Abstract

PURPOSE: Estrase obtained from internal organs of mammalian animals is simply and efficiently purified by utilizing its characteristic affinity to the residual group of glycine or alanine.
CONSTITUTION: Glycine or alanine is bonded through a spacer with a support such agarose or dextran and the resultant support is brought into contact with crude estrase obtained from internal organs of mammalian animals. Then, contaminating foreign proteins are eluted with a buffer solution free from sodium chloride, preferably a phosphate buffer solution of about 7.4pH, then, the estrase held on the support is eluted with a buffer solution containing less than 0.2M sodium chloride, preferably by the gradient method in which the concentration of sodium chloride gradually increases from 0 to 0.2M.
