Abstract
PURPOSE:To obtain an oral agent for the remedy of thrombosis, etc., absorbable through the epithelial cells of small intestine in high efficiency, preventing the decomposition of antithrombin factors, by encapsulating an aqueous solution of a mixture of heparin and antithrombin III in liposome. CONSTITUTION:An aqueous solution of a mixture of heparin and antithrombin III is encapsulated in liposome. Preferably, the solution is further added with a proteinase-inhibitor effective to inhibit the action of decomposition enzymes and prevent the decomposition, e.g. aprotinin, dimethylcarbamoylmethyl-p-(p-guanidinobenzoyloxy)phenyl acetate mesylate, etc. together with the liposome-membrane constituting material. Each component is freeze-dried and mixed with each other before encapsulation. The liposome is preferably a multi-layered liposome having a size of about 1mu, and having 2-3 layers. The osmotic pressure of the aqueous phase in the liposome is preferably higher than that of the absorbing cell.
