PURPOSE: To obtain the title compound useful as an antitumor agent of raw material for other agents in high yield and purity, by decarboxylating 5-fluoroorotic acid under a specific degree of vacuum and a specified temperature.
CONSTITUTION: 5-Fluoroorotic acid is decarboxylated preferably in an inert gas atmosphere under vacuum at 230°C or above, preferably 250W290°C, and finally a degree of vacuum ≤1 mm Hg, preferably ≤0.5 mm Hg to give the title compound, which easily sublimes to afford 5-fluorouracil as a sublimate by cooling. The starting material 5-fluoroorotic acid is obtained by fluoroinating orotic acid with fluorine gas.
