Abstract

PURPOSE: To purify a ricin-specific enzyme in high efficiency and purity, by using an affinity chromatography with a specific adsorbent.
CONSTITUTION: It has been found that a ricinal of formula is an extremely specific inhibitor against a ricin-specific enzyme and the bond (reaction) between the enzyme and its inhibitor is reversible under a specific condition. Taking advantage of the properties, ricinal is bonded to an activated carrier through the α-amino group to obtain an adsorbent and a ricin-specific enzyme is extracted and purified by affinity chromatography using said adsorbent. A spacer consisting of an amino acid residue or peptide residue is preferably inserted between the activated carrier and ricinal.
