Abstract

PURPOSE:To quantify blood hemoglobin precisely and easily without using an oxided substance such as potassium ferricyanide by optically measuring bound material of methemoglobin and aliphatic high sulfonate. CONSTITUTION:The aliphatic high sulfonate (ex. dodecane sulfonic acid sodium) shown by the expression (R is alkyl of C11-16; M is a positive ion of m value) is solved in water or buffer liquid with pH 5-10 approx., or shortly, nonionic surfactant being added, and thereby a reagent is formed. Next, the sample blood (or diluted liquid with physiological salt solution) and the aforesaid reagent are mixed at the normal temperature and the bound material of methemoglobin and aliphatic high sulfonate thus produced is quantified with comparison of color in the vicinity of wavelength 534nm. In addition, the aforesaid method unnecessitates the combined use of a hemolytic agent and is free from variation of an absorption curve within the range of pH 5-10 approx. and from the influence of albumen, bilirubin, hyperlipemia blood, white corpuscle and the like.
