Abstract

PURPOSE:To store an useful substance of plant bodies in a liquid, remove only noxious browned substances and enable cultivation without damaging the plant bodies, by using an adsorbent attaining adsorption equilibrium with a culture fluid. CONSTITUTION:Active carbon is filled in an adsorption column 15 having 20mm inside diameter and 65mm length and an MS culture medium is fed thereto at 5ml/min flow rate for 12hr to provide the action carbon in an adsorption equilibrium state. Calluses 2 of the genus Cymbidium are then placed in a culture column 11 equipped with a perforated plate having 60mm inside diameter and 150mm length and an MS culture medium is passed through the adsorption column 15 for a given time and then sprayed on the calluses. The culture fluid is subsequently circulated through this loop and continuous operation is carried out. An illuminator 19 is lit for 12hr and put out for 12hr and temperature is kept at 23 deg.C to carry out cultivation for about 90 days. Browning of the culture fluid is not observed and green calluses are propagated.
