Abstract

PURPOSE:To perform highly accurate measurement with ease by using polystyrene beads bonded with anti-human-alpha1-microglobulin alpha-1-microglobulin antibody and anti-human-alpha1-microglobulin antibody marked with peroxidase. CONSTITUTION:Polystyrene beads are immersed in the anti-human-alpha1-microglobulin antibody soln. obtd. by applying a general method for separating and refining antibody protein to antiserums, whereby anti-human-alpha1-microglobulin antibody bond polystyrene beads are formed. Said polystyrene beads are immersed in a test tube contg. the liquid to be tested so that the alpha1-microglobulin (antigen) in the liquid to be tested is bound to the anti-human-alpha1-microglobulin insolubilized and stuck on the beads, after which the beads are washed and are immersed in a separately prepd. anti-human-alpha1-microglobulin antibody soln marked with peroxidase, so that the anti-human-alpha1-microglobulin antibody marked with peroxidase is bound to the antigen bound to the anti-human-alpha1-microglobulin antibody on the beads. Thence, the beads are washed and are immersed in an enzyme substrate soln. to allow the beads to form a color developement, and the absorbance thereof is measured, whereby the antigen is determined.
