Abstract

PURPOSE: To prepare specific anti-hCG-β-antibody capable of detecting the free hCG-β in the body fluid, independently, and useful for the diagnosis of malignant tumor, abnormal pregnancy, etc., by treating body fluid containing anti-hCG-β- antibody with purified hCG containing little hCG-β.
CONSTITUTION: Human chorionic gonadotropin (hCG, pref. having a titer of ≥10,000 IU/mg) is treated with a modifier such as urea, etc., and purified to obtain its β-subunit (hCG-β). It is administered to an animal, e.g. a rabbit to form an hCG-β-antibody in the body fluid of the animal. When the concentration of the antibody in the fluid exceeds 10-8 mol/1, the body liquid is extracted from the animal, and concentrated in option. A high purity hCG containing ≤3.5%, pref. ≤1% of hCG-β is added to the body fluid, and incubated at 37W40°C for 0.5W1 hour. The specific anti-hCG-β-subunit antibody is obtained by removing the precipitate from the incubated liquid.
