Abstract
PURPOSE: At least one selected from manganese, manganese salts, and protamine sulfate is added to and mixed with an aqueous suspension of crushed human myelogenic leukocites and the supernatant is treated to separate and recover human- originating myeloperoxidase of high quality in high yield.
CONSTITUTION: An aqueous suspension of human myelogenic leukocites is treated with a homogenizer at 0°C for 5min to prepare an aqueous suspension of crushed leukocites, which is subjected to following treatments. The suspension is centrifuged into the precipitate and the supernatant and the yellowish green supernatant is recovered as a fraction containing myeloperoxidase. The fraction is suspended in a buffer solution of pH6W8.0 containing potassium phosphate and homogenized. Then, at least one selected from Mn, Mn salts and protamine sulfate is added to the homogenized suspension. The resultant suspension is centrifuged again to separate the myeloperoxidase fraction, i.e., the supernatant, which is subjected to cation exchange column chromatography to recover nyeloperoxidase.
