Abstract

PURPOSE:To transplant a microorganism without damaging the agar surface by piercing with a needle, by changing over the transplanting needle at the set position from a locked state top a free state and stabilizing the tip of the transplating needle in a tilted position. CONSTITUTION:A base part 2 for holding a transplating needle is rotatably mounted by a pin 3 eccentric to the central aix (S) thereof, and constituted to stabilize the transplanting needle with the tip 11 at the tilted position in the free state of the needle. The transplanting needle is then changed over to the locked state, and the tip 11 is pierced in the surface of an agar (K) to fish the microorganism. The transplanting needle is then changed over to the free state to inoculate the microorganism to a transplating laboratory dish. After the tip 11 reaches the surface of the agar (K), the transplanting needle escapes in the direction indicated by arrow (P) without piercing the surface of the agar (K).

(Prior art) When transplanting cultured bacteria to another petri dish, conventionally the work was mostly done manually using a transplant needle, but when the amount is large, it becomes extremely burdensome and it is difficult to continue the work. In addition, when this moving work is performed using a machine, the position of the transplant needle is insufficiently controlled, especially when inoculating the bacteria caught from the original petri dish onto the agar surface of the transplant dish. However, during inoculation, the transplant needle was stuck into the agar surface of the transplant dish, and the transplant needle was moved while being stuck, which caused the agar surface to become rough.

First, FIGS. 1 and 2 will be explained. The transplant needle 1 is fixed to a base 2, and the whole is rotatably attached to a surrounding wall (not shown) by means of a bottle 3. The position of the bottle 3 is eccentric from the axis S, so that when the transplant needle 1 is free, the tip 11 of the transplant needle 1 is at a position slightly to the side from directly below, as shown in FIG. , for example, so as to be stable at an angle of about 15" to 20 degrees. On the other hand, a protrusion 21 is provided on the base 2, and the piston 5 from the lock cylinder 4
It is configured so that the two are pressed against each other.

The transplant needle 1 is rotated clockwise in the figure by the pushing motion by the piston 5. The stopper 6 stops this rotation at a fixed position. Due to the stopper 6, the transplant needle 1 is held at its tip 1.

Needles such as those shown in (B) and (C) can be used. The transplant needle 1 shown in (A) is made by connecting a conductive material 1b such as copper to a nichrome wire, heat-resistant steel, or other electrical resistor material 1a with a rounded tip 11, and is capable of self-heating sterilization. It's a needle. The transplant needle shown in (B) is a metal rod whose tip 11 is bent to give it a rounded shape.

Returning to FIGS. 1 and 2 to continue the explanation, upon transplantation, it is first necessary to collect bacteria from the cultured original petri dish G. This fishing bacterium holds the transplant needle 1 in a locked state,
The whole, ie, transplant needle 1° lock cylinder 4 and piston 5. This is done by lowering the entire body including the stopper 6.

By descending, the tip of the transplant 1 pierces the agar surface, and bacteria from colonies formed on the agar surface adhere to the transplant needle 1. By raising the entire body, the bacteria can be caught while still attached to the hook 1. Once the fishing bacteria have settled, the next step is inoculation. This inoculation is carried out with the transplant needle 1 in a free state, that is, the entire transplant needle 1. Lock cylinder 4 and piston 5. This is done by lowering the entire body including the stopper 6. In this case, the degree of descent is determined by the extent to which the transplant needle 1 slightly bites into the surface of the agar of the transplant petri dish I. However, since the transplant needle 1 is held in a free state with its tip @ 11 slightly on the side from the direction directly below, when the tip 11 reaches the surface of the agar (as shown by the two-dot chain line in Fig. 2) ), even if the needle falls further than that, the needle 1 escapes in the direction indicated by the P arrow and does not penetrate the surface of the agar. Therefore, the agar surface is prevented from being roughened by the needles 1.

FIG. 4 shows an example in which the transplant needle device according to the present invention is incorporated into an automatic transplant device. That is, a rotor 62 is provided on a base 61, and the base 61 itself moves up and down in the vertical direction (Z-axis direction) along an elevator shaft 63. A plurality of transplant needles are attached at regular intervals on the circumference of the rotor 62. The bottle 3 for rotatably supporting the transplant needle 1 is supported by the wall of the rotor 62 into which the base 2 is fitted.

The rotor 62 is driven by a driving source 65. Deceleration v! 166, the new transplant needle 1 is sequentially moved to the downward center position by rotating by a predetermined rotation angle. On the other hand, there is an X-Y table 71 on which a cultured original petri dish G and a transplanting petri dish I can be fixedly placed as a pair, and axes 72 and 73 for moving the table 7 in the X-axis direction and the X-axis direction. , a drive source and its transmission mechanism (not shown) are provided. As a result, the X-Y table 71 is moved smoothly, and the original Petri dish G and the transplanting Petri dish ■
and are placed under the transplant needle 1 alternately to transplant the bacteria. The details of the transplant operation will be explained with reference to FIG.

First, the rotor 62 rotates and the new transplant needle 1 is set in a downward position. Next, the X-Y table 71 moves so that the position of a predetermined colony on the original petri dish G is located below the transplant needle. Next, the base 61 itself descends along the lift shaft 63. As a result, the transplant needle 1 is lightly pierced into the predetermined colony position.

Next, the base 61 is raised, and bacteria are attracted to the transplant needle 1. Next, the X-Y table 71 is moved to place the transplantation Petri dish I at a predetermined position directly below the transplantation needle 1. Next, transplant needle 1
The lock is released, and the transplant needle 1 becomes free (FIG. 2). Then, the base 61 descends again, and the transplant needle 1 comes into contact with the agar at a predetermined position in the transplant Petri dish (2) to perform inoculation.

In this case, when inoculating in a linear manner, the X-Y table 71 is moved by a predetermined amount while the transplant needle 1 remains inoculating the agar, and linear inoculation is performed. In the case of spot inoculation, the X-Y table 71 does not move. When the inoculation is finished, the base 61 is raised and the transplant needle 1 is locked on. Since each colony may be transplanted from a plurality of colonies in the original petri dish G, it may be transplanted to different positions many times in the same petri dish. Once the porting has been performed this predetermined number of times, the porting work ends.
