Abstract

PURPOSE: Acidic mucopolysaccharide is treated with aqueous dimethylsulfoxide under an acidic condition of a specific pH to effect selective cleavage of β-D-hexosaminide linkages, thus producing depolymerization products such as disaccharide or its oligosaccharide used as a medicine.
CONSTITUTION: An acidic mucopolysaccharide such as chondroitin-6-sulfate is dissolved in aqueous dimethylsulfoxide at a pH higher than 2, preferably 3W5.5 and heat-treated at room temperature to 120°C, preferably 80W120°C under stirring to effect depolymerization of the acidic mucopolysaccharide. After completion of the reaction, water and sodium hydroxide are added and mixture is fractionated by the column chromatography to produce disaccharide consisting of uronic acid and hexosamine and its oligosaccharide. The amount of water in dimethylsulfoxide is preferably in the range from the stoichiometric amount necessary for hydrolysis to less than 20wt%.
