Abstract

PURPOSE: To measure the specific hemagglutination inhibiting antibody by using the hemagglutinin obtd. by treating the bacterium liquid of the agglutinin deficient strains to the fresh red blood cells of haemophilus paragallinarum with an aq. potassium rhodanate soln.
CONSTITUTION: The agglutinin deficient strains to the fresh red blood cells of the haemophilus paragallinarum are cultured in the adequate media. Serotype 2 bacterium strain, H-18 strain, etc., are used as the bacterium strain for the agglutinin deficient strains. The bacteria are then collected from the cultures and are treated with the aq. soln. of the potassium rhodanate contg. a physiological salt soln. and acoustic waves by which the surface materials of the cells are removed. The treated liquid is refined to obtain the hemagglutinin. This hemagglutinin has substantially no agglutinatability to the fresh red blood cells but has the high agglutinatability to the glutaraldehyde fixed red blood cells. The above-mentioned hemagglutinin is thus used as the antigen and the liquid suspension of the glutaraldehyde fixed red blood cells is used as the liquid suspension of the red blood cells in the measurement of the hemagglutination inhibiting antibody and, therefore, the specific hemagglutination inhibiting antibody is measurable and the infectious coryza, etc., are prevented.
