Abstract

PURPOSE:To obtain a sharp and stable chromatogram, by a method wherein, in case an amino acid is analyzed, a sample is colored in a reaction tank by the use of a coloring reagent beforehand, and the component is separated by the liquid chromatography in which a distribution type negative-phase separating tube is employed. CONSTITUTION:A sample in a chemical reaction tank 5 and a coloring reaction reagent solution 1 as well, fed by a piston type filler 2, are heated and agitated by means of a stirrer and a heater 6. The colored sample so obtained is automatically poured into a liquid chromatocolumn through a rotary valve sample weighing device 10. The sample, automatically poured, is fed to a distribution reversed-phase separating tube 16 by means of a transferred phase 15 fed by a liquid feed pump 14 for separation into components. The components so separated are detected, in order, by means of a detector 17. A coloring reagent for an amine chemical compound sample is preferably Oo-phthalaldehyde. This constitution permits analysis of a standard amino acid for a short period of time, say, 30 minutes.
