Abstract

1,183,499. Preparing microscope specimens. BAXTER LABORATORIES Inc. Oct. 31, 1967 [Dec. 5, 1966], No. 49309/67. Heading G2J. A reagent for staining biological specimens, such as blood, for microscopic examination, comprises:- (a) a polychrome methylene blue solution such that when diluted with water to a concentration of one part of polychrome methylene blue solution in 2, 000 parts of total solution the ratio of the spectrophotometrically determined optical density in a 1 centimetre cell at a wavelength of 660 mÁto that at a wavelength of 620 mÁ is between 0À8 and 1À5, and (b) an eosin solution such that when diluted in water to a concentration of one part of eosin solution in 100 parts of total solution the spectrophotometrically determined optical density in a 1 centimetre cell at a wavelength of 520mÁ is between 0À3 and 0À7. In an example, the polychrome methylene blue solution is prepared by heating a solution of 1% methylene blue and 0À25% sodium carbonate to 95‹C. for four hours, filtering, ageing and adjusting the pH to 6À85 using sulphuric acid, and diluting the 1% methylene blue to 0À8% with potassium phosphate buffer having a pH of 6À9. The eosin solution is prepared by adding equal amounts of eosin Y and eosin B to a solution of Na 2 HPO 4 and Na H 2 PO 4 to provide a 0À05% solution of eosin, storing, adding 0À1% sodium azide to prevent bacterial growth and filtering. To stain a biological specimen it is immersed on a microscope slide in the polychrome methylene blue solution for 10-20 seconds, thoroughly rinsed, immersed for 10-20 seconds in the eosin solution, rinsed again and then dried in air.
