Abstract
PURPOSE: To determine phosphoric acid in biospecimen such as serum, urine, etc., other phosphoric acid-containing specimen, or a phosphoric acid-producing reaction system, by reacting the specimen with a system containing pyruvic acid and pyruvate oxidase, and measuring the produced H2O2 or consumed O2.
CONSTITUTION: The objective kit is composed of (A) pyruvate oxidase obtained from the cells of Pediococcus, etc. and (B) pyruvic acid, and optionally (C) flavine- adenine dinucleotide (FAD), thiamine pyrophosphate, and calcium ions, etc. e.g. one or more ion-generating salts selected from e.g. CaCl2, CoCl2, MnCl2, etc. A necessary amount of the reaction liquid is taken out from the kit, and made to react with a specimen at 20W40°C for 5W60min. The amount of the phosphoric acid in the specimen can be determined either by determining the oxygen consumption shown in the formula, or by determining the produced H2O2 by colorimetry or fluorometry using 4-aminoantipyrine and phenol or N,N-dimethylaniline, homovanillic acid, etc. as an indicator. The method is useful not only for the determination of phosphoric acid or a phsophoric acid-producing system, but also for the deterination of phosphatase activity, fatty acid, or a component necessary for the examina tion of biospecimen which generates phosphoric acid by the reaction.
