NEW MATERIAL:The amylase having the following physical and chemical properties. Action and substrate specificity, acting to soluble starch or starch-based sugar and pullulan and scarcely acting to raw starch, dextran, maltotetraose, maltose and isomaltose; optimum pH and stable pH; optimum pH to soluble starch is 7.0-8.5 and the stable pH is 5.0-9.0, optimum pH to pullulan is 7.0 and the stable pH is 5.0-8.0; optimum temperature and heat-stability, optimum active temperature is 50 deg.C, stable up to 40 deg.C, inactivated at 60 deg.C in 30min; isoelectric point, 4.13 by electrophoresis; molecular weight, 218,000 by SDS-PAGE process. USE:Production of malto-oligosaccharide. PREPARATION:The objective amylase having pullulanase activity can be produced e.g. by culturing Bacillus circulans F-2 (FERM P-7361) belonging to Bacillus genus in a medium, separating the bacterial cells from the cultured product, collecting a pullulanase-active fraction from the supernatant liquid and subjecting the fraction to extraction, salting-out, ion exchange treatment and purification by hydrophobic chromatography.
