最近1984年に、ウィルスHTLV−IIIは後天性免疫不全症候群(エイズ
)病の最も可能性の高い原因ウィルスとして現われた(ガロ等、イン ザ マテ
リアル イ〉フォメーシ璽ン ディスクロージャー、ポスト112゜5 (Ga
1lo、 et al、 in the Material Informat

H9細胞ti当にHTLV−mウィルスに感染でせ、そして培養する場合にも、
不朽化されたものが生じる。HTLVp24 コア抗原は、不朽化H9/HTL
V−n[細胞系から分離及び精製されている(ガロ等、米国特許第5erial

総括的方法
本発明において、血清中の自然な抗体は、HTLV−11感染を中和する能力に
よシ評価される。HTLV −1[感染は、HTLV−[1p24 に対する特
異的モノクローナル抗体によってウィルスのコア蛋白質、p24 の表出を追う
ことによシ調べられる。
本発明において、効果はかなりの量のウィルスを効果的に中和する抗体を含む血
清を使用することである。この抗体中和は、全体または部分でろって良い、そし
て中和する抗体価の童の算出は、略述した方法を用いて行なわれても良い。この
方法は、めらゆる種からの血清に適用でき、そしてそれ故にHTLV−11中和
抗体反応を引き出す効果のある潜在的ワクチン製造を評価するために有用である
。最後の段階で、3日以内にウィルスの感染を観察することができない場合には
、血清は完全にウィルスの感染を中和した。血清中の中和抗体がウィルスのレセ
プターへの初期の付着に関係するウィルスのエンベロープの糖蛋白質に結合し、
この様にウィルスの感染作用を阻止すると思われる。
また、抗原−抗体反応並びにウィルスの抗原を中和し保護に有用であるエイズ及
び関連した患者の血清中の抗体の存在に依存するHTLV−flit試験する特
別な目的のために、使用することにも努力した。

HTLV −[1を中和し、それを保護し及び特異的MAB例えば抗p 2a

体を検出する本方法を上記参考文献のどれ本開示していない。
本発明
後天性免疫不全症候群(エイズ)の多くの患者の細胞からヒトT−細胞白血病(
向リンパ性)ウィルスタイプIII(HTLV−[7) の分離は、このウィル
スがこの疾病の病因であるという最初の証拠を提示した。この結論は、大部分の
エイズ及びエイズ関連コンプレックス(ABC)患者の血清中K HTLV−I
IIの特異的抗体の存在を示すそれらの血清疫学的研究を包含する多くの一連の
研究の結果によって強化されてきた。加えて、同性愛男性、第■因子の血友病患
者、静脈内薬剤投与者及びハイチ人を包含するエイズの危険性として初期に認め
られたあらゆる群の血清中に、ウィルスの特異的抗体が発見された。さらに最近
のそして広範囲にわたる血清学的研究は、エイズまたはABC患者の異性愛の相
手及びアフリカのめる地域、特にエイズ並びにHTLV−Illは風土性である
と思われるザイール及びルワンダからの個人を包含して、ウィルスにきらされて
いる付加的な人々を認めた。
これらの血清疫学的研究は、HTLV −1[感染の伝達の様式及び範囲に多く
の示唆を与えたけれども、血清陽性の個人のHTLV−[1の特異的抗体の可能
な保Stたは治療効果に関する報告はなかった。それ故に、研究はエイズ及びA
BC患者がウィルス感染を阻害できる抗体活性を有するかどうかを決定するため
に行なわれた。そのような自然の防御機構は感染された宿主に細胞をウィルスの
広がった細胞から回避することを可能にし、それ故に疾病の進行が阻止または予
防され得る。数種の動物のレトロウィルス系において、中和抗体は、標的細胞レ
セプターへのウィルスの初期の付着に関係するウィルスのエンベロープ塘蛋白質

このレセプターに対するウィルスの結合を阻止することによシ、ウィルス中和抗
体は効果的にウィルスの感染を阻害し得る。ここに報告された研究において、
)ITLV−■はエイズまたはABC患者の特異的な中和抗体を引き出、すかど
うか及び該抗体は何らかの保護効果を示し得るのかどうかがめられた。
HT細胞系のH9クローン(特定の方法及び例)は無細胞HTLV −III感
染の標的として使用され、そしてウィルス中和抗体活性について数種の血清を初
めに分析した。
H9細胞の感染は1間接免疫螢光試験(1ndirec、t immunefl
uorescence assay )でモノクローナル抗体を使用してHTL
V−[1p24 の表出を追跡することによシ評価された。ウィルス中和活性に
対し血清陽性または陰性の予備培養されたHTLV −■ウィルスでH9細胞の
感染の動力学を第1図に示す。感染後3日めまでに、健康な通常の献血者の血清
で前処理されたHTLV−[1と共に培養されたH9細胞(D タイタイ80
% 73(、HTLV−1[p24 O表出によシ示されるように感染された。
一方、ABC患者からの血清で前処理したウィルスに感染された場合の3日メK
Id、 H9細1taO10%LカHTLV−111 p24 f表出しなかっ
た。この感染の阻害が細胞の抗原よりむしろウィルスの抗原により実現されたこ
とは、同じ血清で処理したウィルスよりもHTLV−[1と処理し続いて細胞の
前処理によるH9細胞の方がよシ容易に感染することによシ示された(第1a図
)。ろる種の血清の阻害活性は、活性が滴定可能であるから、高血清濃度の非特
異的効果のみではなかった。第1b図に滴定した数種の血清により示されるよう
に、阻害活性を有する血清は、エイズの危険性のある群の患者及び健常者の全て
のカテゴリーに見い出された。
検出された阻害活性が、細胞の抗原よりむしろウィルスの抗原に対して影響され
ることを確認するために、血清を無細胞ウィルスの調製物または感染もしくは未
感染H9M胞に吸収させた。細胞での吸収はあまり効果はなかったけれども、ウ
ィルス調製物での吸収は下の第1表に示すように阻害効果を有する血清の力価を
実質的に減じた。
H9細胞のHTLV−11感染を中和することができる自然の抗体は、成人工イ
ズ患者の60%に及びARCの成人の80釜に検出されるが、しかし通常の健常
な異性愛者の対照においては0%である。抗体力価の相乗平均は。
エイズ患者に比べABC患者では2倍高く、2人の抗体陽性な健康異性愛者でさ
え高かった。この発見は、ウィルス中和抗体は生体内である種の保線効果を発揮
する可能性のあることを示唆する。これらの抗体の存在は、有利な治療に利用さ
れ得るHTLV−11に対する免疫学的応答ヲ示す。また、これらの研究に使用
された方法論は、将来のワクチン研究に直接利用できる。
従って、血清IgGが、H9細胞に対するHTLV−IIIの感染性をウィルス
に結合することによシ中和することができるという系を明らか圧したので、多く
のヒト血清をこの抗体活性のために分析した。その結果を下記の第2表に示す。
エイズまたはABCの患者の多くは、そのような活性を示さなかった健康な異性
愛の個人と対照的に。
ウィルス中和抗体を有することは明白でるる。抗体価は。
両患者群では500以上に及ぶが、全般的な力価は低かった。しかしながら、A
BC患者は研究されたエイズ患者の抗体価に比較して、2倍高い相乗平均の抗体
価を有することがわかった。エイズ発症の危険性を有する健康々同性愛者の間で
は、抗体価の相乗平均は、2人だけの抗体陽性の個人について決定されたもので
あるが、2?i1の患者群のどちらかの値より実質的に高かった。よシ軽いまた
は重要でない疾病の徴候でのより高い力価のこの傾向は、中和抗体の保護効果を
示唆する。
HTLV −1[1中和抗体活性は、通常の健康な異性愛者には検出されなかっ
た(第2表)。しかしながら、かろうじて検出可能な力価(13の)は急性の単
核症の患者からの4血清試料のひとつに得られた。この結果は、高レベルの血友
病の抗体を有する血清中のウィルスの抗原との弱い交叉反応性を示唆するであろ
う。
他のレトロウィルス系において、多くのエンベロープ循蛋白は、中和抗体の標的
でろる。これらの自然に起こるウィルス中和抗体は、生体内での保護効果に関し
て意味深いものである。
本発明の結果は、よシ軽い疾病の人またはHTLV−■に感染されたが、まだ病
気の徴候も示していない人が、よシ高い中和抗体価を有する傾向を示す。このこ
とは、ヒトワクチン研究が値うちのおることを示唆する。一方。
HTLV −[iの生物学全体において中和抗体の役割がビスナウィルス(vi
、sna virus )系においてみられたものと類似しているであろうこと
を推測することは興味深い。第一に肺及び中枢神経系に影響するヒツジのゆりく
シと進行する疾病を起こす非発ガン性レトロウィルスであるとスナウイルスの感
染は、永続性でおる。ウィルスによって引きだされた、中和抗体は、病気の途上
で生じる突然変異ウィルスによる感染を阻害することができない狭い範囲の特異
性を有することがわかった。このように、中和抗体は選択圧を及ぼし、非中和の
突然変異ウィルスの複製を導く。比較的最近感染てれた動物から取り出した′初
期の血清°は、3年取上前感染された動物から得られた°後期の血清ゝに比較し
て、より制限された中和範囲を有することもまた興味深い。これら1後期の血清
”は、全ての先祖の株を包含したよ多広い範囲のビスナ突然変異体を中和するこ
とができた。このことは、特に分離から分離までの明白な遺伝子の変化、特にウ
ィルスのエンベロープ部分での変化のために、及びHTLV−n[(Dビスナウ
ィルスとの明白な類縁のためにも、HTLV −■に関係がおる(ゴング等、サ
イエンス、印刷中(Gonda。
et al 、 鉦14. in press )。
エイズ及びARC,を者並びにHTLV −n[に感染された健常人の血清中の
HTLV−Ill中和抗体の立証は、治療に有利に利用されても良い病気進行の
途上に免疫学的応答を明らかにする重要な発見である。嘔らに適当なワクチン研
究が初めからウィルス感染を妨げることに効果的でめることを示す。ここに記載
した方法論は、これらの将来の手J@ヲ追跡することに有用でろろうし、さらに
HTLV−[1感染の生物学に関する基礎的な研究に有用でもめろう。さらに研
究は、患者血清のウィルス中和抗体の存在が予後の価値を有するかどうかを決定
するであろうし、または適当な養生療法を示すでろろう。
図面の説明
通常の健康な異性愛者(6)及びARC患者(@からの血清を第1A図で比較し
ている。平行実験において、これらの同じ血清を、H9細胞と共に1時間、4℃
で予備培養した。細胞t−PBSで洗浄し、無細胞HTLV−Ill調製物で培
養し、そして実施例2のように培養した。血清とH9細胞のこの処理の結果は、
通常の健康な異性愛者の血清(ハ)及び人孔C患者の血清(Aを表わす。
第1B図において代表的な滴定は、ウィルスの中和抗体活性に関し陰性の2種の
血清を示す:エイズ患者(6)及びABC患者(ロ)。ウィルス中和抗体に関し
陽性の代表的な血清は、小児エイズの場合(@、ABC患者(1−) 、健康な
同性愛者(ハ)及び成人工イズ患者に)から得られた。得られた全ての数値を、
標準抗体陰性血清の存在下で得られる感染のレベルに標準化した。

(a) IgGはPBSに平衡化した蛋白質A−セファロースに吸収させること
により15 mlヒト血清から精製した。次にPBSでカラムの十分な洗浄を行
ない。
IgGを11Mグリシン−MCI 、 pH2,8で溶出し、溶出液t 2 M
)リス−HCl 、 pHao テ中和し、10mM炭酸水素アンモニウムに
対し十分に透析し、そして凍結乾燥した。精製した画分をPBS Q、5ゴに溶
解し、濾過滅菌し、実施例2に記載するような中和抗体活性の滴定用の媒体に希
釈した。
ウィルス吸収実験のために、2ないし5x108ウィルス粒子/+di含む無細
胞ウィルス上清62df実施例2に記載するようにベレットにした。ウイルスノ
ヘレットを吸収させるべき血清の1;10の希釈液1004に再懸濁し、4℃で
一晩培養する。ウィルスを遠心によシ再びベレットとし、吸収された血清はウィ
ルス中和抗体活性の滴定のために確保した。血清を同様に洗浄した107個の細
胞ベレット上に吸収させ、そして滴定した。
(b) HTLV−Ill p−24−II性al胞ノ百分11)(iiH1試
験血清として同様に処理した標準陰性血清の存在下で得た感染のレベルに標準化
した。ここで抗体価は、ウィルス感染がこの標準陰性血清の存在下で得られるも
のの60%であるときの血清希釈の逆数として表した。

ウィルス吸収実験のために、2ないし5×10”ウィルス粒子/1ljt−含む
無細胞ウィルス上清62iIj’i実施例2に記載するようにベレットにした。
ウィルスのベレットを吸収させるべき血清の1=10の希釈液100μlに再懸
濁し、4℃で一晩培養する。ウィルスを遠心によシ再びベレットとし、吸収され
た血清はウィルス中和抗体活性の滴定のために確保した。血清を同様に洗浄した
101個の細胞ペレット上に吸収づせ、そして滴定した。
HTLv−11p −24−陽性細胞の百分率の値は、試験血清として同様に処
理した標準陰性血清の存在下で得た感染のレベルに標準化した。ここで抗体価は
、ウィルス感染がこの標準陰性血清の存在下で得られるものの60%であるとき
の血清希釈の逆数として表した。
実施例2
)iTLV−■中和抗体のヒト血清をスクリーニングするための方法において、
2ないし5 x 10’ HTLV−11粒子/dを含む媒体t−H9/HTL
V−11細胞から得た。初めに使用したウィルスの童は、ウィルス調製物を滴定
すること及び感染後3日までに50ないし80%の感染したH9Ia胞を得るよ
うな試験の11選択することによシ決定ばれた。これは一般的に標的細胞あたり
実質的に過剰量のウィルス粒子を要求した。細胞を低速遠心により除去し、そし
てウィルスを含有する上清を3時間、32,000xgで遠心した。ウィルスの
ベレットを媒体(20慢ウシ胎児血清及びペニシリン/ストレプトマイシンを含
有するRPMI I 640 )全体積2.25j中に再懸濁した。
未感染H9細胞を媒体中で洗浄し、ポリプレ/2μ9/dを含有する媒体中室温
で20分間培養した。細胞を媒体中で洗浄し、そして媒体中に4X10’/il
の濃度に再■
懸した。試験すべき血清を56℃、30分間加熱不活性化し、そして濾過滅菌し
た。各試験のためにウィルス懸濁液20μl及び血清1:10希釈液20μit
−混合し、そしてマイクロタイタープレー) (m1crotiter pla
te )ρウェル(well)中で1時間4℃で、そして次に15分間室温で培
養した。H9細vL(10μt)を各ウェルに加えそして培養を1時間37℃で
続けた。各混合物の一部分(15μりを別のマイクロタイタープレートの2つの
ウェル中の200μlの媒体中に移した。培養を57℃・5% CO,インキエ
ペーター中で行なった。 3日後、各々のウェル中の培養物を取シ除き、燐酸緩
衝化食塩水(PBS)で2度及びPBS:水、1:1で1度洗浄した。
細胞を同溶液およそ30μ/に懸濁させ、セして5ないし10 μ/分tHTL
V−111p24 K対fルモ/ / a−tk抗体を使用して間接固定細胞免
疫螢光試験用の8−ウエルノトキンプラズモシススライド(toxoplasm
osis jlides )上にスポットした。
中和抗体活性を示す血清及び1:10希釈液を引き続き連続して希釈し、試験は
抗体価を決定するために繰シ返した。
下記の請求の範囲中及び明細書中、血清とは実質的な童の抗HTLV −lを含
む血清を意味する。これは成人及210の範囲である。

Claims (10)
- 1.(a)HTLV−IIIウイルスをHTLV−IIIに対する抗体を含有す る血清中の自然のヒト抗体で処理し、そして(b)自然の抗体により生じた保護 の測定を決定するために残りのウイルスの感染性を試験することよりなるHTL V−IIウイルス感染性を中和する方法。
- 2.試験がHTLV−IIIに特異的なモノクローナル抗体で行なわれる請求の 範囲第1項記載の方法。
- 3.モノクローナル抗体が抗HTLV−IIIp24である請求の範囲第2項記 載の方法。
- 4.(a)HTLV−IIIウイルスを自然のヒト抗体を含有する血清と処理し ; (b)その結果の培養物をH9細胞で感染及び培養し;そして (c)HTLV−IIIまたはHTLV−IIIp24に特異的なモノクローナ ル抗体で残りの感染性を試験することからなるHTLV−IIIウイルス感染性 を中和する方法。
- 5.モノクローナル抗体がHTLV−IIIp24に特異的である請求の範囲第 4項記載の方法。
- 6.モノクローナル抗体がHTLV−IIIに特異的である請求の範囲第4項記 載の方法。
- 7.さらに50ないし80%のH9細胞が感染されるように試験のためにウイル スの量を選択することからなる請求の範囲第4項記載の方法。
- 8.HTLVまたはp24HTLVに特異的なモノクローナル抗体の添加により 、残りのウイルスの感染性を計測することからなる、中和抗体を含有する血清の 添加によりHTLV−IIIウイルスの保護効果を試験する方法。
- 9.さらにウイルスの感染をさせるために、モノクローナル抗体の添加の前に3 日保持することからなる請求の範囲第8項記載の方法。
- 10.免疫螢光試験が抗原及びモノクローナル抗体の反応に利用される請求の範 囲第8項記載の方法。
