Abstract
PURPOSE:To obtain a luciferase gene originated from Luciola cruciata and useful for the quick and efficient production of luciferase useful as an enzyme for the determination of ATP, by using gene recombination technique. CONSTITUTION:m-RNA is prepared from the tail of Luciola cruciata and a c-DNA is synthesized from the m-RNA. The c-DNA is integrated into a vector DNA (e.g. plasmid pUC) to obtain various recombinant DNAs. Various recombinant strains are prepared by transforming E.coli, etc., with said DNAs. Microbial cell containing said recombinant is selected from the strains by a colony hybridization method using a probe consisting of a <32>P-labeled DNA containing a gene coding luciferase originated from Photinus pyraris. The plasmid DNA is digested with a restriction enzyme to obtain a DNA containing a c-DNA coding luciferase.
