Claims (1)
- 【特許請求の範囲】 1.アスペルギルス・ニガー・バール・アワモリ(Aspergillus niger var .a wamori )から単離されることを特徴とする転位性配列。 2.Seq.ID No.3で示されるDNA配列を含むことを特徴とする請求の範囲第 1項記載の転位性配列。 3.44bpの逆方向反復配列を含むことを特徴とする請求の範囲第1項記載の転 位性配列の断片。 4.Seq.ID No.4またはSeq.ID No.5で示されるDNA配列を含むことを特徴と する請求の範囲第3項記載の断片。 5.糸状菌から転位性配列を単離する方法であって、請求の範囲第4項記載の 断片のうちの1個をプローブとして使用することを特徴とする方法。 6.請求の範囲第5項記載の方法に従って単離されることを特徴とする転位性 配列。 7.Seq.ID No.6で示されるDNA配列を含むことを特徴とする請求の範囲第 1項記載の転位性配列。 8.請求の範囲第7項記載の転位性配列によってコードされていることを特徴 とする推定トランスポゼース。 9.Seq.ID No.7で示されるアミノ酸配列を含むことを特徴とする請求の範囲 第8項記載のトランスポゼース。 10.遺伝子を不活化する方法であって、該遺伝子を含む宿主細胞内に、不活化 すべき遺伝子内のTA配列を標的とする転位性配列を形質転換し、該転位性配列 が該遺伝子に挿入されて不活化され、および該遺伝子による遺伝子発現を阻止す るようにすることを含むことを特徴とする方法。 11.遺伝子を活性化する方法であって、該遺伝子を含む宿主細胞内に、宿主細 胞のゲノム内のTA配列において転位性配列の標的となる所望のDNA配列が挿 入された転位性配列を形質転換し、該転位性配列が該遺伝子に挿入されて、活性 化された遺伝子産物の発現を制御するようにすることを特徴とする方法。
