Abstract
Description
細胞膜に局在するいくつかのヒアルロン酸結合受容体については、今日までに、例えば、CD44(リンパ球抗原群Clusters of Differentiationのうちの一つ)、LYVE−1(lymphatic vessel endothelial HA receptor)、RHAMM(Receptor for Hyaluronan−Mediated Motility)、LEC(Liver Endothelial Cell clearance)受容体などが同定されており、これらヒアルロン酸結合受容体の中で、LYVE−1は322個のアミノ酸残基からなる受容体として見出された(J.Cell Biol.144,789−801(1999)参照)。
LYVE−1のmRNAは脾臓、リンパ節、胎児肝臓、心臓、肺など全身に広く発現されており、LYVE−1受容体は、それら組織内のリンパ管内皮に局在することが明らかにされている。このことから、LYVE−1は、形態的に毛細血管と区別が困難なリンパ管の同定に利用できると考えられる。
例えば、悪性腫瘍のリンパ行性転移においては、腫瘍が周囲の組織へと浸潤した後、リンパ管壁を破壊性に浸潤してリンパ管に侵入し、リンパ行性に運搬されて、原発巣とは離れた部位に転移する行程を経るが、この浸潤過程において、リンパ管を含む宿主組織の破壊がもたらされる。
従って、病理組織におけるリンパ管の詳細な所見が得られれば、その腫瘍の破壊性浸潤性増殖の有用な病理像となるものと考えられる。
これまでに、腫瘍がリンパ管壁を破壊性に浸潤してリンパ管に浸潤する様子をとらえた報告がいくつかなされている(高沢 博:「実験的リンパ行性転移形成における移植部リンパ管の電顕的研究」日本癌学会総会記事、30,296(1971)、荒木 京二郎:「癌の浸潤初期とリンパ管侵襲の電顕的研究−N−Methyl−N’−nitro−n−nitrosoguanidine(MNNG)誘発ラット胃癌における観察−」岡山医誌、91,659−669(1979)、James N.,et al:「Growth and metastasis of Lewis lung carcinoma in the footpad of mice.」Expl.Cell Biol.、56,221−228(1992)、Carr I.,et al:「The fine structure of neoplastic invasion:invasion of liver,skeletal muscle and lymphatic vessels by the Rd/3 tumour.」J.Pathol.、118,91−99(1976)、Paku S.,et al:「Ultrastructural analysis of experimentally induced invasion in the rat lung by tumour cells metastasizing lymphatically.」Anticancer Res.、6,957−966(1986)参照)。
しかし、これらの報告は電子顕微鏡による細胞レベルの研究であり、組織においてリンパ管に浸潤する腫瘍の様子を組織レベルで観察した報告はない。
一方、病理組織における病理像を染色によって観察するための、組織等の染色方法が種々開発されてきた。例えば、酵素抗体法などの免疫組織化学的な染色方法は、特異性、再現性等に優れており、本発明による方法もこの範疇に属するものである。しかしながら、リンパ管を染色の標的とし、これを選択的に染色する方法はこれまで知られていない。
すると、種々のペプチドフラグメントの中でも、LYVE−1受容体のアミノ酸配列上の領域であって、▲1▼親水性領域であること、▲2▼糖付加されない領域であること、▲3▼βターン構造をとる領域であること及び▲4▼細胞内領域であることの4つの条件を満たすアミノ酸配列のうち、GenBankにAccession number AF118108の下に登録されたアミノ酸配列における第297位〜第308位に相当する、以下の配列:
で表されるペプチドフラグメントを用いて、上記の抗体の調製及び免疫染色を行ったところ、その染色がリンパ管に特異的であることがわかった。さらに、本発明の抗体を用いた免疫染色の結果から、生体組織におけるリンパ管の詳細な所見を得ることができた。また、特に、通常は確認が困難な早期の悪性腫瘍の破壊性浸潤性増殖についての病理学的所見を得ることができたのである。
すなわち、本発明は、配列番号1に記載のアミノ酸配列からなるペプチドフラグメント又はそのC末端若しくはN末端にリンカーとして一つのアミノ酸が付加されたペプチドフラグメントに関するものであり、さらに当該ペプチドフラグメントに対する抗体にも関する。
また、本発明は、以下の配列:
で表される、上記酸配番号1で表されるペプチドフラグメントをコードするDNAフラグメントにも関する。
また、本発明は、ヒトリンパ管特異的ヒアルロナン受容体−1(LYVE−1)に特異的である、上記抗体にも関する。
また、本発明は、配列番号1に記載のアミノ酸配列において、1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなり、かつ上記抗体によって認識されるペプチドフラグメント又はそのC末端若しくはN末端にリンカーとして一つのアミノ酸が付加されたペプチドフラグメント及びそれらをコードするDNAフラグメントにも関する。
さらに、本発明は、上記抗体を調製するための、上記ペプチドフラグメントの使用にも関する。
加えて、本発明は、上記抗体を含むことを特徴とする、ヒトリンパ管免疫染色用組成物及びヒトリンパ管免疫染色用試薬キットにも関する。
ここで、当該ヒトリンパ管免疫染色用試薬キットは、本発明の抗体の他、3%過酸化水素水等の内因性パーオキシダーゼのブロッキング試薬、ビオチン化標識抗ウサギイムノグロブリン抗体、パーオキシダーゼ標識ストレプトアビジン、トリス塩酸緩衝液等の基質緩衝液、DAB(3,3′−ジアミノベンジジン四塩酸塩)等の発色試薬及び/又は0.8%過酸化水素水等の基質試薬等を含むことができる。
また、本発明は、採取したヒトリンパ管を、本発明の抗体を用いて特異的に免疫染色する方法にも関する。
さらに、本発明は、本発明の抗体を用いてヒトリンパ管を特異的に免疫染色することによる、悪性腫瘍における浸潤度の測定方法、及び当該浸潤度を測定することによる、悪性腫瘍の悪性度の診断方法にも関する。
さらに加えて、本発明は、本発明の抗体を用いたヒトリンパ球の分離、精製、すなわちリンパ管細胞及びその前駆細胞の分取にも関する。細胞の分取には、例えば、細胞分離機器(セルソーター)、磁気細胞分離法等を利用することができる。また、分取した細胞は、例えば、リンパ管浮腫の軽減を目的としたリンパ管新生治療等に用いることができる。
ペプチドフラグメントの調製
本発明のペプチドフラグメントの調製は、市販の自動ペプチド合成装置を用いて容易に行うことができる。
抗原の調製
ペプチドフラグメントの抗原性を高めるために、ペプチドフラグメントにキャリアタンパク質を結合させる。また、ペプチドフラグメントとキャリアタンパク質との結合には、立体障害等による抗原−抗体反応の阻害を避けるため、リンカー及びスペーサーを用いる。すなわち、抗原として、ペプチドフラグメント−リンカー−スペーサー−キャリアタンパク質の結合物を用いるのが有利である。
抗原の調製において用いることのできるリンカーとしては、例えば、システイン、グルタミン酸、チロシン等の、ペプチドフラグメントとスペーサーとの結合を仲介する能力、すなわち、リンク能力を有するアミノ酸が挙げられ、好ましくはシステインを挙げることができる。
ここで、リンカーは、ペプチドフラグメントのC末端又はN末端のどちらに結合することもできるが、N末端に結合することが好ましい。
リンカーをN末端に結合するほうが好ましい理由は次のとおりである。すなわち、ペプチドフラグメントはC末端から合成されるため、リンカーをN末端に結合することにより、不完全長のペプチドフラグメントがキャリアタンパク質と結合されるのを防ぐことができる。また、C末端にシステインがある場合は、ラセミ化が起こることがあり、その防止措置を講じなければならず、収量も低くなるからである。
また、抗原の調製において用いることのできるスペーサーとしては、例えば、MBS(m−マレイミドベンゾイル−N−ヒドロキシスクシニミドエステル)、SMCC(スクシニミジル 4−(N−マレイミドメチル)シクロヘキサン−1−カルボン酸)、Sulfo−SMCC(スルホスクシニミジル 4−(N−マレイミドメチル)シクロヘキサン−1−カルボン酸)又はSulfo−MBS(m−マレイミドベンゾイル−N−ヒドロキシスルホスクシニミドエステル)等の架橋試薬を挙げることができ、MBSが好ましい。
さらに、抗原の調製において用いることのできるキャリアタンパク質としては、例えば、牛アルブミン、卵白アルブミン、ミオグロビン及びスカシ貝ヘモシアニン(KLH)等のタンパク質が挙げられ、好ましくはKLHが有利に用いられる。
ペプチドフラグメントとリンカーとの結合は、自動ペプチド合成装置により容易に行うことが出来る。また、スペーサー及びキャリアタンパク質の結合も従来の方法により容易に行うことができる。
抗体の調製
上記抗原の調製において得られた抗原をアジュバントと共に乳化して動物に免疫する。免疫後、動物を放血させ、抗血清を分離する。この抗血清からアフィニティークロマトグラフィー法により抗体を分取する。
抗体の調製においては、免疫動物として、例えば、マウス、ウサギ、ラット、羊等を用いることができるが、好ましい免疫動物としては、ウサギ又は羊を挙げることができる。
また、一般的なモノクローナル抗体調製方法により、本抗体をモノクローナル抗体として調製することもできる。
抗体の免疫特異性の検討
LYVE−1遺伝子を発現させた細胞より通常の方法にてタンパク質試料を得、これをウェスタンブロットティングによりナイロンフィルターに転写してフィルター試料とした。このフィルター試料を複数作製した。フィルター試料の一部は本発明のLYVE−1抗体と反応させ、残りのフィルター試料を、本発明のLYVE−1抗体と免疫に使用したペプチドを予め反応させておいた試液と反応させた。反応させたフィルターは、酵素結合第2抗体を用いた染色方法等の一般的な染色方法により染色した。
その結果、本発明のLYVE−1抗体と反応させたフィルター試料にはLYVE−1とLYVE−1抗体とが結合したバンドが確認され、本発明のLYVE−1抗体と免疫に使用したペプチドを予め反応させておいた試液を反応させたフィルター試料にはそのバンドが確認されないことから、本発明のLYVE−1抗体はLYVE−1に特異的であることが確認された。
組織染色
ヒト正常組織の固定パラフィン切片又は凍結切片を作製し、本発明のLYVE−1抗体を反応させた。そして、酵素抗体法、蛍光抗体法等の一般的な染色法により染色を行った。
その結果、組織切片のリンパ管のみが染まり、その他の部分は染まらなかった。この結果から、本発明のLYVE−1抗体は、リンパ管を特異的に認識する抗体であることがわかった。さらに、癌浸潤が見られる標本においてLYVE−1抗体に陽性を示す部分がリンパ管であることからも、本発明のLYVE−1抗体がリンパ管に特異的であることがわかった。
以下、実施例により本発明をさらに具体的に説明する。
図2は、本発明の抗体を用いたヒトの各種正常及び悪性腫瘍の組織の染色図である。図2の各染色図において、矢印(arrow)はLYVE−1抗体により染色されたリンパ管を指し、矢じり(arrowhead)はvon Willebrad factor(vWf)により染色された血管を指す。
図2−aは、全長のLYVE−1のcDNAを組み込み、蛋白発現させたcos7細胞をLYVE−1抗体により免疫染色した図である。図2−aにおいて、LYVE−1は主に細胞表面の細胞膜に発現がみられ、細胞質にも軽度の発現を認めた。
図2−b〜dは、胃組織の連続切片の染色図である。図2−bはHematoxylin−Eosin染色(HE染色)によるもの、図2−cはLYVE−1抗体による染色、そして、図2−dはvWf抗体を用いた免疫染色図である。
図2−e〜mは、ヒトの各種正常組織のLYVE−1抗体を用いた免疫染色図である。図2−eは食道、図2−fは結腸、図2−gは図2−fのLYVE−1陽性管腔部の強拡大像、図2−hは心臓、図2−iは肺、図2−jは腎臓、図2−kは前立腺、図2−1は卵巣、そして図2−mは膵臓組織の染色結果である。
図2−n〜qは、ヒトの各種正常組織の中で洞構造を持つ主な臓器の染色図である。図2−nでは肝臓の類洞内皮細胞に認められるLYVE−1発現を認め、図2−oはその強拡大像である。図2−pでは脾臓の脾洞の内皮細胞に、また、図2−qではリンパ節内のりンパ管の内皮細胞及び網状線維に認められるLYVE−1発現を示したものである。
図2−r〜xは、ヒト悪性腫瘍の病変部組織の染色図である。そのうち、図2−r〜tは、食道癌の組織に関するものであり、図2−rにおいては食道癌(扁平上皮癌)の腫瘍細胞塊がLYVE−1陽性管腔内に認められ、リンパ管侵襲が示されている。また、図2−sはその部分の強拡大像である。図2−tは図2−rの連続切片を用いたvWf抗体の染色図である。
図2−u及びvは、肺癌の組織に関するものである。図2−uにおいては、肺気管支周囲結合織内のLYVE−1陽性管腔内に肺癌(腺癌)の腫瘍細胞塊を示したものである。図2−vは図2−uの連続切片を用いたvWf抗体の染色図である。
図2−w及びxは、ヒトの心臓に発生した血管肉腫の組織染色図で、図2−wではLYVE−1陽性管腔内に血管肉腫の腫瘍塊を認め、その連続切片を用いてvWf染色を行った図2−xでは、vWf一部陽性の腫瘍細胞がvWf陽性管腔内にも認められる。
図2−r〜xにおいて、LYVE−1陽性内皮細胞をもつリンパ管内への腫瘍細胞浸潤を確認することができる。
各図における拡大率はそれぞれ次の通りである。a;550倍、g及びs;350倍、o;300倍、j、u及びv;250倍、w及びx;225倍、k及びp;185倍、b〜d、f、l、m及びq;130倍、h、i、n、r及びt;110倍、e;50倍。
図3−Aは、心臓筋層内の結合組織内、心外膜脂肪織内に認められるLYVE−1陽性管腔を示したものである。
図3−Bは、肺胸膜下、血管周囲などに認められるLYVE−1陽性管腔を示したものである。
図3−Cは、腎ボウマン嚢周囲、尿細管周囲の問質に認められるLYVE−1陽性管腔を示したものである。
図3−Dは、膵臓の血管周囲を主とする間質に認められるLYVE−1陽性管腔を示したものである。
図3−Eは、扁桃の上皮下のリンパ小節内に認められるLYVE−1陽性管腔を示したものである。
図3−Fは、胸腺の小葉間結合組織内に認められるLYVE−1陽性管腔を示したものである。
図3−Gは、胃の粘膜固有層から漿膜まで全層に認められるLYVE−1陽性管腔を示したものである。特に粘膜筋板沿い、筋層に多く見られる。
図3−Hは、小腸の粘膜固有層の粘膜上皮近傍から漿膜の間質に至る組織の染色図である。
図3−Iは、舌の上皮下の結合織内に認められるLYVE−1陽性管腔を示したものである。
図3−Jは、食道の上皮下の結合織内に認められるLYVE−1陽性管腔を示したものである。
図3−Kは、結腸の粘膜下層から漿膜までの間質に認められるLYVE−1陽性管腔を示したものである。
図3−Lは、膀胱の粘膜下層から漿膜までの間質に認められるLYVE−1陽性管腔を示したものである。

図3−Pは、胎盤絨毛の間質に認められるLYVE−1陽性発現細胞を示したものである。
図3−Qは、肝類洞血管の内皮細胞に認められるLYVE−1発現を示したものである。
図3−Rは、脾洞の内皮細胞に認められるLYVE−1発現を示したものである。
図3−Sは、輸出入リンパ管内皮細胞および細網細胞に認められるLYVE−1発現を示したものである。
図3−Tは、副腎の洞様血管内皮細胞に認められるLYVE−1発現を示したものである。
図3−Uは、食道癌におけるリンパ管侵襲の染色像である。食道の粘膜下層のLYVE−1陽性管腔内に腫瘍塊が認められる。
図3−Vは、胃癌におけるリンパ管侵襲の染色像である。組織のLYVE−1陽性管腔内に腫瘍塊が認められる。
図3−Wは、EMR切除を行った胃癌組織におけるリンパ管侵襲を示した図である。粘膜固有層のLYVE−1陽1生管腔内に腫瘍塊が認められる。
図3−Xは、図3−Wの拡大図です。
図3−Yは、心臓の血管肉腫におけるリンパ管侵襲を示した図である。LYVE−1陽性管腔内に腫瘍塊が認められる。
図3−Zは、リンパ管腫の不規則に増生する管腔の内皮細胞に部分的に認められるLYVE−1発現を示したものである。
図4は、抗体の特異性を確認したウェスタンブロット図である。
レーン1及び3はLYVE−1遺伝子を組み込んだCos−7細胞より得られた試料と反応させた図であり、レーン2及び4はLYVE−1遺伝子を組み込んでいないCos−7細胞より得られた試料と反応させた図である。更に、レーン3及び4は、予め過剰量の本発明のペプチドフラグメントを反応させておいたLYVE−1抗体を用いて反応させたものである。
レーン1で発色が認められレーン3で発色が認められないことから、本発明の抗体はLYVE−1タンパク質に特異的であることがわかる。
配列番号1に記載のアミノ酸配列を有する抗原ペプチドフラグメントは、Genosys社製ABcus7自動ペプチド合成装置を用いて固相法により合成した。その際、N末端にリンカーとなるシステインを付加し、リンカーを付加した抗原ペプチドフラグメント30mgを得た。合成後、抗原ペプチドフラグメントを高速液体クロマトグラフィーにて精製した。
得られた抗原ペプチドフラグメントの確認は次のようにして行った。すなわち、抗原ペプチドフラグメントをベンジル化し、0.1%トリフルオロ酢酸を加えた後、ABI社製VoyagerSystem1162質量分析器にて測定した。測定にあたっては、標準コントロールサンプルを測定し、その理論分子量の差異を補正した後に、合成したペプチドフラグメントを測定し、理論値と一致することを確認した(図1)。
合成した抗原ペプチドフラグメントの抗原性を高めるために、抗原ペプチドフラグメントにリンカー及びスペーサーを介してヘモシアニン(KLH)を結合させた。まず、スペーサーとKLHの結合は次のようにして行った。KLH16mgを10mM燐酸ナトリウムバッファー(pH7.2)1mlに溶解し、これにMBS2.8mgのジメチルホルムアミド1ml溶液を加える。室温中で30分撹拌後、溶液を16,000rpmで遠心し、その上清をセファデックスG25によるカラムクロマトグラフィーにて精製し、KLH−MBS結合物質を得た(含有量は1.8mg/ml)。次に、1mgの上記抗原ペプチドフラグメントとKLH−MBS結合物質(KLH1mg相当)および1/2体積の0.2M燐酸ナトリウムバッファーを混合し、窒素ガス存在下で室温にて3時間反応することにより、抗原を得た。
実施例2(LYVE−1抗体の調製)
抗原(抗原ペプチド100μg相当)を、フロイントの完全アジュバントと共にウサギ背部皮下に免疫した。
以後、2週間に一度の割合で、抗原ペプチド100μgに相当する量の抗原をフロイント不完全アジュバントと共に同様に免疫し、合計5回免疫した。第2回目の免疫以降、抗原及びセルロースアセテート膜を使用したドットブロット法にて抗体価上昇の確認を行った。抗体価の上昇を確認した後、全採血を行い、抗血清を分離した。
次に、抗体の精製を以下のようにして行った。まず、Affige10(BIO−RAD社)に抗原ペプチドを結合させて、精製用アフィニティーカラムを作製した。そして、非動化血清10mlを同量のPBS(燐酸緩衝食塩水)で希釈し、0.45μmのフィルターに通した後、アフィニティーカラムに添加した。カラムを約50mlのPBSにて洗浄した後、5mlの50mMクエン酸バッファーで抗体を溶出した。溶出した抗体に2Mトリスバッファー(pH9.5)を少量加え、溶出抗体溶液のpHを7.4に補正した。280nMの波長の吸光度を測定することにより、抗体濃度を計測した。
実施例3(LYVE−1抗体の特異性の検討)
LYVE−1のタンパク質翻訳域の全長遺伝子を、真核細胞発現プラスミドベクターに組み込み、リポソーム試薬を使用して、このプラスミドベクターをCos−7細胞に導入し、一過性にLYVE−1タンパク質を発現させた。導入24時間後にCos−7細胞をPBSで洗い、Lysisバッファー(20mMトリス塩酸バッファー、140mM 食塩水、0.1% Nonidet P−40、1μg/mL 蛋白分解阻害剤(ペプスタチン、リューペプチン及びアプロチニン)の混合液)により溶解した。この混合液を15,000rpmにて遠心した後、その上清をPAGE(ポリアクリルアミドゲル電気泳動)にて分画した後、ナイロンフィルターに転写し、フィルター試料とした。このフィルター試料の一部をLYVE−1抗体と反応させた。
一方、抗体を抗原ペプチドと4℃にて一晩反応させ、抗体吸収試液とし、同様にフィルター試料の一部を抗体吸収試液と反応させた。その後、ペルオキシダーゼ標識抗ウサギIgG山羊血清と反応させた後、化学発光検出試薬(ECL Western blottingキット)を反応させた。検出は、フルオロイメージアナライザーLAS−1000(富士フィルム)を用いて行った。その結果、LYVE−1抗体と反応させたフィルター試料にはLYVE−1とLYVE−1抗体とが反応した特異的なバンドが確認されたが、抗体吸収試液と反応させたフィルター試料ではそのバンドは確認されなかった。このことより、本抗体がLYVE−1特異的な抗体であることが証明された。
実施例4(LYVE−1抗体を用いた組織染色)
下記表1に示すヒトの各種正常及び病理学的組織のパラフィン固定切片を使用し、LYVE−1の局在について検索した。
各切片を脱パラフィン後にガラススライド上に置き、ガラススライド上の組織とLYVE−1抗体を室温にて30分反応させた。これをPBSで洗浄した後、ビオチン標識抗ウサギIgG山羊血清と室温にて30分反応させた。
反応物をPBSで洗浄し、さらにストレプトアビジンペルオキシダーゼ結合試薬と室温にて30分反応させた。PBS洗浄後3,3−ジアミノベンジジン塩酸を基質として発色を行った。各組織の染色像を図2に示す。
また、同一組織におけるリンパ管内皮細胞の染色と血管内皮細胞の染色を比較するため、von Wilebrand factor(vWF)による血管内皮細胞の染色も同様にして行った。
その結果、ヒトの各種正常及び病理学的組織において、LYVE−1抗体によりリンパ管内皮細胞は染色されたが、血管内皮細胞は染色されなかった。また、vWFにより血管内皮細胞は染色されたが、リンパ管内皮細胞は染色されなかった。各組織の染色像を図3に示す。
上記実験の結果から、LYVE−1抗体はリンパ管内皮細胞を特異的に認識する抗体であること、及びLYVE−1抗体はリンパ管内の癌細胞浸潤部の特定に有用であることが証明された。
本発明により、LYVE−1のペプチドフラグメント及びそのペプチドに対する抗体が提供される。本発明の抗体を利用して、生体組織中のリンパ管のみを選択的に染色することができる。特に、本発明の抗体は、生体組織における腫瘍によるリンパ管の破壊性浸潤性増殖についての病理学的所見を得るための使用において有用であり、ひいては、本発明の抗体を利用して腫瘍の悪性度を診断するための指標を得ることができ、癌患者の治療及び予後に役立つものとなる。
さらに、本発明の抗体は、ヒトリンパ球の分離、精製を利用したリンパ管の新生治療に用いることができる。すなわち、本発明の抗体を用いて、例えば細胞分離機器(セルソーター)、磁気細胞分離法等によりリンパ管細胞及びその前駆体細胞を分取し、これを、例えば、リンパ管浮腫の軽減を目的としたリンパ管新生治療等に用いることができる。(Lymphangiogenesis in tumors:What do we know?,Microscopy Research and Technique Volume 60,Issue 2,171−180,February 2003;Lymphangiogenesis and Tumor Metastasis,Myth or Reality?,Clinical Cancer Research Vol.7,462−468,March 2001;Expression of the Fms−Like Tyrosine Kinase 4 Gene Becomes Restricted to Lymphatic Endothelium During Development,Proceedings of the National Academy of Sciences,Vol 92,3566−3570,April 1995)
Claims (11)
- 配列番号1に記載のアミノ酸配列からなるペプチドフラグメント又はそのC末端若しくはN末端にリンカーとして一つのアミノ酸が付加されたペプチドフラグメント。
- 請求の範囲第1項に記載のペプチドフラグメントに対する抗体。
- ヒトリンパ管特異的ヒアルロナン受容体−1(LYVE−1)に特異的である、請求の範囲第2項に記載の抗体。
- 配列番号1に記載のアミノ酸配列において、1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなり、かつ請求の範囲第2項又は第3項に記載の抗体によって認識されるペプチドフラグメント又はそのC末端若しくはN末端にリンカーとして一つのアミノ酸が付加されたペプチドフラグメント。
- 請求の範囲第2項又は第3項に記載の抗体を調製するための、請求の範囲第1項又は第4項に記載のペプチドフラグメントの使用。
- 請求の範囲第2項又は第3項に記載の抗体を含むことを特徴とする、ヒトリンパ管免疫染色用組成物。
- 請求の範囲第2項又は第3項に記載の抗体を含むことを特徴とする、ヒトリンパ管免疫染色用試薬キット。
- 採取したヒト組織におけるリンパ管を、請求の範囲第2項又は第3項に記載の抗体を用いて特異的に免疫染色する方法。
- 悪性腫瘍の悪性度の鑑別方法であって、次のステップ;
a)採取したヒト組織におけるリンパ管を、請求の範囲第2項又は第3項に記載の抗体を用いて特異的に免疫染色するステップ、
b)ステップa)で染色した染色像における悪性腫瘍の浸潤度を測定するステップ、
c)ステップb)に基づいて悪性腫瘍の悪性度を鑑別するステップ、
からなる方法。 - 請求の範囲第2項又は第3項に記載の抗体を用いて、ヒトリンパ球を分離及び/又は精製する方法。
- ヒトリンパ管の新生治療における、請求の範囲第2項又は第3項に記載の抗体の使用。
