Abstract

PURPOSE: To prepare pseudouridines in high yield, by cultivating a bacterium belonging to the genus Pseudomonas in a medium comprising a uracil donor and a saccharide source.
CONSTITUTION: A bacterium (e.g., Pseudomonas THD-9) belonging to the genus Pseudomonas, capable of producing pseudouridine and/or a 5'-0-acetylpseudouridine is inoculated into a liquid medium comprising 20W50mM uracil donor (e.g., uracil) and 20W300mM saccharide(e.g., ribose) which the bacterium to be used can assimilate in a molar ratio of the uracil donor to saccharide of 1:1W1:15, cultivated aerobically at a pH of 6W8 at 15W35°C for 2W4 days, and pseudouridine and/or 5'-0-acetylpseudouridine formed in the medium is collected.
