Abstract

PURPOSE: To determine the sero-enzyme with a good reproduciblity, free from variation in color with temperatur and time, by reaction of a diazotized p-nitroaniline liberated from the substrate with vanillic acid, and by colorimitry of the formed coloring matter.
CONSTITUTION: p-Nitroaniline liberated by using a substrate, e.g. L-leucyl p-nitroanilide, according to sero-enzyme concentration, is reacted with a nitrite under acidic conditions, to give a p-nitrobenzene-diazonium salt, which is coupled with vanillic acid in the presence of an alkali carbonate, e.g. sodium carbonate, to form an azo coloring matter (C). The colorimetry of C provides the sero-enzyme value.
