Abstract

PURPOSE:To obtain the title substance of high purity in high yield, which can be expected to be used as a medicine, by acyl rearrangement of 7-acyl-7- deacetylforskolin derivative only with heat. CONSTITUTION:The compound of formula I (R<1> is hydroxyl which may be esterified; R<2> is acyl; R<3> is 2-3C hydrocarbon group) is heated at 150-250 deg.C in case of no solvent for 15-60min or at 70 deg.C to the boiling point of the solvent for 3-8hr to give the compound of formula II. Direct heating is preferably conducted under fusing, any solvents whether they dissolve the compound of formula I or not can be used, when a solvent is employed, and the solvent is preferably anhydrous, in order to avoid hydrolysis of the acyl group. The solvent is for example DMF, DMSO or acetonitrile. When R<2> is dimethylaminopropionyl, the treatment of the reaction mixture with the amine corresponding the amine component inhabits the formation of by-products.
