Claims (1)
- 【特許請求の範囲】 1.所定の標的RNA配列に結合するリガンドを識別するための高処理量方法で あって、 (a)前記標的RNA配列に結合することが不明の複数の化合物をテストリガ ンドとして選択し; (b)前記標的RNA配列をそれぞれの前記テストリガンドの存在下で保温し てテスト組み合わせを作製し; (c)前記標的RNA配列をテストリガンドの非存在下で保温してコントロー ル組み合わせを作製し; (d)それぞれの組み合わせにおいて前記標的RNA配列のコンホメーション を測定し; (e)リガンドとして、コントロール組み合わせにおける前記標的RNAコン ホメーションに比べてテスト組み合わせにおいて前記標的RNAコンホメーショ ンにおける測定可能な変化を起こすいずれかのテストリガンドを選択し; (f)複数の前記テストリガンドについて工程(b)−(e)を繰り返し、前 記標的RNA配列に結合するリガンドを識別する 工程を含む方法。 2.請求項1記載の方法であって、前記標的RNAコンホメーションにおける前 記測定可能な変化が、前記標的RNAコンホメーションにおける、少ない重畳か ら多い重畳への、多い重畳から少ない重畳への、又は第1の重畳コンホメーショ ンから第2のオルタナティブ重畳コンホメーションへの変化を含む方法。 3.請求項1記載の方法であって、前記テスト及びコントロール組み合わせを、 前記コントロール組み合わせにおける前記標的RNA配列の検出可能なフラクシ ョンを変性させる条件に供する工程をさらに含む方法。 4.請求項3記載の方法であって、前記供する工程が、温度の変更、塩濃度の変 更、変性化合物の添加及びこれらの組み合わせのうち少なくとも1つを含む前記 方法。 5.請求項1記載の方法であって、前記標的RNAが長さ約5から約500ヌク レオチドである方法。 6.請求項1記載の方法であって、前記標的RNAが、放射核種、蛍光化合物、 親和性標識及びこれらの組み合わせからなる群より選択される標識を含む方法。 7.請求項1記載の方法であって、前記測定工程が: (i)前記テスト及びコントロール組み合わせを、オリゴヌクレオチドが標的 RNA配列の所定のコンホメーションと優先的にハイブリダイズする条件下でオ リゴヌクレオチドと接触させる工程;及び (ii)前記オリゴヌクレオチドとのハイブリッド中に存在する前記標的RN A配列のフラクションを測定する工程を含む方法であって、(ii)において測 定される前記フラクションが前記所定のコンホメーションにある標的RNAのフ ラクションを示す方法。 8.請求項7記載の方法であって、前記オリゴヌクレオチドが、放射核種、蛍光 化合物、親和性標識及びこれらの組み合わせからなる群より選択される標識を含 む方法。 9.請求項1記載の方法であって、前記標的RNA配列がさらに第1の蛍光プロ ーブ及び第2の蛍光プローブを含み、 (i)前記第1のプローブの前記蛍光放出波長の最大が前記第2のプローブの 蛍光吸収波長の最大と重なり、 (ii)前記第1のプローブ及び第2のプローブが標的RNA内に位置し、そ れにより第1及び第2のプローブ間の蛍光エネルギー転移が、標的RNAが所定 のコンホメーションにある場合のみ起こる 方法。
