Abstract
PURPOSE: A microorganism demanding amino acid is cultured under amino acid- starving conditions to culture the microorganism with selectively increased specific genetic information in the cells without using of chloramphenicol.
CONSTITUTION: A microorganism that demands amino acid and has a new plasmid made by building a DNA carrying external genetic information into the plasmid multiplying DNA in relax type is cultured in a medium containing amino acid in an amount equal to or less than that permits the microorganism the maximum multiplication. After the amino acid is consumed out and the multiplication of the microorganism substantially stops, the culture system is stood for at least one hour, then the amino acid is added by an amount required for multiplication to the medium to continue the cultivation. Thereby more amounts of the objective amino acid, nucleic acid, nucleoside, antibiotic or the like is produced than in conventional methods.
