Abstract
PURPOSE:To readily obtain an alpha-amylase useful for producing industrial starch saccharides, etc., in high yield at a low cost, by cultivating a microorganism of the genus Thermus capable of producing the alpha-amylase. CONSTITUTION:A strain FERM P-8881 of the Thermus having the following mycological properties. Shape; bacillus. Gram staining property; negative. Growth pH; 5.0-8.5, etc., is aerobically cultivated in a culture medium containing pullulan, soybean cake, etc., at about 60 deg.C for 72 hr to provide a culture, which is then purified by a method for filtering, salting out, ultrafiltering, etc., to afford the aimed alpha-amylase having the following physico-chemical properties. Action; specifically hydrolyzing straight-chainlike structure of alpha-1,4- glucopyranoside bonds in the amylopectin, amylose, starch, etc., and partial hydrolyzates thereof and forming alpha-type ionomer of maltose, etc. Substrate specificity; hydrolyzing straight-chainlike malstooligosaccharides of alpha-1,4- glucopyranoside bonds having a higher polymerization degree than that of maltotetraose. Optimum pH; 5.5-6.0. Stable pH; pH 5-9 at 50 deg.C for 30min. Optimum action temperature; about 70 deg.C. Molecular weight; 135000+ or -5000, etc.
