本発明は、N−(2−ブロモアルキル)−アミド誘導体の製造方法に関し、該方
法においては、ニトリル及び、水もしくは酸の存在下で、アルケン性不飽和化合
物が、正の臭素イオン供与体と接触させられる。
そのような方法は、ジェー・ベーガーら、Journal fur praktische Chemi、第
311巻、1969年、第15〜35頁に記載されており、そこでは用いられる正のハ
ロゲンイオン供与体が、大抵塩素分子であり、いくつかの場合に臭素分子である
。
記載されている方法の欠点は、収率[選択された基質(substarate)に依存する
]が低い乃至非常に低いことである。
本発明の目的は、所望するN−(2−ブロモアルキル)−アミド誘導体が比較
的高い収率で得られる方法である。
これは、本発明に従い、正のハロゲンイオン供与体として、N−ブロモイミド
、N−ブロモヒダントイン及びN−ブロモアミドからなる群からのN−ブロモ化
合物を用いることによって達成される。
実は、驚くことに、正のハロゲンイオン供与体としてのそのようなN−ブロモ
化合物の利用は、広い範囲のアルケン性不飽和化合物について高い収率を達成す

使用されるニトリルは、好ましくはアセトニトリルである。ニトリルの使用さ
れるべき量は、好ましくはできる限り低く選ばれ、特にアルケンに対して30モ
ル当量より少なく、好ましくは1〜10モル当量のニトリルである。
本発明で使用されることができる好適な正のハロゲンの供与体は、例えばN−
ジブロモヒダントイン、N−ブロモサクシンイミド、及びN−ブロモアセトアミ
ドである。1,3-ジブロモ-5,5-ジメチルヒダントイン(DBDMH)が好ましく用いられ
る。
反応混合物中に存在する水及び/又は酸の量は、通常、アルケンの量を基準に
計算して、水のみが用いられる場合は、0.5〜4モル当量、及び、酸のみが用いら
れる場合は、

本発明に従い調製されたN−(2−ブロモアルキル)−アミド誘導体は、例え
ばβ−アミド酸、β−アミノ酸、及びβ−アミド−α−ヒドロキシカルボン酸の
合成に用いられることができる。このため、α−β−不飽和酸が、アルケン性不
飽和化合物として、本発明に従う方法によって最初にα−ブロモ−β−アミド酸
に転化され、次いで、β−アミド酸に還元される。β−アミド酸の加水分解が、

閉環化合物の、所望される1,2-アミノアルコールへの加水分解は、好ましくは
酸性条件下で行われる。なぜならば、酸性環境においては、これらの反応段階が

閉環及び加水分解についての用語、酸性条件とは、本発明の範囲内において、

条件下で不活性であり且つその中に水が溶解するところの水と相溶性の溶媒であ
り得る。好ましくは水が用いられる。使用されるべき水の量は重要でなく、当業
者によって容易な方法で決定され得る。
閉環及び加水分解が行われる温度は、重要でなく、通常、0〜120℃、好ましく
は40〜105℃である。
N−(2−ブロモアルキル)−アミド誘導体、加水分解の間に中間体として形
成される1,2-アミノアルコールのO−アシル誘導体(アシル基は用いられたニト
リルから誘導される)、及び1,2-アミノアルコールは、必要であれば、当業者に
知られる方法に従い、各々別々に得ることができる。
1,2-アミノアルコールの調製に特に適切な方法は、アルケンからのN−(2−
ブロモアルキル)−アミド誘導体の調製、閉環及び加水分解が、それらの間に、
中間的に形成
される生成物の単離無しに遂行されれば、すなわち、例えばインデンのシス-1-
アミノ-2-インダノールへの転化がいわゆるワンポット反応により行われた場合

おける閉環及び加水分解は、高い収率を与えることが見出だされ、及び良い生成
物を高い収率で得るために中間生成物の単離及び生成は不要であることが見出だ
