動物及び植物の細胞を検出するための染色剤及び毛管スライド
本発明は染色剤、スライド及び血液サンプルを準備する方法に関する。
ヒト又は動物の血液中には、例えば赤血球細胞(RBCs)及びいくつかの型
の白血球細胞(WBCs)のような、様々な種類の細胞や微粒子が見られる。健
康個体では、各種の細胞数は複雑な身体メカニズムによってコントロールされ得
る。通常範囲からのあらゆる変化は、体内の深刻な生理学的又は病理学的な変化
を示し得る。それゆえ、血液中の各種の細胞数又は微粒子数の決定を可能とする
ことは重要である。この数は血液或いは患者又は植物のその他の生化学的流動体
又は生化学的物質、の適切なサンプルを準備し、前記サンプルを例えば顕微鏡等
でスクリーニングすることによって得られる。植物の細胞成分の染色に染色剤が
用いられてもよく、ここでの「植物」という特定には水中及びその他の環境サン
プル中に存在する菌類、バクテリア等を含んでいる。
従来は、最初に血液をスライドにのせ、空中で乾かし、その血液標本を(細胞
を固定するために)メタノール等の浴槽に浸し、最後に前記血液標本を染色する
ことによって塗抹標本サンプルが準備されている。
従来の染色剤及び染色方法は複雑で時間のかかる一連の過程を含んでいる。結
果として、全ての染色過程が完了するまでにはある程度の時間、例えば1時間か
かり、満足の行く結果を得るためにはオペレーターが熟練しており、経験を積ん
でいなければならない。塗抹標本作成過程において、血液中の細胞及び微粒子は
均一に散布されなかったり、又は傷つけられ、それゆえスクリーニング中に検出
するのが困難又は不可能となる。
さらに、従来の多くの染色剤は、構成成分が混合されると、例えば2〜3日の
短い時間しか安定ではなく、バルクで購入した場合、迅速に使用しないと容易に
汚染される。さらに、ある染色剤はある種の血液細胞しか分染しないか、及び/
又はある種の動物の血液細胞の染色にしか効果がなかったりする。
上記に述べられた問題と複雑さを考慮すると、上記のような方法で少量のサン
プルを準備することや、例えば外科又は診療所等のこのような技術を専門として
いない人によって準備されることは好適でない。結果として、分析のために血液
が遠方の契約した研究所に送られることもある。これはお金がかかり、比較的大
量の血液サンプルが提供される必要があり、かなりの時間遅延が含まれる。さら
に付け加えると、移送中に血液が変化したり、血液中の微粒子や細胞が損なわれ
るかも知れない。
本発明の目的は熟練していない人が困難なく高い品質のテスト標本を作成でき
るような方法を提供することである。
ひとつの局面から、本発明は以下の構成要素、
(i)被検査物質の特性に応じてその浸透圧が変更可能な酸性又は塩基性の緩
衝溶液と、
(ii)固定性の有機膜調整剤と、
(iii)サンプル中の細胞核及び細胞質成分を染色する化合物と、及び
(iv)溶媒と、
を含む、動物又は植物サンプルの染色剤を提供する。
好ましくは、前記緩衝溶液が分子量294.1のクエン酸3ナトリウム2水和物(
式C6H5O7Na3・2H2O)である。
好ましくは、前記膜調整剤が分子量90.12の2−エトキシエタノール(エチレ
ングリコールモノエチルエーテル)(式C2H5OCH2CH2OH)である。
好ましくは、前記染料化合物が分子量305.8のアズールB(c.l.52010;BSC
によって特定された、植物組織中の細胞RNA及びDNAを分染するアズール1
)(式C15H16CIN3S)、又はアズールBの誘導体である。
好ましくは、前記アズールB、又はその誘導体が、約80%の色素含有量を有
する。
好ましくは、前記溶媒が脱イオン水又は蒸留水である。
典型的には、染色剤を使用してヒト又は動物から採取された血液サンプルを染
色でき、並びに細胞及び/又は前記細胞中の微粒子の種類を分染できる。
本発明に係る染色剤は室温でも良好な安定性が得られるという利点があり、血
液細胞の種類及び細胞内構造に対してよいコントラストを与え、様々な異なる種
類の動物の血液染色に効果的に用いられるであろう。
別の局面からは、本発明は一容量の液体と以下の構成成分、
(i)被検査物質の特性に応じてその浸透圧が変更可能な酸性又は塩基性の緩
衝溶液と、
(ii)固定性の有機膜調整剤と、
(iii)サンプル中の細胞核及び細胞質成分を染色する化合物と、及び
(iv)溶媒と、
を有する染色剤とを混合する過程と、並びに混合物を静置する過程と、を含む染
色方法を提供する。
好ましくは前記染色剤が以下の構成成分、
(i)分子量294.1のクエン酸3ナトリウム2水和物(式C6H5O7Na3・2
H2O)と、
(ii)分子量90.12の2−エトキシエタノール(エチレングリコールモノエ
チルエーテル)(式C2H5OCH2CH2OH)と、
(iii)分子量305.8のアズールB(c.l.52010;BSCによって特定された

好ましくは、前記アズールBが約80%の色素含有量を有する。
典型的に、前記液体は血液であってもよい。
本発明に係る染色方法は、素早く簡単な一段階の過程を有し、新鮮な血液サン
プルの染色に使用できるという利点を有する。
典型的に、前記血液と前記染色剤が1:4の割合で混合されてもよく、緩やか
な攪拌で混合されてもよい。
好ましくは、前記混合物が1分から60分の間、15℃から60℃の間の温
度で、静置されてもよい。
さらにまた別の局面からは、本発明は毛管作用によって液体をそれ自身の中に
引き入れるようにしたスライドを提供する。
典型的に、前記液体は染色されている、又は染色されていない血液サンプルで
ある。
本発明に係るスライドの利点は、血液一滴を毛管チャンバーの一方の端に単に
導入し、毛管作用によってチャンバーの長さに沿って流れさせ、細胞の単分子膜
を形成することにより、塗抹標本を作る必要なしに、質のよい単分子膜の血液フ
ィルムを作成することができるということである。それゆえに本発明に係るスラ
イドは、容易に使用でき、従来どおりの方法で分析するための新鮮な血液サンプ
ルの素早く簡単な準備を促進する。
好ましくは、前記スライドは共に配置されたベースとカバーを含み、毛管チャ
ンバーを形成している。
本発明に係るスライドのさらなる利点は、毛管チャンバーが、血液等の分析さ
れるテストサンプルの制御された量又は既知量を提供し、そのため定量分析が可
能である点である。従来のスライドは定性分析にしか使用できなかった。
好ましくは、前記ベース及び前記カバーがガラス及び/又はプラスチック素材

本発明に係る準備方法は、素早く簡単であり、質のよい単分子血液フィルム膜
を生産し、例えば、血液細胞数の計量を容易にするという利点を有する。特別な
技術又はテクニックを必要とせず、例えば一般の開業医の外科等での小規模の使

本発明に係る染色剤は以下の手順により準備される。
クエン酸3ナトリウム(16.0g)を蒸留水に溶解し、1.01(哺乳動物
の赤血球細胞の等浸透圧の50%に相当)にする。2−エトキシエタノール
(200ml)と前記クエン酸3ナトリウム水溶液(800ml)を攪拌して混
合する(これを第一の緩衝溶液とする)。緩衝溶液のpHは8.9である(未調整)
。アズールB(1.0g)を500mlの前記第一の緩衝溶液に加え、1.01

まず図1及び2に言及すると、スライド1はベース2とカバー3を有し、両方
