Abstract

PURPOSE: To produce sensitized blood cells capable of performing detection of the subject antibody with high sensitivity and easily, by coupling hemolytic streptococcal nicotinamide adenine dinucleotidase (NADase) to red cells processed by a coupling agent.
CONSTITUTION: Live red cells or fixed red cells ar processed using a coupling agent such as tannic acid to obtain blood cells processed by the coupling agent. A solution containing NADase, which is separated and purified by a hemolytic streptococal cultivation filtrate, is brought into contact with the above-described processed blood cells to produce NADase-sensitized blood cells. In order to preserve these sensitized blood cells for a long period of time, fixed red cells are used as red cells, and the thus obtained sensitized blood cells are suspended in a preserving solution to be preserved, or preserved by freeze-drying the blood cells together with a medium containing a protecting agent. In the case of using the freeze-dried article, a diluting solution is added to the sensitized blood cells to prepare a diagnosis solution.
