Abstract

PURPOSE: To prepare an antibiotic substance, sisomicin, by culturing sisomicin- producing fungi belonging to Dactylosporangium genus.
CONSTITUTION: Fungi capable of producing an antibiotic substance, sisomicin, and belonging to Dactylsoporangium genus, e.g. Dactylosporangium thailandense G367, are cultured in a conventional cultivation medium at 25W35°C for 100W200hr under aeration, and after adjusting the pH of the cultured medium to acidic range, the medium is neutralized and filtered to obtain the filtrate of the cultivation products. The filtrate is treated with an anion exchange resin, the adsorbed active substance is eluted with 2N aq. ammonia, and the elute is concentrated, adjusted on pH, and treated with an anion exchange resin. The adsorbed material is eluted with aq. ammonia having concentration gradient extending over 0 and 0.35N, and the active fraction is concentrated under reduced pressure, and freeze-dried to obtain the objective antibiotic substance, sisomicin.
