Microlonospora Carbonacea由来の新規オルトソマイシン
発明の要旨
化合物A、B、C、D、E、F、G、H、およびJを、微生物Micromonospora
Carbonacea var Africana(SCC 2146と命名)の発酵ブロス(broth)から単離
した。これらの化合物は、オルトソマイシン(orthosomycin)として同定された
。これらの化合物は抗菌剤である。培養物由来の成分(化合物A)は、最も強力
な抗菌剤であることが見出された。
本発明は、新規な抗菌性化合物A、B、C、D、E、F、G、H、およびJ、

発明の詳細な説明
微生物の発酵
Micromonospora carbonacea亜種Africana、株PF6-3の凍結培養物1mLを、種培
地100mLを含む300mLのエーレンマイヤー振とうフラスコに移した。培地の組成(
g/L)は、以下の通りであった:牛肉抽出物(Difco)3、Tryptone(Difco)5
、Cerelose(CPC、2001)1、ジャガイモデキストリン(Avebe、WPD-650)24、
酵母抽出物(Universal、Taston)5、炭酸カルシウム(Pfizer、Albaglos)1
、およびシリコーン消泡剤(Union Carbide、SAG-471、30%懸濁液)0.3mL/L。
フラスコを30℃で激しく撹拌しながら48時間インキュベート(300rpm、1インチ
ストローク)し、そして培養物30mLを6×2Lフラスコ(それぞれ、同一の種培
地を500mL含む)に移した。上記のような48時間のインキュベーションの後、培
養物3Lを、さらなる24時間の培養のために、同一の培地300Lを含む接種発酵器
(inoculation fermenter)に移した。最終的に、接種発酵器の内容物を、産生
培地10,000Lを含むより大きい発酵器に移した。産生培地の組成(g/L)は、以下

%〜100%飽和に維持しながら、通気および激しい撹拌下で、36℃において120時
間〜140時間発酵させた。1分あたり80標準立方フィート〜240標準立方フィート
の空気流を用いて、約120時間〜130時間、発酵を行った。
単離
発酵ブロスを、約25℃に冷却した。発酵ブロスの半量を、別の容器に移し、激
しく撹拌し、そして2N NaOHを用いて、pHを10.5に調整した。300L XAD-7樹脂
(Rohm&Haas非官能性アクリルエステルポリマー製吸着剤)を、発酵ブロスにチ
ャージし、そして0.5時間激しく撹拌した。pHを9.25に下げ、そして3.5時間激し
く撹拌した。pHを、さらに7.00に下げ、そして樹脂をスクリーニングによりブロ

H8に調整された0.1Mリン酸ナトリウム一塩基緩衝液140Lで予め洗浄した)を10L
/分でカラムにチャージすることにより、樹脂ダウンフローから溶出させた。溶
出物を、150L〜200Lのカット(cut)中に回収した。カットを含む抗生物質複合
体を合わせ、そして水酸化ナトリウムでpH8に調整された0.1Mリン酸ナトリウム
一塩基緩衝液140Lを用いて抽出し、次いで脱イオン水2×60Lで抽出した。残留
する水の共沸蒸留により、酢酸エチル層を、30℃未満の温度において、最初の容
積の10分の1(約50mL)まで真空濃縮した。濃縮物を、ヘプタン100L(2容積)

酸化
10kg〜11kgの粗抗生物質複合体を、抗生物質13-384の複合体のニトロソ成分を
酸化するために、激しく撹拌しながら80/20酢酸エチル-アセトン(10容積)に溶
解させた。重炭酸ナトリウム2kgと、バナジルアセチルアセトネート(acetyace
tonate)触媒50gを添加した。温度を25℃〜30℃に維持しながら、tert-ブチル過
酸化水素(2,2,4-トリメチルペンタン中の3M溶液)5.5Lを、0.5時間かけてゆ
っくり添加した。酸化の進行を、HPLC(高圧液体クロマトグラフィー)によりモ

最初の精製
粗酸化物質の約2.5kgを、酢酸エチル9Lに溶解させ、そして、酢酸イソプロ

則シリカゲル(70μm〜250μm)70kgで充填された10フィート×直径1フィート

抗生物質複合体のさらなる精製
精製を、2工程のクロマトグラフィー手順により達成した。最初の精製で得ら
れたヘッドカット(ID 33285-104-2)およびテイルカット(ID 33285-104-1)の

調整し、そしてジオールカラム床容積に基づく画分を集めた。次いで、新規な成
分を含む画分を、265nmにおけるUVシグナルモニタリングに基づくピーク収集を
行いながら、PVA-Sil(ポリビニルアルコール官能基化シリカゲル)上の半分取
(semi-preparative)高圧液体クロマトグラフィーを用いて精製した。個々の成

実施例1
富化されたテイルカット(ID 33285-104-1)5gを、CH2Cl2:MeOH(96:4 v/v
)20mL中に溶解させ、そしてガラスカラム(600×50mm、1.18L)中に含まれる予
備コンディショニングした(pre-conditioned)ジオール結合シリカ(40μ〜63
μ、不規則媒体)200g(約400mL)に付与した。サンプル付与前のジオールカラ
ムコンディショニング工程は、MeOH1Lに続いて最初の移動相CH2Cl2:ヘプタン
:MeOH(60:40:2 v/v/v)1.2Lを通過させる工程を包含した。サンプル付与後

た。4回のさらなる注入がなされた後、計24mgの化合物A(98%より高い純度)
を得た。
複合体2(100mg)を、CH2Cl2:ヘプタン:MeOH(78:20:2 v/v/v)溶媒系
を移動相として用いた以外は上記と同様の半分取HPLC条件を用いて、さらに精製
した。2つの純粋な成分(化合物E(1.5mg)および化合物F(9.5mg))が得ら
れた。しかし、第1の成分(2.4mg)は、スペクトルデータの分析に基づいて、
2つの成分(化合物Cおよび化合物D)の混合物として同定された。
実施例2
富化されたヘッドカット5g(ID 33285-104-2)を、CH2Cl2:MeOH(96:4 v
/v)25mL中に溶解させ、そして再生したジオール結合シリカゲルカラム200gに付
与した。再生工程は、MeOHを通し、続いて最初の移動相CH2Cl2:ヘプタン:MeOH
(60:40:2 v/v/v)1.5Lを通す工程を包含した。サンプル付与後、移動相2.8L

なる2種溶媒系(binary solvent system)が選択され、これにより、第3の実

CH2Cl2:MeOH(96:4 v/v)1.0mL中に溶解させ、そして、n-ブチルクロリド:M
eOH(93:7 v/v)で平衡化された半分取PVA-Silカラム(250×20mm)上に注入
した。流速は15mL/分であり、そしてUV検出は265mnで行った。3つの成分を集め
た。最初の2成分(9.5mgおよび11.5mg)は不安定であり、同定することができ
なかった。第3の成分(39mg)のみが、化合物Bとして同定された。
複合体4(61mg)を、異なる移動相CH2Cl2:MeOH(97.5:2.5 v/v)を用いる
以外は上記の条件と同様である、改変された半分取HPLC法によりさらに精製した
。この複合体から、3つの純粋な成分(化合物G(6.1mg)、化合物H(8.7mg)
、および化合物J(27.5mg))を得た。
物理化学的特性
全ての化合物は、溶媒の除去後に白色粉末として得られた。この化合物は、メ
タノール、ジメチルスルホキシド、酢酸エチル、アセトン、およびクロロホルム
に可溶であり;ジエチルエーテル、ジクロロメタン、および1-クロロブタンに一

本発明の化合物の構造決定
化合物の構造は、分光学的データ(紫外(UV)、赤外(IR)、高速原子衝撃質量分

実施例1
化合物Aの構造解明を、質量分析データおよびNMRスペクトルデータの分析に
より達成した。FABマススペクトルデータは、参照試料(13-384-成分-1、エバニ
ノマイシン)と比較して34 amuの分子量の増加を示した。トリクロロ含有分子イ
オンクラスターを、FABマススペクトル中に観測した。両方の観測により、化合
物A中に第3の塩素原子の存在が明らかになった。分子の右側上の芳香族エステ
ルフラグメントへのこれらの余分の塩素原子の結合を、参照試料(13-384-成分-

本発明の化合物の生物学的特性
