Abstract

PURPOSE: To detect the subject antibody easily and with high sensitivity, by bringing a latex reagent prepared by sensitizing high specific gravity latex particles with a staphylococcal α toxin into contact with a body fluid or its diluted solution and to observe a sedimentation pattern by a microtiter method.
CONSTITUTION: A staphylococcal α toxin is purified, and particles of high specific gravity latex having a specific gravity of 1.1 or more, or polymer latex such as chlorostyrene, vinyl chloride or the like are sensitized with this purified toxin. After the sensitization of latex particles, the latex particles are washed with an aqueous medium to eliminate surplus antigen. Then, the unsensitized surfaces of the latex particles are saturated with bovine serum albumin or the like, and subjected to a treatment for preventing nonspecific agglutination. The thus prepared latex reagent is brought into contact with a body fluid such as human or animal serum, or its diluted solution on a plate for microtiter, and a sedimentation pattern of latex is observed whereby the existence of the α toxin antibody is discriminated.
