Abstract
PURPOSE: To prepare the titled compound useful as an intermediate for the preparation of a compound having blood platelet coagulation inhibiting activity, in high yield, by reacting a novel 2-halogeno-3,4-dihydro-condensed pyrimidine compound with a halogenoacetic acid ester in the presence of phase transfer catalyst.
CONSTITUTION: A novel compound of formula I (group of formula II is benzene ring or thiophene ring which may be substituted with lower alkyl, lower alkoxy, phenyl, halogen, etc.; etc.; X1 is halogen) is made to react with a halogenoacetic acid ester of formula III (R is lower alkyl; X2 is X1) in the presence of a phase transfer catalyst such as tetra-n-butyl ammonium iodide, at room temperature for several to several tens minutes to obtain the objective compound of formula IV in an yield of as high as 80W95%. Heating of the resultant compound together with NH3 affords the compound of formula V having blood platelet coagulation inhibiting activity.
EFFECT: Mild reaction condition, easy pretreatment, and high yield separation of the objective compound.
