Claims (1)
- 【特許請求の範囲】 1.GnRH−Rのヌクレオチド配列の一部に相補的なアンチセンス配列をコー ドし、かつ細胞におけるGnRH−R遺伝子の翻訳を阻害するオリゴヌクレオチ ド。 2.GnRH−Rのアミノ末端領域をコードするヌクレオチド配列に相補的な、 請求項1に記載のオリゴヌクレオチド。 3.GnRH−Rのエピトープに免疫特異的に結合するモノクローナル抗体。 4.GnRHのGnRH−Rへの結合を競合的に阻害する、請求項3に記載のモ ノクローナル抗体。 5.ヒトGnRH−Rに結合する、請求項3または4に記載のモノクローナル抗 体。 6.GnRH−Rを用いて宿主種を免疫感作することを含んでなる避妊法。 7.GnRH−R融合タンパク質を用いて宿主種を免疫感作することを含んでな る避妊法。 8.GnRH−Rシグナル伝達を変調することのできる化合物を同定する方法で あって、 (a)純粋または半純粋形態の、膜調製物中の、または全生存細胞もしくは 固定細胞中のGnRH−Rまたはその機能性誘導体に前記化合物を接触させ; (b)工程(a)の混合物を、GnRHまたはGnRH類似体の存在下で、 前記化合物がシグナル伝達を促進または抑制するのに十分な期間インキュベート し; (c)シグナル伝達を測定し; (d)上記シグナル伝達活性を前記化合物の不在下でインキュベートしたG nRH−Rのシグナル伝達活性と比較し、それによって前記化合物がシグナル伝 達を刺激するか又は抑制するかを決定する、 ことを含んでなる該方法。 9.化学的または生物学的調製物中の、GnRH−Rに結合可能な分子を同定す る方法であって、 (a)GnRH−Rまたはその機能性断片を固相マトリックスに固定し; (b)前記化合物を結合させるのに十分な期間、上記化学的または生物学的 調 製物を工程(a)で作成した固相マトリックスに接触させ; (c)固相マトリックスから未結合の物質をすべて洗い落とし; (d)固相に結合した前記化合物の存在を検出する、 ことを含んでなる該方法。 10.化学的または生物学的調製物中の、GnRH−Rに結合可能な分子を同定す る方法であって、 (a)GnRH−Rまたはその機能性断片を固相マトリックスに固定し; (b)前記化合物を結合させるのに十分な期間、上記化学的または生物学的 調製物を工程(a)で作成した固相マトリックスに接触させ; (c)固相マトリックスから未結合の物質をすべて洗い流し; (d)固相に結合した前記化合物の存在を検出し; (e)固相マトリックスから結合した化合物を溶出させ、それにより該化合 物を単離する、 ことを含んでなる該方法。 11.GnRH−Rに結合可能なペプチドを同定する方法であって、 (a)固相支持体に結合させた、アミノ酸の全ての可能な組合せからなるラ ンダムペプチドライブラリーを、前記ペプチドを結合させるのに十分な期間Gn RH−Rタンパク質またはその機能性断片に接触させ; (b)未結合ペプチドをすべて洗い流し; (c)GnRH−Rに結合した前記ペプチドの存在を検出する、 ことを含んでなる該方法。 12.請求項8、9、10または11に記載の方法によって同定または単離された化合 物を含んでなる、GnRH−Rの生物活性を抑制する医薬組成物。 13.請求項8、9、10または11に記載の方法によって同定または単離された化合 物を含んでなる、GnRH−Rの生物活性を活性化する医薬組成物。 14.請求項8、9、10または11に記載の方法によって同定または単離された化合 物を含んでなる、動物の腫瘍を治療するための医薬組成物。 15.請求項8、9、10または11に記載の方法によって同定または単離された化合 物を含んでなる、避妊のための医薬組成物。
