV421-conjugated donkey anti-rabbit IgG Ab (406410, Biolegend)をそれぞれ4℃, 30分間反応させ、FACSバッファーで2回洗浄後、FACS analyzer (LSRFortessaX-20, BD)にて各細胞を分画しTGFbeta3改変抗体分子の細胞特異性を確認した。
FACS解析の結果、病態モデルの脾細胞でないことからCD138陽性plasma細胞への染色を評価することができなかったが、B220を発現するB細胞への結合特異性を確認することができた(図24)。
B細胞への結合特異性が確認されたTGFbeta3改変抗体分子がSmadシグナルを実際に入力する生理活性が持つことを、リン酸化Smad2/3の検出によって評価した。日本チャールスリバーより購入した10-14週齢・オスのC57BL/6Nマウスから、脾臓を単離した。脾細胞を調製して0.5%BSA含有PBSに再懸濁し、latent TGFbeta3, TGFbeta3改変抗体分子, 5μg/mLのPlasminプロテアーゼを添加して30分間37℃のCO2インキュベーターで反応した後、BD PhosflowTM Lyse/Fix buffer (558049, BD)にて細胞を固定し、2回洗浄後にBD PhosflowTM Perm buffer III (558050, BD)にて細胞膜の透過処理を行った。さらにB220 (BUV395, 563793, BD), CD4 (PE-CF594, 562285, BD)に対する蛍光標識抗体とともにPE標識抗リン酸化Smad2/3抗体 (562586, BD PhosflowTM Smad2 (pS465/pS467)/Smad3 (pS423/pS425), BD) にて染色した後、FACS analyzerにてB細胞におけるリン酸化Smad2/3を評価することで、TGFbeta3改変抗体分子のSmadシグナルの入力効果を検証した。
その結果、未刺激に対して、mature TGFbeta1, mature TGFbeta3はリン酸化Smad2/3の割合が亢進しており、Plasmin処理したlatent TGFbeta3, TGFbeta3改変分子においても同様にリン酸化Smad2/3の割合が亢進していた。一方でTGFbeta3がコンジュゲートされていない1D3-mF18やPlasminプロテアーゼのみにおいてリン酸化Smad2/3の割合は未刺激時と変化がなかった(図25)。この結果から、B細胞への結合特異性およびSmadシグナル入力効果というTGFbeta3改変抗体分子のコンセプトが確認された。
日本チャールスリバーから購入した10-14週齢・オスのC57BL/6Nマウスの脾臓を単離し、Mouse B cell isolation kit (130-090-862, Miltenyi Biotec)によって分画したB細胞 (negative fraction)を精製した。
CellTraceTM CFSE (Carboxyfluorescein diacetate succinimidyl ester) Cell Proliferation Kit for Flow Cytometry (C34554, Molecular Probes)を用いて、20分間37℃のWater bathで反応した後、PBSで2回洗浄を行い10,000 cells/wellとなるようにround-bottom 96-well plateに分注した。その後、latent TGFbeta3, TGFbeta3改変抗体分子, 0.5μg/mLのPlasminプロテアーゼを添加した。さらに細胞増殖の刺激には5μg/mL のanti-IgM (715-005-140, Jackson)、5μg/mL のanti-CD40 (HM40-3, BD Pharmingen)、2ng/mL のrecombinant murine IL-4 (214-14, Peprotech)の刺激をそれぞれ加え、3日間37℃のCO2インキュベーターにて培養後、細胞分裂をFACS analyzer (LSRFortessaX-20, BD)を用いてTGFbeta3改変抗体分子のB細胞増殖抑制効果を検証した。標準品として、mature TGFbeta1リコンビナントタンパク (240-B-010/CF, R&D Systems)とmature TGFbeta3リコンビナントタンパク(243-B3-010/CF, R&D Systems)を用いて同じタイミングで刺激を開始した。
mature TGFbeta1のB細胞増殖抑制効果は弱かったのに対して、mature TGFbeta3と同様に、Plasminプロテアーゼ処理したTGFbeta3改変抗体分子またlatent TGFbeta3においてもB細胞増殖抑制効果が確認された(図26)。一方で、TGFbeta3がコンジュゲートされていない1D3-mF18においてはPlasminプロテアーゼ処理しても活性は全く確認されなかった。この結果は、TGFbeta1よりも TGFbeta3のB細胞増殖抑制効果ははるかに強く、TGFbeta3改変抗体分子という分子系にすることによってもその生理活性は保持されることを示している。
Mature型およびLatent型のTGFbeta3は分子の性質上、非特異的な結合および非常に速い血中消失速度が予想され、そのままの分子系では治療薬としての開発には多くの課題が想定されている。持続的に活性化B細胞を抑制するために、TGFbeta3改変抗体分子という分子系が血中半減期を伸ばすことができるか検証した。
日本チャールスリバーより購入した6週齢・オスのC57BL/6J マウスに対して、Latent TGF-beta3およびTGF-beta3改変抗体分子を1 mg/kgで尾静脈内投与した。投与後、頸静脈より経時的に採血し、血液を遠心分離することで血漿サンプルを得た。血漿サンプル中のTGF-beta3改変抗体分子およびTGF-beta3濃度をELISA法で定量した。血漿中TGF-beta3およびTGF-beta3改変抗体分子の濃度データを解析し、血漿中濃度下面積(AUCinf)および半減期(T1/2)を算出した。
TGF-beta3改変抗体分子の抗体部分のAUCおよび半減期はそれぞれ8100 ng・day/mLおよび2.09 day であった。一方、TGF-beta3 のAUCinfは、Latent TGFbeta3 として投与したときは146 ng・hour/mLであったのに対し、TGFbeta3改変抗体分子として投与したときは982 ng・hour/mL であった。また、TGF-beta3 のT1/2は、Latent TGFbeta3 として投与したときは0.116 hourであったのに対し、TGF-beta3改変抗体分子として投与したときは2.44 hourであった。本研究により、TGF-beta3を改変抗体分子として投与することで、AUCが上昇しT1/2が延長されることが明らかになった(図27および表3)。
本結果は、TGFbeta3改変抗体分子という分子形に改変することによって、Latent TGFbeta3と比較してSmadシグナル入力効果の持続時間延長が可能となること、また治療薬としての可能性を示唆している。
以下の手法によりTGFbeta3改変抗体分子によるB細胞への特異的な活性を評価する。
A) Ex vivo: B6.Cg-Tg(SBE/TK-luc)7Twc/Jマウス(SBE-Luc Tgm, 005999, Jackson Laboratory)から脾臓・リンパ節を単離する。TGFbeta3改変抗体分子、T細胞・B細胞・NK細胞・単球・樹状細胞等のマーカーに対する蛍光標識抗体を反応させてB細胞特異性を確認する。また、分画したB細胞にPlasminプロテアーゼを添加しLuciferase活性を測定する。これにより、TGFbeta3が誘導する細胞内シグナルを評価する。
B) In vivo imaging: SBE-Luc Tgマウスに対しAlexa Fluor 647標識したTGFbeta3改変抗体分子を投与し、IVIS Spectrum CT (Perkin Elmer)を用いてin vivo imagingを行う。蛍光シグナルによって抗体の分布と発光シグナルによるTGFb3を介したSmadシグナルの分布が生体内で重なることを確認することにより、B細胞へのリクルートおよびSmadシグナル入力効果というTGFbeta3改変抗体分子のコンセプトを確認する。
C) B細胞におけるSmadシグナルの確認: 上記実施例で示した1D3-mF18, 1D3-mF18-latent TGFb3, latent-TGFb3に加え、抗CD19抗体(1D3)以外のB細胞表面に結合しない抗体を有し、latent TGFb3を連結した改変抗体分子の4種のタンパク質を作成する。C57BL/6NマウスまたはSBE-Luc Tgマウスに対しこれらを尾静脈投与し、脾臓・リンパ節を単離してpSmadの割合を比較検証する。また、上記4種類のタンパクを尾静脈投与して脾臓・リンパ節を単離後、T細胞・B細胞・NK細胞・単球・樹状細胞等のマーカーに対する蛍光標識抗体を指標に各細胞を分画してRNAを抽出し、次世代シークエンサーを用いて網羅的な発現遺伝子解析を行うことによって、各細胞のTGFbeta3シグナル関連遺伝子の発現変化を評価することでTGFbeta3改変抗体分子のコンセプトを確認する。
以上の評価により、TGFbeta3改変抗体分子が標的とするB細胞に特異的に活性を有することが証明される。
Claims (10)
- TGF-beta3を含むB細胞活性化の抑制剤。
- 前記B細胞は自己反応性B細胞である請求項1に記載の抑制剤。
- 前記TGF-beta3はB細胞による抗体産生を抑制する請求項1又は請求項2に記載の抑制剤。
- 請求項1乃至3のいずれかに記載の抑制剤を含む自己免疫疾患の治療剤。
- TGF-beta3と抗体又は抗体断片とが連結された分子を含む自己免疫疾患の治療剤。
- 前記抗体又は抗体断片はB細胞を認識する可変領域を含む請求項5に記載の治療剤。
- 前記可変領域はCD19、CD20、CD40、CD22、IL21R、BAFF-R、BCMA、TACI、CD27、又はCD138に含まれるいずれか1つまたは複数をマーカーとして前記B細胞を認識する請求項6に記載の治療剤。
- TGF-beta3を認識する第一の可変領域と、B細胞を認識する第二の可変領域とを含む多重特異性抗体を含む自己免疫疾患の治療剤。
- 前記自己免疫疾患は全身性エリテマトーデス、天疱瘡、多発性硬化症、視神経脊髄炎、ANCA関連血管炎、関節リウマチ、移植臓器拒絶、シェーグレン症候群、若年性皮膚筋炎、重症筋無力症、又はバセドウ病、橋本病等の自己免疫性甲状腺疾患のいずれかである請求項4〜8のいずれかに記載の治療剤。
- 請求項4〜9のいずれかの治療剤を含む医療用キット。
