Abstract
PURPOSE: To prepare the titled substance having high thrombolytic activity, in high concentration and yield, by separating from a solution obtained by culturing cells capable of producing plasminogen-activating factor, in the presence of a specific compound.
CONSTITUTION: Plasminogen-activating factor having a molecular weight of 45,000W68,000 is separated from a solution containing the factor and obtained by contacting (A) cells capable of producing plasminogen-activating factor and originated from human kidney or lung, with (B) a solution containing a nitrogen source, a carbon source, inorganic salts, and if necessary, organic nutrients. The separation is carried out in the presence of at least one metal-chelating agent (e.g. ethylenediaminetetraacetic acid). The chelating agent suppresses the decomposition and denaturation of the factor during the separation process, and hence ensures the preparation of the factor having uniform molecular weight, in high purity and concentration.
