Abstract

PURPOSE:To measure easily a helper T cell function in vitro, by culturing a T cell isolated from bright red blood of a patient along with a B cell isolated from bright red blood of a normal person under a specific condition and by using <3>H-labelled thymidine. CONSTITUTION:A helper T cell (having a function of helping antibody production) is isolated from bright red blood of a patient and cultured in the presence of a Tm cell having a suppressor function (suppressing antibody production) controlled by mitomycin treatment, a B cell and B cell mitogen (PWM promoting mitosis) isolated from bright red blood of a normal person and <3>H-labelled thymidine. On the other hand, a Tm cell of a normal person is cultured in the same way by adding the same B cell and PWM of a normal person and the same <3>H-labelled thymidine. An amount of DNA synthesized in a process where the B cell differentiates and proliferates is measured by determining, with a toluene scintillator, the <3>H-labelled substance incorporated in the B cell collected after the proliferation. An abnormal part of a patient suffering from immune disease is diagnozed bosed on differences in a helper function between the patient and the normal person by comparing their radioactivities, to thereby facilitate medical treatment.
