Abstract

PURPOSE:To enable accurate determination of a substance to be inspected in a sample to be inspected with a handy operation, by using a liposome containing phospholipid, cholesterol and a crosslinking agent as selected from phosphatidic acid or the like to remove a complement component from the sample being inspected effectively. CONSTITUTION:First, a liposome (pretreatment reagent) containing phospholipid, cholesterol and a crosslinking agent as selected from phosphatidic acid or the like is added to a sample to be inspected and allowed to stand at a room temperature for about 10min. As a result, an antigen or an antibody in the liposome reagent reacts with a substance to be inspected and a complement is activated by an antigen-antibody reaction to release labelled substance from the liposome in proportion to the level of the antigen-antibody reaction. Then, a determination is performed by an analysis method corresponding to the labelled substance and, for example, the amount of the substance being inspected in the sample can be measured according to a calibration curve prepared beforehand. This can remove a complement component in the substance being inspected effectively thereby enabling accurate determination of the substance being inspected.
