Description
本発明は少なくとも1個の遊離ホルミル基を有
する高分子化合物、特にホルミル基含有ポリサツ
カライド誘導体とアミノ基含有2官能配位子化合
物が結合して成る、少なくとも1個の遊離ホルミ
ル基を有する高分子化合物に関する。
本発明の高分子化合物は文献未載の新規物質で
あり、特定臓器の描出、動定疾患の検出、生理活
性物質の動態検査、疾病の治療などの核医学的用
途に適した、安定な放射性金属元素標識つき放射
性薬品を提供することが出来るものである。
特定臓器の描出、特定疾患の検出、動態検査な
どを目的とした非侵襲的核医学診断のために、従
来ヨード−131で標識された生理活性物質が汎用
されてきた。たとえば、血液循環系の描出や動態
検査に用いられるヨード−131標識ヒト血清アル
ブミン、血栓の検出に用いられるヨード−131標
識フイブリノーゲンなどが挙げられる。しかしな
がら、ヨード−131は半減期が約8日と長く、か
つ、核医学診断に有用なガンマー線の他に、ベー
タ線を放出するため、被検者に多量の放射線被曝
を与える欠点がある。
そこで核医学診断により適した物理的特性を有
する放射性金属を、他の方法により生理活性物質
に導入し、有用な放射性診断剤を得ようとする試
みが続けられている。たとえば、生理活性物質に
直接、放射性金属塩を作用させる標識法で得られ
るものとして、テクネチウム−99m標識ヒト血清
アルブミン、インジウム−111標識ブレオマイシ
ンなどが知られている。さらにジエチレントリア
ミン五酢酸(DTPA)、3−オキソブチラールビ
ス(N−メチルチオセミカルバゾン)カルボン
酸、デフエロキサミンなどの2官能配位子化合物
の各種金属に対する強いキレート形成能と、それ
らの化合物末端のアミノ基およびカルボキシル基
の種々の生理活性物質に対する反応性に基づい
て、これら2官能配位子化合物を介して放射性金
属および生理活性物質を結合させる方法も提案さ
れている。
これらの方法で得られた標識化合物は比較的安
定であり、しかも生理活性物質の活性を保持して
いるので、核医学診断目的に非常に興味ある薬剤
である。しかしながら、これらの方法によつて得
られた放射性診断剤は、分子量の大きい生理活性
物質、たとえば血栓診断やガン診断に使用される
それぞれ分子量約34万のフイブリノーゲンや分子
量約16万のIgGを用いた場合、診断に必要な高比
放射能のものが得られない欠点がある。
本発明者らは、この欠点を解決すべく種々研究
を重ねた結果、先にジアルデヒドデンプンにアミ
ノ基含有2官能配位子化合物とアミノ基含有生理
活性物質が結合した高分子化合物を開発すること
に成功した(特開昭59−105002号明細書および特
開昭59−106425号明細書参照)。この高分子化合
物は、1分子当たり多数の配位子をもつものであ
り、このことはとりも直さず1分子当たりに結合
する放射性金属イオンの数が従来の2官能配位子
化合物に比して格段に多いことを意味する。そし
てこの高分子化合物を使用することにより、生理
活性物質の変性および活性低下を来すことなく高
比放射能の放射性診断剤が得られる事実が見出だ
された。
上記の知見に基づいて更に研究を進めた結果、
ジアルデヒドデンプンに代えて他のホルミル基含
有ポリサツカライド誘導体を使用しても同様に生
理活性物質の変性や活性低下を起こすことなく高
比放射能の放射性医薬品が得られる事実が見出だ
された。一般にジアルデヒドデンプンは分子量分
布が広く、網状構造を有するので、その繰り返し
構造の数ほどには2官能配位子化合物が効果的に
結合しにくい傾向が認められるが、ジアルデヒド
アミロースのように分子量分布が狭くかつ直鎖構
造を有するものを使用すれば多数の2官能配位子
化合物を効果的に結合することが出来る。
本発明は以上の知見に基づいて完成されたもの
であつて、その要旨は分子中に少なくとも3個の
ホルミル基を有するポリサツカライド誘導体(た
だし、ジアルデヒドデンプンを除く。)()とア
ミノ基含有2官能配位子化合物()が前者1分
子当たり後者が少なくとも2分子の割合において
メチレンイミノ結合(−CH=N−)またはメチ
レンアミン結合(−CH2NH−)を介して結合し
て成る少なくとも1個の遊離ホルミル基を有する
高分子化合物()、特に(a)くり返し単位2〜
1000のジアルデヒドアミロースまたはジアルデヒ
ドデキストランと(b)デフエロキサミンまたは3−
オキソブチラールビス(N−メチルチオセミカル
バゾン)カルボン酸・ジアミン縮合体が前者1分
子当たり後者少なくとも2分子の割合においてメ
チレンイミン結合(−CH=N−)またはメチレ
ンアミン結合(−CH2NH−)を介して結合して
成る少なくとも1個の遊離ホルミル基を有する高
分子化合物に存する。
本発明の目的とする上記高分子化合物()は
ポリサツカライド誘導体()と2官能配位子化
合物()が結合して構成されたものである。
ポリサツカライド誘導体()は分子中に少な
くとも3個のホルミル基を持つことが必要であ
り、ホルミル基の数が多いほど好ましい。それら
のホルミル基のうち少なくとも2個は2官能配位
子化合物()との結合に役立つものであり、他
の少なくとも1個は高分子化合物()において
遊離のまま残留し、後に高分子化合物()に生
理活性物質()を結合させるのに役立つ。
ポリサツカライド誘導体()としては、たと
えば適宜に置換されていることもあるポリサツカ
ライドを酸化剤(たとえば過ヨード酸ナトリウ
ム)で処理して得られる、原則としてサツカライ
ド単位毎に1個または2個のホルミル基を有する
ものが使用されうる。ポリサツカライドとして
は、オリコサツカライトでもよいが、本発明の目
的から理解されるようにペント−サン、ヘキソ−
サン、ポリグルコサミン、ポリウロン酸、グリコ
サミノグリカン、グリコウロノグリカン、ヘテロ
ヘキソ−サンなど高次のポリサツカライドが好ま
しい。具体例としては、アミロース、アミロペク
チン、デキストラン、セルロース、イヌリン、ペ
クチン酸、プルランなどが挙げられ、それらの混
合物や脱水縮合物であつてもよい。一般にサツカ
ライド単位が3000以下、特に1000以下のものが望
ましい。
2官能配位子化合物()としては、放射性金
属元素()に対し強固なキレート結合を形成
し、かつ比較的緩和な条件下でポリサツカライド
誘導体()のホルミル基と反応し得るアミノ基
を有するものが使用される。このような2官能配
位子化合物()の具体としてはデフエロキサミ
ン(メルク・インデツクス、第9版、374頁
(1976年))、式:
(式中、R1およびR2それぞれ水素、C1〜C3アル
キルまたはフエニルを表す。)で表される3−ア
ミノメチレン−2,4−ペンタンジオン−ビス
(チオセミカルバゾン)誘導体(ヨーロツパ特許
出願第54920号明細出)、式:
(式中、R3およびR4はそれぞれ水素またはC1〜
C3アルキル、nは0〜3の整数を表す。)で表さ
れる1−(p−アミノアルキル)フエニルプロパ
ン−1,2−ジオン−ビス(チオセミカルバゾ
ン)誘導体(オーストライア特許第533722号明細
書)などが挙げられる。
そのもの自体はアミノ基を有していなくても容
易にアミノ基またはアミノ基含有構造を形成し得
る基または構造を有している場合は、放射性金属
元素を捕捉する性質を有する限り、このものもま
たアミノ基またはアミノ基含有構造を形成せしめ
たうえで、2官能配位子化合物()として使用
し得る。たとえば、カルボキシル基を有するもの
は、これにアルキレンジアミンを反応させること
によつて容易にアミノ基を導入することが出来、
本発明において2官能配位子化合物()の1種
として使用することが出来る。このような2官能
配位子化合物()の具体例としては、ジエチレ
ントリアミン五酢酸(DTPA)、エチレンジアミ
ン四酢酸(EDTA)、ジメルカプトアセチルエチ
レンジアミン(フリツツバーク(Fritzberg)
ら:ジヤーナル・オブ・ヌクレアー・メデイスン

C1〜C3アルキル)で表わされる2−プロピオン
アルデヒド−ビス(チオセミカルバゾン)誘導体
(アメリカ特許第4287362号明細書)などが挙げら
れる。
高分子化合物()を製造するには、たとえば
ポリサツカライド誘導体()と2官能配位子化
合物()を縮合させて前者のホルミル基と後者
のアミノ基の間でメチレンイミン結合を形成せし
め、必要に応じこのメチレンイミン結合を還元し
てメチレンアミン結合に変換すればよい。上記縮
合反応はホルミル基とアミノ基を縮合させるため
に採用される自体常套の手段で行えばよく、上記
還元反応もメチレンイミン結合をメチレンアミン
結合に変換するてめに採用されている自体常套の
手段、たとえば、は水素化ホウ素ナトリウムのよ
うな金属水素化物を使用することにより行なわれ
る。上記還元反応に際し、サツカライド単位中の
ホルミル基が一部還元された化合物が副生するこ
とがあるが、これによつて本発明の目的が妨げら
れることはない。反応試剤、反応条件などの相違
によりポリサツカライド誘導体()1分子に結
合する2官能配位子化合物()の分子数は異な
るが、一般には5またはそれ以上、特に10または
それ以上が好ましい。ただし、この反応で得られ
た高分子化合物()のポリサツカライ誘導体
()部分における少なくとも1個のホルミル基
は生理活性物質()との結合のため遊離のまま
残留すべきである。
ポリサツカライド誘導体()としてアミロー
ス由来のものを使用する場合を例に挙げて高分子
化合物()の構造を式で示せば次の通りであ
る:
(式中、Xは2官能配位子化合物()からアミ
ノ基を除去した残基、Yは−CHOまたは−
CH2OH、Pは2〜1000の整数、q、r、sおよ
びtはそれぞれ0〜1000の整数を表す。ただし、
q+r+s+tは2〜1000の整数である。)。
なお、上式においてジアルデヒドアミロース
(a)はアミロースを原料とし、これを過ヨー
ド酸のような酸化剤で部分的あるいは全体的に酸
化することにより得られる鎖状高分子物質であつ
て、それ自体市販されている。そのサツカライド
単位は通常2〜1000であり、特に2〜500が好ま
しい。
ここに生成した高分子化合物()は、必要に
応じ、高分子物質に適用されるカラムクロマトグ
ラフイー、ゲルろ過法、透析法などの常套の精製
法により精製されてもよい。
本発明の高分子化合物()は、その分子中に
少なくとも1個の遊離ホルミル基が存在するか
ら、そのホルミル基を介してアミノ基を有する生
理活性物質()と結合し、放射性医薬品調製用
キヤリヤーを提供することが出来る。ここで言う
生理活性物質とは、適当な器官または組織あるい
は特定の病巣に蓄積するか、特定の生理状態に対
応して特異な挙動を示す物質を意味し、生体内に
おけるこのような物質の挙動を追跡することによ
つて診断上有用な情報を得ることが出来るもので
ある。生理活性物質は一般にそれ自体でアミノ基
を有しているものが多いが、そのような生理活性
物質はもとより、それ自体ではアミノ基を有して
いないものであつても、これに適宜の方法でアミ
ノ基またはアミノ基含有構造を導入したものを使
用することが出来る。ホルミル基とアミノ基の間
の反応は比較的緩和な条件下で進行するから、本
発明において有利に利用することが出来る。
生理活性物質()の具体例としては、血液蛋
白質(たとえばヒト血清アルブミン、フイブリノ
ーゲン)、酵素(たとえばウロキナーゼ、ストレ
プトキナーゼ)、ホルモン(たとえば甲状腺刺激
ホルモン、副甲状腺ホルモン)、免疫抗体(たと
えばIgGおよびその断片のF(ab)2′、Fab′、
Fab)、モノクローナル抗体、抗生物質(たとえ
ばブレオマイシン、カナマイシン)、糖類、脂肪
酸、アミノ酸などが挙げられる。
本発明の高分子化合物()に対して生理活性
物質()を結合せしめるには、たとえば高分子
化合物()と生理活性物質()を縮合させて
前者のホルミル基と後者のアミノ基の間でメチレ
ンイミン結合を形成せしめ、必要に応じこのメチ
レンイミン結合を還元してメチレンアミン結合に
変換すればよい。上記縮合反応も還元反応も共に
自体常套の方法で行えばよい。高分子化合物
()の1分子当たり導入される生理活性物質の
分子数は、反応試剤、反応条件などによつて異な
るが、通常は10またはそれ以下、特に3またはそ
れ以下が好ましい。
ここに生成した高分子化合物()と生理活性
物質()の結合体は必要に応じ高分子物質適用
されるカラムクロマトグラフイー、ゲルろ過法、
透析法などの常套の精製法により精製されてもよ
い。
この高分子化合物()−生理活性物質()
結合体は、前記のように放射性医薬品調製用キヤ
リヤーとして有用なものである。すなわち、該結
合体には高分子化合物()の部分に2官能配位
子化合物()が複数個存在しており、これによ
つて複数個の放射性金属元素()を捕捉するこ
とが可能であり、このように複数個の放射性金属
元素()を捕捉せしめた結合体はそれ自体放射
性医薬品として使用されうる。
ここに放射性金属元素()としては、放射能
を有する金属元素であつて、核医学的診断や治療
に適した物理的または化学的特性を有し、しかも
2官能配位子化合物()の配位子構造により容
易に捕捉されうるものが使用される。その具体例
としては、ガリウム−67、ガリウム−68、タリウ
ム−201、インジウム−111、テクネチウム−99m
などがあり、これらは通常、塩、特に水溶性塩の
形で使用され、水性媒体中において高分子化合物
()−生理活性物質()結合体と接触せしめて
その標識化を行う。ただし、放射性金属元素
()が安定なキレート錯体を形成しうる原子価
状態にある場合には(たとえばカリウム−67、イ
ンジウム−111)、反応系に他の試剤を存在せしめ
る必要はないが、安定なキレート錯体を形成する
ために原子価状態を変化せしめる必要がある場合
には(たとえばテクネチウム−99m)、反応系に
還元剤または酸化剤を存在せしめる必要があろ
う。還元剤としては、2価のスズ塩(たとえばハ
ロゲン化スズ、硫酸スズ、硝酸スズ、酢酸スズ、
クエン酸スズ)が挙げられる。
たとえば、放射性金属元素()としてテクネ
チウム−99mを使用する場合、高分子化合物
()−生理活性物質()結合体を水性媒体中還
元剤としての第1スズ塩の存在下パーテクネテー
トの形のテクネチウム−99mと処理することによ
つてテクネチウム−99m標識高分子化合物()
−生理活性物質()結合体を調製することがで
きる。上記調製に際し、各試剤の混合順序につい
て格別の制限はないが、通常、水性媒質中で最初
に第1スズ塩とパーテクネテートを混合すること
は避けた方が望ましい。第1スズ塩はパーテクネ
テートを充分に還元出来る量で使用するのが好ま
しい。
このようにして得られた標識高分子化合物
()−生理活性物質()結合体が放射性診断剤
として有用であるためには、診断を可能とする充
分な放射能と放射能濃度を有することが必要であ
る。たとえば放射性金属元素としてテクネチウム
−99mを使用した場合、投与時に約0.5〜5.0ml当
たり0.1〜50mCiの放射能濃度を有することが望
ましい。また、このような標識高分子化合物
()−生理活性物質()結合体は調製後直ちに
投与されてもよいが、好ましくは調製後適当時間
保存に耐えうる程度の安定性を有することが望ま
しい。なおまた、標識高分子化合物()−生理
活性物質()結合体には、必要に応じPH調節剤
(たとえば酸、塩基、緩衝剤)、安定剤(たとえば
アスコルビン酸)、等張剤(たとえば塩化ナトリ
ウム)などが配合されてもよい。
本発明高分子化合物()を用いて得られた放
射性医薬品の一例をポリサツカライド誘導体
()としてジアルデヒドアミロース、2官能配
位子化合物()としてデフエロキサシン、生理
活性物質()としてヒト血清アルブミン、放射
性金属元素()としてガリウム−67を使用する
場合につき説明すれば次の通りである。
まず、ジアルデヒドアミロースをデフエロキサ
ミンと縮合させた後、必要に応じて還元すること
によりジアルデヒドアミロース−デフエロキサミ
ン結合体を調製し、この結合体にヒト血清アルブ
ミンを縮合させた後、必要に応じて還元すること
によりヒト血清アルブミン−ジアルデヒドアミロ
ース−デフエロキサミン結合体を得る。この結合
体と3価のガリウムイオンの形でガリウム−67を
含む水溶液を接触させることにより、安定で高比
放射能のガリウム−67標識ヒト血清アルブミン−
ジアルデヒドアミロース−デフエロキサミン結合
体を得る。
この標識結合体の電気泳動的挙動は、ヒト血清
アルブミンの挙動と全く同じである。また、この
標識結合体のラツト体内分布は従来のヨード−
131標識ヒト血清アルブミンとほぼ同じである。
血中濃度の半減期は約30時間で、ヨード−131標
識ヒド血清アルブミンの血中濃度の半減期(5.7
時間)よりも充分長い。すなわち、本発明による
標識結合体は、心プール像の描出に有用であるこ
とが理解出来る。
本発明による上記ヒト血清アルブミン−ジアル
デヒドロアミロース−デフエロキサミン結合体
(試料1)と従来法によりヒト血清アルブミンと
デフエロキサンを直接結合させたもの(試料2)
とのガリウム−67、1mCiに対する標識能を比較
したところ、次のような結果が得られた。

Based on the reactivity of the carboxyl group to various physiologically active substances, methods for bonding radioactive metals and physiologically active substances via these bifunctional ligand compounds have also been proposed. The labeled compounds obtained by these methods are relatively stable and retain the activity of physiologically active substances, so they are very interesting drugs for nuclear medicine diagnostic purposes. 実施例 3
ジアルデヒドアミロース−3−オキソブチラー
ルビス(N−メチルチオセミカルバゾン)カル
ボン酸アミン誘導体の結合体(縮合還元体)の
製造:−
3−オキソブチラールビス(N−メチルチオセ
ミカルバゾン)カルボン酸(以下、KTSと略
す。)(132mg)を無水ジオキサン(5ml)に溶解
し、10℃付近に冷却したのち、トリ−n−ブチル
アミン(0.12ml)、更にイソブチルクロロホルメ
イト(64μ)を加え、同温度で約50分間撹拌し
て、混合酸無水物を得た。
別にN−tert−ブチルオキシカルボニル−1,
6−ヘキサンジアミン(104mg)を無水ジオキサ
ン(2ml)に溶解した溶液を調製し、この溶液を
混合酸無水物溶液に加え、10℃付近で約15時間撹
拌し、KTS−N−tert−ブチルオキシカルボニル
−1,6−ヘキサンジアミン縮合体を得た。この
縮合体溶液に濃塩酸(1〜2滴)を加えてPH2に
下げることによりN−tert−ブチルオキシカルボ
ニル基を離脱せしめ、KTS−ヘキサンジアミン
縮合体の溶液を得た。これをDAA(83mg)のジメ
チルスルホキシド(5ml)溶液に加えた後、水素
化ホウ素ナトリウム(17.2mg)を加え、室温で約
3時間反応させ、DAA−ヘキサンジアミン−
KTS結合体の溶液を得た。
上記溶液を通常の透析チユーブに入れ、常法に
より30時間透析することにより、未反応試薬を除
去し、さらに凍結乾燥することにより、目的とす
る高分子化合物を得た。
ヘキサンジアミン−KTS結合体の最大吸収は、
波長334nmに存在し、そのEmaxは4.37×104であ
ることを確認した。上記高分子化合物を水に溶解
して3mg/mlの濃度とし、水を対照として334n
mで吸光度を測定した。その結果、DAA1分子あ
たりKTS10.5個が結合していることが確認され
た。従つて、上記高分子化合物の平均分子量は約
7000と算出される。
実施例 4
ジアルデヒドデキストラン−デフエロキサミン
結合体(縮合還元体)の製造:−
DFO(2.8g)を水(30ml)に溶解し、この溶液
に等モル量(432mg)のトリエチルアミンを加え、
室温にて10分間撹拌した。これをジアルデヒドデ
キストラン(以下、DADと略す。)(1g;ホル
ミル基含量5.1μmol/mg)の水(40ml)溶液に加
え、15分間室温にて撹拌した。これに水素化ホウ
素ナトリウム(167mg)を加え、約1時間室温で
撹拌しながら還元した。生成したDAD−DFO結
合体を精製するため、透析チユーブに導入し、水
に対して3日間透析した後、下記のゲルクロマト
グラフイーを実施した。

A solution of 6-hexanediamine (104 mg) dissolved in anhydrous dioxane (2 ml) was prepared, and this solution was added to the mixed acid anhydride solution and stirred at around 10°C for about 15 hours. A carbonyl-1,6-hexanediamine condensate was obtained. Concentrated hydrochloric acid (1 to 2 drops) was added to this condensate solution to lower the pH to 2, thereby removing the N-tert-butyloxycarbonyl group to obtain a solution of KTS-hexanediamine condensate. After adding this to a solution of DAA (83 mg) in dimethyl sulfoxide (5 ml), sodium borohydride (17.2 mg) was added, and the mixture was reacted at room temperature for about 3 hours.

The above solution was placed in an ordinary dialysis tube and dialyzed for 30 hours by a conventional method to remove unreacted reagents, and then freeze-dried to obtain the desired polymer compound.
