Abstract

PURPOSE: To obtain the title compound useful as a raw matierial for preparing a spirolactone which is a diuretic, by reducing the product of reaction between androst -4-en-3,17-dione and a metal acetylide at a specific pH, and by treating the reduced reaction product with an acid.
CONSTITUTION: Androst-4-en-3,17-dione is reacted with a metal acetylide, e.g. potassium acetylide, in the presence of a hydrophilic organic solvent, e.g. tetrahydrofuran, to ethynylate the carbonyl group at the 17-position selectively. The pH of the reaction mixture is adjusted to 1W13, preferably 2-12, with a weak acid. The carbonyl group at the 3-position of the ethynylated compound is reduced to give a steroid mixture consisting mainly of pregn-5-en-20-yne-3,17-diol, which is treated with an acid at a pH≤2. The objective substance is separated from the acid-treated mixture.
