Abstract
Description
(1) 以下の何れかの抗体から選択される、エイズウイルスのエンベロープ糖タンパク質gp120のV3ループを認識するモノクローナル抗体。
(a) H鎖の可変領域(VH)のアミノ酸配列として配列番号1に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号2に記載のアミノ酸配列を有する抗体;
(b) H鎖の可変領域(VH)のアミノ酸配列として配列番号3に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号4に記載のアミノ酸配列を有する抗体:
(a) H鎖の可変領域(VH)のアミノ酸配列として配列番号5に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号6に記載のアミノ酸配列を有する抗体;
(b) H鎖の可変領域(VH)のアミノ酸配列として配列番号7に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号8に記載のアミノ酸配列を有する抗体;
(c) H鎖の可変領域(VH)のアミノ酸配列として配列番号9に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号10に記載のアミノ酸配列を有する抗体:
(4) 受託番号FERM BP−11020、受託番号FERM BP−11022、又は受託番号FERM BP−11023を有する細胞が産生する、エイズウイルスのエンベロープ糖タンパク質gp120のCD4結合部位を認識するモノクローナル抗体。
(5) 長期非進行症例のHIV感染患者のB細胞をgp120に対する結合性についてスクリーニングし、gp120結合抗体を産生するB細胞を選択することによって得られる細胞によって製造される、(1)から(4)の何れかに記載の抗体。
(6) 長期非進行症例のHIV感染患者のB細胞をgp120に対する結合性についてスクリーニングし、gp120結合抗体を産生するB細胞を選択し、選択されたB細胞が産生する抗体を採取することを含む、(1)から(5)の何れかに記載の抗体の製造方法。
(8) 受託番号FERM BP−11020、受託番号FERM BP−11022、又は受託番号FERM BP−11023を有する、(2)に記載の抗体を産生する細胞。
(9) (1)から(5)に記載の少なくとも1種類以上のモノクローナル抗体を含む、HIV感染症の予防及び/又は治療薬。
(10) (1)又は(3)に記載のモノクローナル抗体と、(2)又は(4)に記載のモノクローナル抗体とを組み合わせて含む、HIV感染症の予防及び/又は治療薬。
1.本発明のモノクローナル抗体
本発明のモノクローナル抗体の第一の例は、以下の何れかの抗体から選択される、エイズウイルスのエンベロープ糖タンパク質gp120のV3ループを認識するモノクローナル抗体である。
(a) H鎖の可変領域(VH)のアミノ酸配列として配列番号1に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号2に記載のアミノ酸配列を有する抗体;
(b) H鎖の可変領域(VH)のアミノ酸配列として配列番号3に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号4に記載のアミノ酸配列を有する抗体:
(a) H鎖の可変領域(VH)のアミノ酸配列として配列番号5に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号6に記載のアミノ酸配列を有する抗体;
(b) H鎖の可変領域(VH)のアミノ酸配列として配列番号7に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号8に記載のアミノ酸配列を有する抗体;
(c) H鎖の可変領域(VH)のアミノ酸配列として配列番号9に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号10に記載のアミノ酸配列を有する抗体:
本発明は、上記した本発明のモノクローナル抗体を産生する細胞にも関する。本発明のモノクローナル抗体は当該細胞を用いて製造することができる。以下、本発明のモノクローナル抗体を産生する細胞の取得方法を説明する。
本発明の抗体をコードする遺伝子は、上記した本発明のモノクローナル抗体を産生するEBV 形質転換ヒト細胞に由来するmRNAを用いてcDNAライブラリーを作製し、その中から本発明のモノクローナル抗体をコードするcDNAを含むプラスミドを単離することによって取得することができる。mRNAは、EBV 形質転換細胞を培養し、グアニジニウムイソチオシアネート溶液で細胞を溶解させた後に抽出することによって調整することができる。得られたmRNAを鋳型としてcDNAを作製し、これをベクターに組み込むことでcDNAライブラリーを作製する。このcDNAライブラリーを用いて、本発明のモノクローナル抗体をコードする遺伝子を取得することができる。
本発明のモノクローナル抗体は、必要に応じて、薬学的に許容される適当な担体、賦形剤、希釈剤などを用いて、HIV感染症の予防及び/又は治療薬として用いることができる。HIV感染症の予防及び/又は治療薬としては例えば注射剤として製剤することができる。HIV感染症の予防及び/又は治療薬の投与量は、患者の症状の程度、年令及び体重、投与方法などに依存し、有効成分である抗体の重量としては通常、約10ng〜約100mg/kg体重の範囲である。
本発明では、本発明のモノクローナル抗体またはその断片を用いて免疫アッセイを行うことによって、HIVウイルスを検出することもできる。この方法では、好ましくは、少なくとも下記(a)及び(b)の工程を含む。
(a)本発明のモノクローナル抗体またはその断片と試料とを反応させる工程;及び
(b)工程(a)で形成した抗原抗体複合体と、検出のための標識抗体とを反応させる工程;
末梢血B細胞にEBウイルスを感染させ、クローニングを繰り返し、抗gp120活性をもつ抗体産生細胞を分離する。作成方法は、基本的に我々が以前ヒトT 細胞白血病ウイルス1型(Human T-cell leukemia virus type 1; HTLV-1)感染症例について行った方法を応用したものである(Matsushita S., et al., Proc. Natl. Acad. Sci. (USA) 83:3672-3676, 1986)。広範なウイルス株に対して、中和活性を持つ一人の長期非進行症例より、末梢血をヘパリン含有採血管に採血し(20〜30ml)、ファイコールハイパックによる比重遠心分離法にて、末梢血単核球(peripheral blood mononuclear cell; PBMC)分画を採取する。得られたPBMCからダイナビーズ、イムノマグネットにてCD8陽性細胞を除去する。具体的には、PBMCをダイナビーズCD8と1時間4℃でインキュベートし、磁気ビーズを磁気器具にて除去し、CD8除去PBMC(CD8-depleted PBMC)を得る。CD8-depleted PBMC1〜10x106に対し、EBウイルス産生株B95-8(ATCC)の培養上清 50%を含む培地(15%fetal calf serum;FCS 含有RPMI1640)2〜5mlに浮遊し、24well culture dish (Linbro,ICN) に移し、37℃、5%CO2含有インキュベーターにて12〜18時間培養する。この間に末梢血中のB細胞にEBウイルスが感染し、その104に1株の割合で不死化細胞が得られると考えられる。培養した細胞を集め遠心して古い培地を捨てて、新しい培地にCD8-depleted PBMC として1 wellあたり1X104以下となるように、96well flat bottom culture plate (Falcon ) に分注し、さらに37℃、5%CO2インキュベーターにて培養する、7日から10日ごとに培地を半分新しいものに交換しながら、B細胞株の増殖を待つ。増殖してきた細胞は、48well,または24wellのculture dish (Linbro,ICN)にExpandし、増殖する細胞の培養上清を回収し、後述の“gp120-capture assay”にてスクリーニングする。スクリーニングで繰り返し陽性と判断されるクローンに関しては。正常人のPBMCにX線照射した細胞をフィーダーとして、限界希釈法にてクローニングし、抗gp120活性をもつ抗体産生細胞を分離する。必要であればクローニングの手順を繰り返す。
抗gp120活性をもつ抗体産生細胞のスクリーニングには“gp120-capture ELISA”をもちいた(J.P.Moore et al., J.Virol.67,p6136-6151,1993)。すなわち、実験前日、96well polypropyren plate (Falcon)にCCbuffer (Carbonate-bicarbonate buffer, pH 9.6)で13.3μg/mlに調整した抗gp120-C5 sheep抗体(D7324, Aalto Bioreagents, Dublin, Ireland) を50μl加え、4℃で一晩静置する。上清を取り除いた後、blocking buffer (2% ウシ血清アルブミン;BSA、0.1% Azide、PBS、pH7.2) 175μlを加え、室温で30分間静置する。ELISA wash buffer (0.15% Tween20、PBS pH7.2)で3回ウェルを洗浄後、単量体gp120(gp120-SF2;Austral Biologicals、San Ramon, California)を50μlずつ加える。2時間室温に静置した後、ELISA wash bufferで3回洗浄し、"gp120-captured plate"を作成する。このplateにB細胞培養上清を加えて室温で2時間反応させる。ELISA wash bufferで3回洗浄し、ELISA bufferで1000倍希釈した抗ヒトIgG-ALP100μlを加えて室温で1時間静置する。ELISA wash bufferで3回洗浄し、Substrate buffer (Diethanol amine buffer pH9.85)にPhospatase substrate (SIGMA)を溶かし各100μlをウェルに加えて室温で10〜30分静置後マイクロプレートリーダーにて吸光度(405nm)を測定する。このようにしてgp120結合抗体を産生するB細胞クローン20種類が得られた(表1)。
gp120に反応する抗体は反応エピトープによって次の4種類に分類される。すなわち、CD4結合部位(CD4binding site;CD4bs)、CD4誘導エピトープ(CD4induced epitope;CD4i)、 第3可変部位(V3loop;V3L)及びこれらに分類できないもの(other epitopes)である。V3Lに結合する抗体かどうかはV3Lのアミノ酸配列を持つ合成ペプチドを用いたELISA(V3-peptide ELISA)で判定する。今回は抗体の由来する症例のウイルスのV3配列に最も近いJR-FL株のV3配列(NNT20; NNTRISIHIGPGRAFVTIGK)を用いた。すなわち、実験の前日にPBSで5μg/mlに希釈したペプチド(NNT20)を96well polypropyren plateの各wellに100μlを加え、4℃で一晩静置する。上清を取り除いた後、blocking buffer 175μlを加え、室温で30分間静置する。ELISA wash bufferで3回洗浄後、B細胞培養上清を加えて室温で2時間反応させる。ELISA wash bufferで3回洗浄し、抗ヒトIgG-ALP100μlを1時間反応させ、洗浄後にALP−Substrateを用いて発色させ、吸光度(405nm)を測定した。得られた20種類の抗体に関してV3-peptide ELISAをおこなったところ、6種類がV3に反応する抗体であった。
HIV-1はウイルス粒子上および慢性感染細胞表面に標的細胞に膜融合をおこす機能的エンベロープを持つことが知られている。作成した抗体がこの機能的エンベロープを認識できるかどうか検討するため、HIV-1JR-FL株慢性感染細胞(PM1/JR-FL)の細胞表面に対する抗体の結合活性をフローサイトメーターにて調べた。また、この反応性に関してsCD4の影響が見られるかどうか検討した。2x105のPM1/JR-FLを試験管にとり、1μg/ml にFACS buffer(2%BSA, 0.02% Azide in PBS; pH 7.23)で希釈調整したsCD4またはFACS bufferのみを50μl加えた。15分反応させ、FACS buffer にて1〜3μg/mlに希釈した単クローン抗体を50μl加え室温で45分反応させた。FACS bufferにて2回洗浄後、FACS buffer で1:200倍に希釈したFITC結合抗ヒトIgG抗体(Sigma)50μl加えて4℃、暗所にて30分間保温した。その後、FACS bufferにて2回洗浄し、0.5mlの固定液{2%(w/v) paraformaldehyde in PBS}で固定した。固定したサンプルは、FACSCalibur(Beckton-Dickinson)にて解析した。取り込んだデータはCell Quest(Beckton-Dickinson)にて解析した。図2に示すようにV3L抗体は5種類とも感染細胞表面への明らかな結合活性が認められた。しかも、この活性はsCD4存在下に増強された。sCD4非存在下では1D9,2F8,3E4,3G8の4クローンに比べ、1C10(0.5γ)及び5G2では強い反応が認められた。CD4bsに分類される抗体のうち3D6(0.5δ),42F9,49G2では感染細胞表面への結合活性が認められたが、4E3と7B5では認められなかった。 gp120単量体での反応と異なり、3量体から成る機能的エンベロープではsCD4によるCD4bs抗体の結合阻害は認められなかった。一方、CD4i抗体に関しては、4種類の抗体ともsCD4非存在下では弱い反応しか認めないが、sCD4存在下にはV3抗体に匹敵するほどの反応性の増強を認めた。Epitopeが同定できなかった5種類の抗体に関してはどれも感染細胞表面への結合活性を認めなかった。
20種類の抗体のうち細胞表面への結合活性及び抗体産生の量などを勘案して15種類に関して細胞を増やし、培養上清を集めてフィルターし、プロテインAカラムまたはプロテインGカラム(GE Healthcare)にて単クローン抗体を精製した。精製した単クローン抗体を用いて、実験室でよく用いられている4種類のHIV-1株のenvelopeを用いたシュードタイプに対する中和活性を抗体濃度10μg/ml及び2μg/mlで検討した(図3)。シュードウイルスの作製については293T細胞をトランスフェクション前日に100mmコラーゲンコートプレート(IWAKI)に4×106になるように蒔き、37℃で一晩培養した後、5μgのpNL4-3-Luc-E-R-または5μgのpSG3ΔEnv(Li M. et.al,J.Virol.79,p10108-10125,2005)、4.5μgのエンベロープ発現ベクターおよび0.5μgのpRSV-REVベクターをEffectene (QIAGEN)にて導入し、37℃、5%CO2にて培養した。24時間後に上清を回収し、0.2μmフィルターに通し、分注して-80℃にて保存した。得られたシュードウイルスはp24 Antigen ELISA kit (ZeptoMetrix Co.)でp24の抗原量を測定した。シュードウイルスを用いたSingle-round中和実験は以下のようにおこなった。実験前日にGHOST-hi5細胞またはTZM-bl細胞を平底96ウェル細胞培養用プレート(Falcon)に200μlあたり2×104になるように蒔いた。コンフルエントの70%程度まで増殖したら、200-500TCID50のpseudotyped virusと各濃度の抗体とを10%ウシ胎児血清(FCS)、0.1mg/ml G418、0.05mg/ml Hygromycin-B、5μg/ml Puromycin、20μg/ml Polybrane(GHOST-hi5の場合)または10%FCS、10μg/ml DEAE-dextran(Pharmacia Biotech, TZM-blの場合)を含むDMEMを混合し、15分間氷上で静置した。培養液を除去した標的細胞に100μlの抗体/ウイルス混合溶液を加えて37℃、5%CO2で2時間インキュベートし、ウイルスを標的細胞に吸着させた。その後さらに100μlの上記の抗生物質等を含むDMEMを加え、37℃、5%CO2にて2日間培養した。上清を除去し、PBS(pH 7.4)にて3回洗い、PBSで5倍希釈した30μlのLysing buffer(Luc PGC50; 東洋インキ、GHOST-hi5の場合)またはLysing solution (Applied Biosystems、TZM-blの場合)を加え15分間振盪した。そのうち20μlを発光測定用のプレート(Coster 3912)へ移し、luciferase基質(ピッカジーン、東洋インキ)を100μl加え、10秒後にTR477 microplate luminometer(Applied Biosystems)にて蛍光強度を測定した (GHOST-hi5の場合)。TZM-bl細胞の場合は、発光測定用のプレートに移したあと、Reaction buffer diluentで50倍希釈したGalacto-Star (Applied Biosystems)を100μl加え、1時間暗所にて静置した。その後、TR477 microplate luminometerにてβ-galactosidase活性を測定した。中和活性は、{1−(t−c)/(n−c)}×100(t; サンプルの蛍光強度、c; 細胞のみのバックグラウンド蛍光強度、n; 抗体無しサンプルの蛍光強度)で計算した。同じ抗体濃度で3ウェルにて実験を行い、独立した実験を2-3回ずつ行った。
臨床分離株を含む17種のウイルス株に対する中和活性を検討した(表2)。中和実験は以下のようなMTT assayを用いた。方法は96well round-bottom micro culture plateにて、各濃度の抗体存在下で100 TCID50のウイルスを2x103のPM1/CCR5に感染させ、37℃で7日間培養した。それぞれのwellから100μlの培養液を取り除き、phosphate-buffered saline (PBS)に溶かしたMTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) 溶液 (7.5mg/ml) 10μlを加え、37℃で3時間インキュベートした。4% Triton X-100 (vol/vol)を含んだacidified isopropanol 100μlを各wellへ加えてformazan crystalを遊離させた。マイクロプレートリーダーにて吸光度(570nm)を測定して、細胞の生存率を算出した。実験は2-3回繰り返した。50%のウイルス感染を阻止する抗体濃度(IC50)を算出し、IC50にあたる抗体濃度を色分けして示した。
gp120はCD4分子と結合後に立体構造変化を起こすことが知られている。一方、CD4bs抗体が立体構造変化を起こすかどうかに関しての研究はほとんどなかった。我々は、CD4bs抗体がgp120に結合後にgp120に立体構造変化を起こし他の抗体の反応性を変化させる可能性を考えた。前述のgp120-captureELISAの方法で“gp120-captured plate"を作成し、CD4bs抗体のひとつである0.5δ(3D6)またはコントロールの抗体(8D11)を5μg/mlの濃度で15分反応させ、これにBiotin(Pierce, Rockford IL)を結合させた抗体、1C10,3E4,3D6,4C11を各濃度で2時間反応させた。ELISA wash buffer で洗浄後Avidin-ALP(Zymed)を1時間反応させ基質を加えて発色させる。結果を図4に示す。ビオチン化した0.5γ(1C10)や3E4のV3抗体は0.5δ存在下にgp120に対する反応性が著名に増加した。一方、ビオチン化した0.5δ(3D6)や4C11の反応性は0.5δ存在下に抑制された。このように、CD4bs抗体の0.5δは単独でgp120に立体構造変化を起こしV3抗体(特に0.5γ)の反応性を飛躍的に拡大させることがわかった。
前述のように0.5δは0.5γの反応性を増加させることから、0.5δと0.5γの中和活性におけるcombination効果を検討した。各濃度の0.5δと0.5γの組み合わせを用いてJR-FL株に対する中和活性をMTT assay を用いて検討した。図5に示すように二つの抗体の併用で中和活性の相乗的増強が観察された。相乗効果の解析はChow/Talaleyらの分析法と3-Dimensional分析法を用いた。これらのin vitroの観察は、in vivoではV3抗体とCD4bs抗体が相互作用して、さらに強力な中和反応を起こしているのではないかと推測させる。これらのデータから広範囲で強力な中和抗体反応には広範囲の株に反応するV3抗体とCD4bs抗体の両者が必要と考えられた。
国際的にAIDSワクチン開発のデータを評価するため、標準となるウイルスが米国の機関(NIAID ARRP)から供給されている。12種類のシュードウイルスからなるsubtype (clade) B ウイルスのパネル(Standard Virus Panels B;SVPB)を用いた中和データを表3に示す。表3に示すように0.5γ(IC10)は50μg/mlで7/12 、150μg/mlまでの濃度で10/12のウイルスの増殖を50%以上抑制できた。一方、従来技術の抗体であるb12は8/12、 2G12は6/12、447-52Dでは1/12、KD247は3/12という結果であった。従来技術であるb12、2G12 、447-52D(447D)のデータは論文からの引用である(Li M. et.al,J.Virol.79,p10108-10125,2005)。b12と2G12 は50μg/mlまでのデータであり、447-52D(447D)は25μg/mlまでのデータしかないので必ずしも比較できないが、これらの従来技術とは明らかに異なり、0.5γ(1C10)は幅広い中和活性を示していることがわかる。CD4bs抗体であるb12も広汎な中和活性を持つが、自己抗体によく見られるheavy chain CDR3が18アミノ酸と長い特殊な抗体である(Saphire EO.et al., Science239,p1155-1159)。一方、0.5γ(1C10)のCDR3は11アミノ酸からなる。
我々はそれぞれの代表的中和単クローン抗体、抗V3:0.5γ(1C10),5G2, CD4bs: 0.5δ(3D6), 42F9, 49G2について抗原結合部位の遺伝子配列を決定した。結果を図6から図10、及び配列表の配列番号1から20に示す。方法はそれぞれの細胞株からRNAを抽出しMarksらの方法に従ってVH及びVLをPCRで増幅し同定した(Marks, J. D.et.al., J. Mol. Biol., 222: 581-597, 1991)。

これらの新規単クローン抗体はクレイドBのウイルスに対して広範で強力な中和活性を示した。特に、0.5γ(IC10)は現在使用しうる抗体の最高水準にあると考えられる。これらの抗体はワクチン開発に重要な役割を果たすだけでなく、抗体そのものまたはその断片、もしくはその遺伝子をもとに作製される分子はAIDS/HIV感染の予防・治療に用いられる可能性がある。
Subtype (clade) B ウイルスのパネル(SVPB)と同様に, subtype Cウイルスのパネルも供給されている。図11に示すようにsubtype Cパネルの中の2つ (SVPC13とSVPC15)のウイルスに対して、CD4bs抗体の0.5δ(3D6)と49G2と42F9はコントロール抗体に比べ明らかに中和活性が認められた。また、subtype AEウイルスである93TH976.17と93TH966.8に対してもこれらのCD4bs抗体は中和活性を示していた。これら3つのCD4bs抗体は、subtype B以外のウイルスにも中和が見られることから、抗体によるcross-clade 中和の研究の上でも重要であると考えられる。
gp120がCD4に結合した後に、V3はcoreceptor (CCR5またはCXCR4)に結合し、ウイルスは細胞内に侵入する。0.5γや5G2のエピトープであるV3と結合のカウンターパートであるCCR5に対する阻害剤(maraviroc、MVC)は臨床応用の面からも重要な組み合わせと考えられる。そこでV3抗体とCCR5阻害剤(MVC)の組み合わせでJR-FL株に対する中和活性をMTT assayを用いて検討した。図12に示すようにV3抗体とMVCの併用で中和活性の相乗的増強が観察された。相乗効果の解析は3-Dimensional分析法を用いた。これらのin vitroの観察は、V3抗体とCCR5阻害剤が相互作用して、さらに強力なウイルス抑制効果を起こしていると推測させる。これらのデータから臨床での投与においてV3抗体とCCR5阻害剤との併用は、治療効果を増強させうるのではないかと考えられる。
Claims (10)
- 以下の何れかの抗体から選択される、エイズウイルスのエンベロープ糖タンパク質gp120のV3ループを認識するモノクローナル抗体。
(a) H鎖の可変領域(VH)のアミノ酸配列として配列番号1に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号2に記載のアミノ酸配列を有する抗体;
(b) H鎖の可変領域(VH)のアミノ酸配列として配列番号3に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号4に記載のアミノ酸配列を有する抗体: - 以下の何れかの抗体から選択される、エイズウイルスのエンベロープ糖タンパク質gp120のCD4結合部位を認識するモノクローナル抗体。
(a) H鎖の可変領域(VH)のアミノ酸配列として配列番号5に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号6に記載のアミノ酸配列を有する抗体;
(b) H鎖の可変領域(VH)のアミノ酸配列として配列番号7に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号8に記載のアミノ酸配列を有する抗体;
(c) H鎖の可変領域(VH)のアミノ酸配列として配列番号9に記載のアミノ酸配列を有し、L鎖の可変領域(VL)のアミノ酸配列として配列番号10に記載のアミノ酸配列を有する抗体: - 受託番号FERM BP−11021、又は受託番号FERM BP−11060を有する細胞が産生する、エイズウイルスのエンベロープ糖タンパク質gp120のV3ループを認識するモノクローナル抗体またはその断片。
- 受託番号FERM BP−11020、受託番号FERM BP−11022、又は受託番号FERM BP−11023を有する細胞が産生する、エイズウイルスのエンベロープ糖タンパク質gp120のCD4結合部位を認識するモノクローナル抗体。
- 長期非進行症例のHIV感染患者のB細胞をgp120に対する結合性についてスクリーニングし、gp120結合抗体を産生するB細胞を選択することによって得られる細胞によって製造される、請求項1から4の何れかに記載の抗体。
- 長期非進行症例のHIV感染患者のB細胞をgp120に対する結合性についてスクリーニングし、gp120結合抗体を産生するB細胞を選択し、選択されたB細胞が産生する抗体を採取することを含む、請求項1から5の何れかに記載の抗体の製造方法。
- 受託番号FERM BP−11021、又は受託番号FERM BP−11060を有する、請求項1に記載の抗体を産生する細胞。
- 受託番号FERM BP−11020、受託番号FERM BP−11022、又は受託番号FERM BP−11023を有する、請求項2に記載の抗体を産生する細胞。
- 請求項1から5に記載の少なくとも1種類以上のモノクローナル抗体を含む、HIV感染症の予防及び/又は治療薬。
- 請求項1又は3に記載のモノクローナル抗体と、請求項2又は4に記載のモノクローナル抗体とを組み合わせて含む、HIV感染症の予防及び/又は治療薬。
