Abstract

PURPOSE: To prepare the title compound useful as an intermediate of tryptophan (an essential amino acid), economically, in high yield, by reducing hydrolyzed 2- alkyl-4-(1-acyl-3-indolylmethylene)-5-oxazolone.
CONSTITUTION: 2-Alkyl-4-(1-acyl-3-indolylmethylene)-5-oxazolone, pref. 2-methyl- 4-(1-acetyl-3-indolylmethylene)-5-oxazolone, is hydrolyzed pref. in the presence of an acidic catalyst, esp. an aliphatic carboxlytic acid, to obtain α-acylamino-β- (1-acyl-1-indolyl)acrylic acid, which is converted to the objective N,1-diacyltriptophan by reduction, pref. to L-N,1-diacyltriptophan by assymmetric catalytic reduction. Hydrolysis of the objective compound affords tryptophan.
