Abstract

PURPOSE:To achieve a higher measuring sensitivity, by getting an amide type phospholipid contained in phospholipid as component of liposome to improve stability of holding a labeled substance in the liposome. CONSTITUTION:A labeled substance is sealed into a liposome having phospholipid containing an amide type phospholipid and cholesterol as requisite component and an antigen or an antibody reacting specifically with a substance to be inspected is immobilized outside the liposome through a crosslinking agent to obtain a liposome reagent. When the liposome reagent thus obtained, a sample containing the substance being inspected and a complement are mixed in a proper buffer, the complement is activated by an antigen-antibody reaction and the labeled substance is released from the inside of the liposome in proportion to the level of the antigen-antibody reaction. Then, the amount of the substance being inspected in the sample is measured by an analysis method corresponding to the labeled substance. This improves the stability of holding the labeled substance thereby to achieve a higher measuring sensitivity.
