Abstract

PURPOSE: To obtain acid-fast bacteria in high yield, by culturing in a medium containing meat extract and L-asparagine as nitrogen sources.
CONSTITUTION: Acid-fast bacteria such as Mycobacterium tuberculosis, BCG bacteria, etc., are cultured in a medium containing 0.2W5wt% of meat extract and 0.05W2wt% of L-asparagine, usually at 30W40°C and pH6-8. The culturing is carried out, e.g. in a liquid medium without agitation, or with shaking, or in a deep tank with aeration and/or agitation. The yield of the acid-fast bacteria increses 2W4 time compared with the culturing in a conventional medium.
