Abstract

PURPOSE:To measure the activity and existing quantity of the zooplankton in a short time by adding specific coloring matter to a sample containing the zooplankton, extracting the coloring matter taken in by the plankton a prescribed time later and measuring the degree of light absorption or the intensity of fluorescence of extracted liquid. CONSTITUTION:In the method measuring the activity and existing quantity of zooplankton so as to explicate the influence of hot discharged water of a power plant on environments, water solution of neutral red (density of about 0.1-1%) is added to the sample containing the zooplankton, such as sea water, and thereby coloring is made for a prescribed time (1.5hr, approx.). Thereafter the sample is crushed by an ultrasonic homogenizer or the like and then the aforesaid coloring matter is extracted in the 70%-alcohol solution, and the top is adjusted to be of pH3. The degree of light absorption (wavelength: 540nm) or the intensity of fluorescence (excitation wavelength: 517nm, absorption wavelength: 608nm) of the extracted liquid is measured. Since the aforesaid coloring matter is taken in only by living plankton and, moreover, more amount thereof is taken in by more active plankton in a short time, the method is fitted for the measurement of the activity and the existing quantity of plankton.
